A new strategy for the preparation of activated supports containing oxidized lipoic acid (LA) is presented. An amino-agarose gel with low pKa (monoaminoethyl-N-aminoethyl-agarose) was highly substituted with a disulfide ligand (254 μmoles LA/g dried gel) which was subsequently oxidized to disulfide oxides. Antioxidant systems are fundamental components of host–parasite interactions, and often play a key role in parasite survival. Here, we report the cloning, heterologous expression, and characterization of a thioredoxin glutathione reductase (TGR) from Fasciola hepatica. The deduced polypeptide sequence of the cloned open reading frame (ORF) confirmed the experimental N-terminus previously determined for a native F. hepatica TGR showing thioredoxin reductase (TR) activity. The sequence revealed the presence of a fusion between a glutaredoxin (Grx) and a TR domain, similar to that previously reported in Schistosoma mansoni and Echinococcus granulosus. The F. hepatica TGR sequence included an additional redox active center (ACUG; U being selenocysteine) located at the C-terminus. The addition of a recombinant selenocysteine insertion sequence (SECIS) element in the Escherichia coli expression vector, or the substitution of the native selenocysteine by a cysteine, indicated the relevance of this unusual amino acid residue for the activity of F. hepatica TGR. Rabbit vaccination with recombinant F. hepatica TGR reduced the worm burden by 96.7% following experimental infection, further supporting the relevance of TGR as a promising target for anti Fasciola treatments. A new imidazolium hydrated salt (DCF-IMI-H2O) of the nonsteroidal anti-inflammatory drug Diclofenac (DCF) was synthesized by solvent evaporation. This salt was fully characterized by X-ray diffraction (SCXRD, PXRD), infrared spectroscopic (FT-IR) and thermal analysis (TGA, DSC, HSM) techniques. The salt formation is driven by deprotonation of the drug's carboxylic group, with proton transfer to the nitrogen atom of the imidazole ring. The water molecule plays an important role in the 3D arrange observed in the crystal lattice, acting as a bridge between adjacent DCF and coformer molecules. The DCF-IMI-H2O exhibits an improved solubility profile when compared with the pure form of DCF, and similar values when compared with sodium and potassium salts. This research assesses the bench-scale application of a non-conventional support, bone particles, for glucoamylase (GA) immobilization and its subsequent use in cassava starch hydrolysis. Upon determining the appropriate conditions to immobilize GA onto chicken bone particles, such as pH, ionic strength, particle size, and enzyme load, bench-scale immobilization of commercial GA without further purification was performed. Under the selected conditions, 270 GA units per gram of support were adsorbed. Optimal temperature and thermal stability of immobilized GA were only slightly different from those of the free enzyme, while optimal pH became more acidic by about one unit. The feasibility of the use of this immobilized biocatalyst for high glucose syrup production from liquefied cassava starch, at bench scale in batch process using a stirred-tank reactor, was demonstrated. It has been shown that TcCyP19 is inhibited by the immunosuppressive drug cyclosporin A (CsA) and analogs, which also proved to have potent trypanosomicidal activity in vitro. In this work, we continue and expand a previous study on the molecular interactions of CsA, and a set of analogs modeled in complexes with TcCyP19. The modeled complexes were used to evaluate binding free energies by molecular dynamics (MD), applying the Linear Interaction Energy (LIE) method. In addition, putative binding sites were identified by molecular docking. In our analysis, the binding free energy calculations did not correlate with experimental data. The heterogeneity of the non-bonded energies and the variation in the pattern of hydrogen bonds suggest that the systems may not be suitable for the application of the LIE method. Further, the docking calculations identified two other putative binding sites with comparable scoring energies to the active site, a fact that may also explain the lack of correlation found. Kinetic experiments are needed to confirm or reject the multiple binding sites hypothesis. In the meantime, MD simulations at the alternative sites, employing other methods to compute binding free energies, might be successful at finding good correlations with the experimental data. With the objective of analysing, at the molecular level, how the structural differences between the three above-mentioned inhibitors justify their different inhibitory activity on TcCyP19, three-dimensional molecular modelling structures were generated to computationally simulate behaviours and interactions. An energy-minimized model of each binary complex in water with ions was obtained. These models were then used as starting point for molecular dynamic simulations, performed with GROMOS96 program. With the resulting set of co-ordinates and energies, a comparison of the interaction between CsA and both CsA analogues in T. cruzi and human cyclophilins were performed. Within the different magnitudes analysed, the total potential complex energy exhibited the best correlation with the experimental data. Sin embargo, siempre ha habido sobrevivientes incluso de las infecciones más letales, gracias a la eficiencia de la respuesta del sistema inmune innato. De todas maneras, la tasa de mortalidad en la era pre-antibióticos era muchísimo más alta que hoy en día. Sin embargo, la continua generación de resistencia bacteriana hace que hoy en día no se pueda dar por sentado el tratamiento de cualquier infección mediante el uso de agentes antimicrobianos. La emergente generación de resistencia a los principios activos disponibles se enfrenta mediante políticas de racionalización del uso de antibióticos y mediante la continua búsqueda de nuevos compuestos con actividad antimicrobiana. En el presente trabajo, se describen los esfuerzos realizados para la síntesis de análogos simplificados del antibiótico higromicina A (HA). El mismo es un compuesto aislado a partir de Streptomyces hygroscopicus, y fue muy estudiado en la década del 90 debido a la excelente actividad que presenta in vivo frente al microorganismo responsable de la disentería porcina (B. hyodysentereae). Su estructura modular compuesta por tres subunidades lo hace un compuesto ideal para el estudio sistemático de la relación estructura-actividad. Es así que un grupo de la empresa Pfizer ha sintetizado diversos análogos, y uno de ellos en particular (CP-111906), que es un derivado de la molécula original, presentó la misma potencia que el antibiótico natural, pero tiene una complejidad estructural disminuida. En la presente tesis doctoral nos propusimos sintetizar nuevos análogos de HA, aplicando reacciones contempladas dentro del concepto “click chemistry”. Particularmente, se propuso sintetizar una serie de aminociclitoles y azidoinositoles a partir del bromociclohexadiendodiol enantioméricamente puro, obtenido a su vez a partir de una biotransformación de bromobenceno. Los aminociclitoles o azidoinositoles fueron posteriormente acoplados a los residuos aromáticos correspondientes, mediante un enlace amida o un anillo triazólico. Con los análogos sintetizados se pretende evaluar los efectos del reemplazo bioisostérico amida-triazol, el efecto de sustituir el metilenodioxo por un isopropilidenodioxo, y el efecto de cambiar la configuración absoluta del carbono que soporta el grupo funcional nitrogenado. Vitis vinifera flowers and grape fruits are one of the most interesting ecosystem niches for native yeasts development. There are more than a 100 yeast species and millions of strains that participate and contribute to design the microbial terroir. The wine terroir concept is understood when grape and wine micro-regions were delimited by different quality characteristics after humans had been growing vines for more than 10,000 years. Environmental conditions, such as climate, soil composition, water management, winds and air quality, altitude, fauna and flora and microbes, are considered part of the “terroir” and contribute to a unique wine style. If “low input winemaking” strategies are applied, the terroir effect will be expected to be more authentic in terms of quality differentiation. Interestingly, the role of the microbial flora associated with vines was very little study until recently when new genetic technologies for massive species identification were developed. These biotechnologies allowed following their environmental changes and their effect in shaping the microbial profiles of different wine regions. In this chapter we explain the interesting positive effects on flavor diversity and wine quality obtained by using “friendly” native yeasts that allowed the microbial terroir flora to participate and contribute during fermentation. During the last years, the progression to control malaria disease seems to be slowed and WHO (World Health Organization) reported a modeling analysis with the prediction of the increase in malaria morbidity and mortality in sub-Saharan Africa during the COVID-19 pandemic. A rapid way to the discovery of new drugs could be carried out by performing investigations to identify drugs based on repurposing of “old” drugs. The 5-nitrothiazole drug, Nitazoxanide was shown to be active against intestinal protozoa, human helminths, anaerobic bacteria, viruses, etc. In this work, Nitazoxanide and analogs were prepared using two methodologies and evaluated against P. falciparum 3D7. A bithiazole analog, showed attractive inhibitory activity with an EC50 value of 5.9 μM, low propensity to show toxic effect against HepG2 cells at 25 μM, and no cross-resistance with standard antimalarials.
