En el presente trabajo se realizaron estudios para confirmar la existencia de lipoxigenasas activas en los hígados de merluza, enzimas que pueden ser las promotoras del deterioro del aceite contenido en los hígados. Cuando una solución conteniendo una mezcla de ácidos grasos obtenidos a partir del aceite de girasol se incubó con el agregado de un extracto acuoso obtenido a partir de los hígados de merluza, se verificó un gradual y continuo incremento en la concentración de hidroperóxidos en la mezcla. La velocidad de oxidación de los ácidos grasos en las incubaciones resultó directamente proporcional a la cantidad de extracto agregado, pudiendo ser eficientemente disminuida por el agregado de antioxidantes. La actividad lipoxigenante encontrada en los hígados de merluza es del orden de 100.000 veces menor a la que se determina por el mismo método para los porotos de soja. El rango óptimo de actividad se obtiene en un rango de pH de 8.0 a 8.5. La determinación se hace en el residuo sulfúrico, donde previo ajuste del medio (acidez regulada a 3N y equilibrio redox) se extrae por solución clorofórmica de ditizona. Se estudiaron las condiciones para evitar las pérdidas por volatilización y también el proceso irregular de pérdidas por calentamiento de soluciones sulfúricas diluidas. La técnica ajustada permite recuperar el mercurio en una proporción del 95%. Verificada por adición de 0,8 a 10 ug de mercurio a orinas normales. Operando con 67 ml. de orina haciendo la determinación comparativa del mercurio con tipos y blancos se puede alcanzar un límite de positivo neto para contenidos de 12ug de mercurio por litro de orina. Se realizó la determinación de sodio, calcio y cobre en 45 muestras de vinos Uruguayos, con el fin de evaluar si cumplen con los requisitos de la normativa de la región y, establecer si existe relación entre los niveles de estos elementos y el tipo de vino. Las determinaciones se realizaron mediante espectrometría de absorción atómica de llama. Se validó la metodología para determinar los 3 elementos mediante dilución directa. Se obtuvo las cifras de mérito linealidad, precisión, límites de detección, exactitud y efecto de interferencias. Los resultados indicaron que las cifras de mérito eran adecuadas y sin observarse efecto matriz. Todos los vinos analizados cumplieron con la reglamentación vigente para estos elementos según la normativa del MERCOSUR. Title A name given to the resource Determinación de tóxicos metálicos y no metálicos en cabello y su aplicación en la evaluación de la exposición a metales y semimetales tóxicos, en niños del Asentamiento Aquiles Lanza, Montevideo Subject The topic of the resource TOXICOLOGIA ANALÍTICA ECOSALUD Abstract A summary of the resource. El presente proyecto tiene como objetivo optimizar las metodologías analíticas para evaluar la presencia de metales y semimetales tóxicos en la matriz cabello a los efectos de ser utilizado como matriz biológica para la determinación de elementos tóxicos, evaluando su desempeño como posibles biomarcadores de exposición en población presuntamente expuesta. Para ello, en primera instancia, se puso a punto una metodología para el monitoreo y tratamiento de tóxicos metálicos y semimetálicos en una matriz compleja como lo es el cabello. Se optimizaron las metodologías para el tratamiento de la muestra y para las determinaciones de los elementos en estudio se utilizó la técnica de espectroscopía de absorción atómica. Se logró desarrollar un protocolo de digestión para determinación de cuatro elementos, plomo, mercurio, arsénico y manganeso, en forma simultánea y su posterior validación. Luego se realizó un estudio en población presuntamente expuesta a los tóxicos estudiados en el asentamiento Aquiles Lanza, con un enfoque basado en los pilares de investigación de la Ecosalud: transdisciplinaridad, participación social, pensamiento sistémico, equidad social y de género, sustentabilidad y acción. En el estudio poblacional, se encontraron niveles de plomo cuantificables, mientras que del resto de los analitos, si bien se detectaron, los niveles estuvieron por debajo de los límites de cuantificación. Del análisis estadístico, surge que podrían existir correlaciones con el sexo de los niños entre otras variables estudiadas. El estudio realizado, ha contemplado una integración global, utilizando los principios de la Ecosalud.
