Alzheimer’s disease (AD) is the most common form of dementia, affecting approximately 47 M people worldwide. Histological features and genetic risk factors, among other evidence, supported the amyloid hypothesis of the disease. This neuronocentric paradigm is currently undergoing a shift, considering evidence of the role of other cell types, such as microglia and astrocytes, in disease progression. Previously, we described a particular astrocyte subtype obtained from the 3xTg-AD murine model that displays neurotoxic properties in vitro. We continue here our exploratory analysis through the lens of metabolomics to identify potentially altered metabolites and biological pathways. Cell extracts from neurotoxic and control astrocytes were compared using high-resolution mass spectrometrybased metabolomics. Around 12 % of metabolic features demonstrated significant differences between neurotoxic and control astrocytes, including alterations in the key metabolite glutamate. Consistent with our previous transcriptomic study, the present results illustrate many homeostatic and regulatory functions of metabolites, suggesting that neurotoxic 3xTg-AD astrocytes exhibit alterations in the Krebs cycle as well as the prostaglandin pathway.This is the first metabolomic study performed in 3xTg-AD neurotoxic astrocytes. These results provide insight into metabolic alterations potentially associated with neurotoxicity and pathology progression in the 3xTg-AD mouse model and strengthen the therapeutic potential of astrocytes in AD. Biological significance: Our study is the first high-resolution metabolomic characterization of the novel neurotoxic 3xTg-AD astrocytes. We propose key metabolites and pathway alterations, as well as possible associations with gene expression alterations in the model. Our results are in line with recent hypotheses beyond the amyloid cascade, considering the involvement of several stress response cascades during the development of Alzheimer’s disease. This work could inspire other researchers to initiate similar studies in related models. Furthermore, this work illustrates a powerful workflow for metabolite annotation and selection that can be implemented in other studies. Bardet-Biedl syndrome (BBS) is a ciliopathy characterized by retinal degeneration, obesity, polydactyly, renal disease and mental retardation. CCDC28B is a BBS-associated protein that we have previously shown plays a role in cilia length regulation whereby its depletion results in shortened cilia both in cells and Danio rerio (zebrafish). At least part of that role is achieved by its interaction with the mTORC2 component SIN1, but the mechanistic details of this interaction and/or additional functions that CCDC28B might play in the context of cilia remain poorly understood. Here we uncover a novel interaction between CCDC28B and the kinesin 1 molecular motor that is relevant to cilia. CCDC28B interacts with kinesin light chain 1 (KLC1) and the heavy chain KIF5B. Notably, depletion of these kinesin 1 components results in abnormally elongated cilia. Furthermore, through genetic interaction studies we demonstrate that kinesin 1 regulates ciliogenesis through CCDC28B. We show that kinesin 1 regulates the subcellular distribution of CCDC28B, unexpectedly, inhibiting its nuclear accumulation, and a ccdc28b mutant missing a nuclear localization motif fails to rescue the phenotype in zebrafish morphant embryos. Therefore, we uncover a previously unknown role of kinesin 1 in cilia length regulation that relies on the BBS related protein CCDC28B. The interaction of dendritic cells and macrophages with a variety of rigid non-cellular  particles triggers activation of the NLRP3 inflammasome and consequent secretion of  IL-1β. Non-cellular particles can also be generated in the context of helminth infection,  as these large pathogens often shed their outermost structures during growth and/or  moulting. One such structure is the massive, mucin-based, soft and flexible laminated  layer (LL), which protects the larval stages of cestodes of the genus Echinococcus. We  show that particles from the E. granulosus LL (pLL) trigger NLRP3- and caspase-1-  dependent IL-1β in LPS-primed mouse bone marrow-derived dendritic cells (BMDC).  This response can be elicited by pLL particles too large for phagocytosis, and  nonetheless requires actin dynamics, Syk and PI3K. These three requirements had  already been observed in our previous study on the alteration by pLL of BMDC  responses to LPS in terms of CD86, CD40, IL-10 and IL-12: however, we now show  that these alterations are independent of NLRP3 and caspase-1. In other words, an initial interaction with particles requiring actin dynamics, Syk and PI3K but not phagocytosis  elicits both NLRP3-dependent and NLRP3-independent responses. Intraperitoneal  injection of pLL induced IL-1, suggesting that contact with LL materials induces IL- 36 1 in the E. granulosus infection setting. Human colorectal adenocarcinoma cell line Caco-2 was used to study the cell growth inhibition capacity of these compounds, their capacity of altering the cell cycle and possible induction of apoptosis, DNA fragmentation, and genotoxic damage. The IC50 after 24 h of incubation was lower for 1, 2, and 3 (4.8, 46.8, and 8.2 μM, respectively) than for 4 (474.7 μM). Compound 1 induced arrest in the G2/M phase at 24 and 48 h of treatment and apoptosis at the highest doses at 24 h of treatment. These facts were corroborated with caspase 3, caspase 9, and cytochrome c activation and DNA fragmentation at 24 h of treatment. The derivatives studied induced neither significant single strand breaks nor oxidative damage at the different studied times.We concluded that among the series of N5,N10-dioxide phenazine derivatives analyzed, 1, which contains a nitro moiety and a chloroacetamide group, is the most promising as an antitumoral compound. Although nowadays landspreading and incineration are the conventional disposal methods for the primary sludge (PS) and biosludge (BS) generated in the wastewater treatment plants of the pulp mill industry in Uruguay, their direct incineration for energy recovery is the most preferable one. Here, we propose the hydrothermal carbonization (HTC) method as a pre treatment to improve the fuel characteristic of PS and BS. Proximate and ultimate analysis, scanning electron microscopy, and thermogravimetric analysis show that the HTC afects PS and BS diferently regarding carbon fxation, energy densifca tion and energy conversion efciency. Moreover, ash composition measured by X-ray fuorescence indicates that the HTC efectively diminishes the slagging and fouling indexes for both feedstocks, which can increase the life expectancy of boil ers. Alternative PS, BS, and their hydrochars were activated with H3PO4 to produce activated carbons, i.g. the BET specifc surface area was as high as 800 m2 /g for PS-AC, and exhibits CO2 adsorption capacity of 5 mmol/g at 10 bar at 273 K. These studies provide alternatives to direct incineration and landspreading of PS and BS in Uruguay to reduce their environmental impact, either by enhancing their fuel quality or by converting them into much more valuable products to face air pollution. The essential oil profile of Baccharis anomala DC was examined. Samples were collected from in the Pró-Mata area (São Francisco de Paula, Rio Grande do Sul State, Brazil), and GC–MS data and retention indices were used to identify 58 oil components. β-Selinene (40.8%), caryophyllene oxide (9.9%) and spathulenol (6.8%) were the major constituents of the steam-distilled oil. The olfactive profile of the B. anomala oil was described as: sweet, resinous and woody. When the oil was evaluated by gas chromatography–olfactometry (GC–O), the study revealed that the oil has a complex aroma profile formed by 34 odorants with modified frequency above 40, 31 of which could be identified. In this way, the odor, was objectively established for this species. The volatile components isolated from the root of Angelica (Angelica archangelica L.) cultivated in southern Brazil have been studied. The essential oil was obtained by hydrodistillation; the volatile concentrate was obtained by supercritical fluid extraction (SFE). SFE was conducted at 40°C, 50°C and 60°C, at pressures ranging from 90–120 bar. The identification of the components was completed using capillary gas chromatography and mass spectrometry based on the retention time on methyl silicone phases. Chemical analysis revealed that although roughly the same compounds were extracted, the two oils had a widely different percentage composition. Exposure to environmental lead (Pb) is a mild risk factor for amyotrophic lateral sclerosis (ALS), a paralytic disease characterized by progressive degeneration of motor neurons. However, recent evidence has paradoxically linked higher Pb levels in ALS patients with longer survival. We investigated the effects of lowlevel Pb exposure on survival of mice expressing the ALS-linked superoxide dismutase-1 G93A mutation (SOD1G93A). SOD1G93A mice exposed to Pb showed longer survival and increased expression of VEGF in the ventral horn associated with reduced astrocytosis. Pretreatment of cultured SOD1G93A astrocytes with low, non toxic Pb concentrations upregulated VEGF expression and significantly abrogated motor neuron loss in coculture, an effect prevented by neutralizing antibodies to VEGF. The actions of Pb on astrocytes might explain its paradoxical slowing of disease progression in SOD1G93A mice and the improved survival of ALS patients. Understanding how Pb stimulates astrocytic VEGF production and reduces neuroinflammation may yield a new therapeutic approach for treating ALS. This study investigated the microbial community developed in a UASB reactor for hydrogen production and correlated it to reactor performance. The reactor was inoculated with kitchen waste compost and fed with raw cheese whey at two organic loading rates, 20 gCOD/Ld and 30 gCOD/Ld. Hydrogen production was very variable, using an OLR of 30 gCOD/Ld averaged 1.0 LH2/Ld with no methane produced under these conditions. The hydrogen yield was also very variable and far from the theoretical. This low yield could be explained by selection of a mixed fermentative population with presence of hydrogen producing organisms (Clostridium, Ruminococcus and Enterobacter) and other non-hydrogen producing fermenters (Lactobacillus, Dialister and Prevotella). The molecular analysis of the raw cheese whey used for feeding revealed the presence of three predominant organisms that are affiliated with the genera Buttiauxella (a low-yield hydrogen producer) and Streptococcus (a lactic acid-producing fermenter). Although these organisms did not persist in the reactor, the continuous addition of these fermenters could decrease the reactor’s hydrogen yield. In this work, an upflow anaerobic packed bed reactor configuration to produce hydrogen using cheese whey as the substrate was tested. The microbiological composition was linked to the reactor operation. Three different organic loading rates of 22, 33, and 37 g COD/L-d were used at a fixed hydraulic retention time of 24 h. The increase of the organic loading rate from the initial value of 22 g COD/L-d to a value of 37 g COD/L-d and the adjustment of the pH to values greater than 5 had a positive effect on the hydrogen production and values of up to 1 L H2/L-d were achieved. The production of hydrogen was stable under all of the investigated conditions, and the problems frequently reported in this kind of reactors (bed clogging, methanogenesis and solvent production) were not observed during operation. The highest hydrogen yield was 1.1 mol H2/mol lactose far from the maximum theoretical value (8 mol H2/mol lactose), but similar to previous works using cheese whey as substrate. The microbiological analysis showed a mixed population with a low proportion of hydrogen-producing fermenters (Clostridium and Klebsiella) and other non-hydrogen-producing organisms. A series of new 3-alkoxy- or 3-hydroxy-1-[x-(dialkylamino)alkyl]-5-nitroindazoles have been synthesized and their trichomonacidal, antichagasic and antineoplastic properties studied. Five derivatives (5, 6, 8, 9 and 17) showed remarkable trichomonacidal activity against Trichomonas vaginalis at 10 lg/mL concentration. Three compounds (8, 10, 11) exhibited interesting antichagasic activity and these same compounds moderate antineoplastic activity against TK-10 and HT-29 cell lines. Unspecific cytotoxicity against macrophages has also been evaluated and only compounds 9, 10 and 11 resulted cytotoxic at the higher dose evaluated (100 lg/mL), loosing cytotoxicity at lower doses. Russian sturgeon (Acipenser gueldenstaedtii) has been successfully farmed in Uruguay for the past ten years. However, during the Uruguayan summer fish endure high water temperatures and increased bacterial infections that threaten aquaculture. Our understanding of sturgeon's immune system and its interplay with environmental factors like temperature is almost unknown. This study analysed the way in which seasonal variations affect enzymatic blood components of Russian sturgeon's innate defences, including the serum alternative complement pathway (ACP), ceruloplasmin (Cp) and lysozyme activities. Results showed that summertime conditions in the farm altered these defences in different ways, inducing a significant decrease in ACP and Cp, and an increase in lysozyme. In addition, serum levels of total protein and cortisol decreased in summer, suggesting a chronic stress response was induced in parallel. Subsequently, we analysed whether the increase in water river temperature during summer could account for the observed results. To that end, we acclimated juvenile sturgeons to mild (18 °C) or warm (24 °C) temperatures for 37 days. Like in summer, sturgeons exposed to 24 °C showed lower levels of serum ACP, Cp and total proteins, together with a progressive decrease in body weight and increased fish mortality. Administration of an immunostimulant containing Se and Zn slightly reverted the temperature-induced effects on sturgeon's defences. Altogether, our study provides novel data on various physiological parameters of the Russian sturgeon and highlights the impact warm temperature has on stress and innate immunity in this chondrostean fish. The immune system of sturgeons, one of the most ancient and economically valuable fish worldwide, is poorly understood. The lack of molecular tools and data about infection biomarkers hinders the possibility to monitor sturgeon health during farming and detect infection outbreaks. To tackle this issue, we mined publicly available transcriptomic datasets and identified putative positive acute-phase proteins (APPs) of Russian sturgeons that could be induced by a bacterial infection and monitored using non-invasive methods. Teleost literature compelled us to focus on five promising candidates: hepcidin, a warm acclimation associated hemopexin, intelectin, serum amyloid A protein (SAA) and serotransferrin. Among them, SAA was the most upregulated protein at the mRNA level in the liver of sturgeons challenged with heat-inactivated or live Aeromonas hydrophila. To assess whether this upregulation yielded increasing SAA levels in circulation, we developed an in-house ELISA to quantify SAA levels in sturgeon serum. Circulating SAA rose upon bacterial challenge and positively correlated with hepatic saa expression. This is the first time serum SAA has been quantified in an Actinopterygii fish. Hesperozygis myrtoides is a native species from Cerrado and Atlantic Forest of Brazil. The essential oil from aerial parts of this plant, collected at different periods of the year, was obtained by hydrodistillation and analyzed by GC and CG-MS. The most abundant compounds were limonene (2.1–22.7%), isomenthone (14.3–47.7%), neo-isomenthol (1.8–4.4%), pulegone (19.8–57.3%) and isomenthyl acetate (0.3–14.3%). The chromatographic data were processed using hierarchical cluster analysis (HCA) and principal component analysis (PCA) based on the major compounds. The multivariate analyses (MVA) indicated that the variation of chemical composition of principal substances of H. myrtoides essential oil depends on the altitude. The enantiomeric distribution of (R)-(+)-pulegone and (+)-isomenthone were established for the first time for this essential oil by bidimensional GC-chiral analysis. R02,the first member of a new lipase family (Family XVII) Subject The topic of the resource LIPASAS JANIBACTER BIBLIOGRAFIA NACIONAL QUIMICA 2017 Abstract A summary of the resource. Janibacter sp. strain R02 (BNM 560) was isolated in our laboratory from an Antarctic soil sample. A remarkable trait of the strain was its high lipolytic activity, detected in Rhodamine-olive oil supplemented plates. Supernatants of Janibacter sp. R02 displayed superb activity on transesterification of acyl glycerols, thus being a good candidate for lipase prospection. Considering the lack of information concerning lipases of the genus Janibacter, we focused on the identification, cloning, expression and characterization of the extracellular lipases of this strain. By means of sequence alignment and clustering of consensus nucleotide sequences, a DNA fragment of 1272 bp was amplified, cloned and expressed in E. coli. The resulting recombinant enzyme, named LipJ2, showed preference for short to medium chain-length substrates, and displayed maximum activity at 80 °C and pH 8–9, being strongly activated by a mixture of Na+ and K+. The enzyme presented an outstanding stability regarding both pH and temperature. Bioinformatics analysis of the amino acid sequence of LipJ2 revealed the presence of a consensus catalytic triad and a canonical pentapeptide. However, two additional rare motifs were found in LipJ2: an SXXL β-lactamase motif and two putative Y-type oxyanion holes (YAP). Although some of the previous features could allow assigning LipJ2 to the bacterial lipase families VIII or X, the phylogenetic analysis showed that LipJ2 clusters apart from other members of known lipase families, indicating that the newly isolated Janibacter esterase LipJ2 would be the first characterized member of a new family of bacterial lipases.
