A summary of the resource.
<span>An analytical method for sample preparation and subsequent determination of Cd, Pb and Se in beef using aerosol dilution by inductively coupled plasma-mass spectrometry was developed. The use of high matrix introduction technology allowed the introduction of a complex sample into the plasma with higher content of total dissolved solids a residual acidity of 4% and better tolerance to matrix effects. Trueness was verified by the analysis of two reference materials. At the 95% significance level, results were statistically equivalent to the certified or informed values. Analytical precision expressed as relative standard deviation was less than 9% in all cases. After validation, a total of 12 beef samples from Uruguay were analyzed. Obtained levels for Cd and Pb were far below the maximum limits established by international regulations in all samples. Also, results showed that beef from this region is a good source of Se in the range 0.120–0.870 mg kg</span><span>−1</span><span> with an interesting contribution to the daily intake of this essential element.</span>

A new analytical method has been developed and successfully evaluated in routine application for the quantitative analysis of a selected group of organophosphate pesticides (coumaphos, chlorpyrifos and ethion) which can be found at trace levels in propolis tinctures (ethanolic propolis extracts); a valuable commodity used as raw material in the food and pharmaceutical industries for which there have been few attempts for pesticide residue analysis reported in the literature. The proposed methodology is based on matrix solid phase dispersion (MSPD) using aluminum sulfate anh. a novel dispersant material and subsequent column chromatography clean-up in silica gel prior to gas chromatography (GC) with both flame photometric detector (FPD) and mass spectrometry (MS) detection used for the routine quantification and identification of the residues, respectively. The limits of detection, for coumaphos, chlorpyrifos and ethion were below 26.0 �g/kg in FPD and 1.43 �g/kg for MS detection. Mean recoveries were in the range of 85–123% with RSD values below 13%, which suggests that the proposed method is fit for the purpose of analyzing pesticides in propolis tinctures containing high concentration of polyphenolics. The method has been successfully applied in our laboratory for the last 2 year in the analysis of real propolis tinctures samples.

Insolubles determination is one of the parameters usually recommended to evaluate the residual life of oil because their presence at elevated levels in diesel lubricating oil changes the viscosity, prematurely clogs filters and is one of the major factors in causing abrasive engine wear. The proposed method employs visible spectrophotometric detection in association with flow injection analysis. The results obtained by this method were compared with the ones obtained by Fourier transform infrared spectrometry (FT-IR) since this is the most employed method for insolubles determination. The proposed method presented a linear response from 0 to 3% (w/w) of insolubles in pentane (ASTM D-893). The sampling frequency was about 30 samples h−1, with a relative standard deviation (n=5) of 2.4% or better. Accuracy was evaluated analysing 98 real samples and the results obtained with the FIA-spectrophotometric method were plotted against those obtained by the FT-IR method by means of linear regression. Slope and intercept of the straight line obtained were compared with the theoretical values of 1 and 0 by means of the joint-confidence ellipse F-test. At the confidence level of 95% no evidence of a difference was found between both methods.

The use of zero-order and first-derivative flame emission spectrometry has been investigated for the determination of basal concentrations of lithium in serum and urine at the µg dm–3 level. No significant matrix effect was observed; however, it was necessary to make use of background correction techniques. Two or three wavelength measurements and first-derivative spectrometry were used for this purpose; both methods gave similar results. Detection limits for serum of approximately 0.09 µg dm–3 were found by both zero-order and first-derivative emission measurements, while normal lithium levels were found to be 29.3 and 1.17 µg dm–3 for urine and serum, respectively.

Abstract
A summary of the resource.
An analytical method for sample preparation of igneous rocks and subsequent determination of Si, Na, K, Ca, Mg, Al, Fe, Mn, Ba, Sr, and Ti by means of microwave induced emission spectrometry (MP-AES) was developed and validated. The proposed sample preparation procedure avoids the use of perchloric acid and provided accurate results even for silicon determination using an acid digestion with hydrofluoric acid. The determination of major elements in rocks is required for the design of classification diagrams that provides relevant information for geochemical analysis. • MP-AES showed to be an adequate technique to measure major and some trace elements that are relevant for classification of igneous rocks. • This method is in better agreement with the principles of the Green Analytical Chemistry and constitutes a reliable alternative to classical analytical and tedious procedures used for geochemical analysis. • The methodology was successfully applied to chemical classification of rocks from Valle Chico (Uruguay) using a Total Alkali-Silica Diagram (TAS).

With the aim of monitoring multiclass semi-polar pesticide residues in freshwater fatty fish, two QuEChERS approaches (so-called acetate buffered and unbuffered versions) were evaluated for the determination of 77 pesticide residues. Compounds were selected according to the dominant rainfed agriculture activities in South America. Unbuffered QuEChERS was finally chosen for validation purposes owing that it provided the best results in terms of recovery yields. Method performance was evaluated in two instrumental systems, liquid chromatography – tandem mass spectrometry (LC–MS/MS) in Scheduled MRM™ algorithm available on hybrid quadrupole – linear ion trap (QLIT) instrument, and gas chromatography – mass spectrometry (GC–MS) under selected ion monitoring (SIM) mode. Spiking experiments were carried out to determine the trueness, precision, linearity, limit of quantification of the method as well as matrix effect. The Unbuffered QuECHERS method described here: • Was validated for the analysis of 67 pesticide residues in fish muscle tissue. • Presented quantification limits in the range 1–15 μg kg−1 for the vast majority of the studied compounds. • Enable environmental monitoring of pesticide residues in fish due to their low LOQs.

A summary of the resource.
Aiming to select the most suitable sample preparation for the multiresidue analysis of pesticides in globe artichoke (Cynara cardunculus L.), a modified QuEChERS, a matrix solid phase dispersion and a dispersive ethyl acetate extraction were compared. Trueness and precision were determined at 0.2 mg kg−1 for the three methods. The modified QuEChERS showed the best performance. The scope of the method was enlarged to 35 GC and 63 LC amenable pesticides, its overall performance was evaluated and validated to artichoke leaves and fruits according to DG-SANTE Guidelines. Different matrix effects were observed for most of the pesticides which were higher for leaves than fruits. Difenoconazole and flutriafol suffered signal suppression in leaves extracts but showed positive matrix effects in fruits. All pesticides were analyzed at or below their Maximum Residue Levels fixed for globe artichoke by the European Union. The method was successfully applied for the analysis of commercial samples.

A summary of the resource.
The determination of the ratio of phospholipid/ lipophilic compounds in liposomes was achieved after thinlayer chromatography (TLC) by measuring the spot intensities of dipalmitoyl phosphatidylcholine and the lipophilic compound. The liposome components under study were separated on one TLC plate, developed in two steps, and detected after charring the plate with specific visualization reagents. The method shows good reproducibility and provides a simple way to quantify the level of lipophilic compound incorporated in the liposome bilayer.

A summary of the resource.
A multicommutated flow system was designed and evaluated for the determination of selenium by hydride generation atomic absorption spectrometry (HG-AAS). It was applied to the determination of total selenium in samples of cow’s milk (fluid and powder) and infant formulae. Linearity was satisfactory in the range up to 27.5 mg L�1 (h ¼ 0.082 C + 0.0033, h ¼ peak-height, absorbance, C ¼ concentration in mg L�1, r2 ¼ 0.999). Detection (3s) and quantification (10s) limits in solution were LD ¼ 0.08 mg L�1 and LQ ¼ 0.27 mg L�1, corresponding to LD ¼ 3.2 mg kg�1 and LQ ¼ 10.8 mg kg�1 in solid samples, and to LD ¼ 0.8 mg L�1, LQ ¼ 2.7 mg L�1 in fluid milk samples. Trueness was verified by analysis (n ¼ 5) of two reference materials (NIST 1549, Non-fat Milk Powder and NIST 1846 Infant Formula). At the 95% significance level, results were statistically equivalent to the certified values. Instrumental precision (sr(%), n ¼ 5) was in the range 1.4% to 11.7%, analytical precision (sr(%), n ¼ 5) being 4.2 and 9.3% respectively for the determination of the above mentioned reference materials. The sampling frequency of the system was 160 hour�

<p>Two flow methods, based, respectively, on flow-injection analysis (FIA) and on multicommutated flow analysis (MCFA), were compared with regard to their use for the determination of total selenium in infant formulas by hydride-generation atomic absorption spectrometry. The method based on multicommutation provided lower detection and quantification limits (0.08 and 0.27 μg L−1 compared to 0.59 and 1.95 μ L−1, resp.), higher sampling frequency (160 versus. 70 samples per hour), and reduced reagent consumption. Linearity, precision, and accuracy were similar for the two methods compared. It was concluded that, while both methods proved to be appropriate for the purpose, the MCFA-based method exhibited a better performance.</p>

A new method was developed for analysing 4-ethylguaiacol and 4-ethylphenol in the aroma of red wines using dispersive liquid–liquid microextraction (DLLME) coupled with gas chromatography–mass spectrometry detection (GC–MS). Parameters such as extraction solvent, sample volume and disperser solvent were studied and optimised to obtain the best extraction results with the minimum interference from other substances, thus giving clean chromatograms. The response linearity was studied in the usual concentration ranges of analytes in wines (50–1500 �g/L). Repeatability and reproducibility of this method were lower than 5% for both volatile phenols. Limits of detection and limits of quantification were also determined, and the values found were 28 and 95 �g/L for 4-ethylguaiacol and 44 and 147 �g/L for 4-ethylphenol, respectively. This new method has been used for the determination of the volatile phenols concentration in different samples of Tannat wine affected by Brettanomyces contamination.

A summary of the resource.
The determination of several trace elements, including thorium and the lanthanides in coal and coal ashes applying total dissolution and ICP-MS, was studied. The procedures were tested with reference materials (Bituminous Coal NIST 1632a and Fly Ash NIST 1633a). For coal samples, chemical ashing with HNO3, HF, and HClO4 produces reasonable results. Regarding coal ash samples, some difficulties related to the determination of rubidium, cesium, thorium, and the lanthanides were found and a proposed solution is discussed. Finally, coal, bottom, electrostatic precipitator (ESP), and fly ash samples from a Brazilian coalfired power plant were analyzed and an enrichment of Mn, Zn, Ge, As, Se, Mo, Cd, Sb, Ag, Pb, Bi, and U in fly ash was observed. The values obtained for refractory elements, such as thorium, cerium, and scandium, in the ESP samples were compared with those obtained by INAA, with good agreement between both results. The use of a large number of elements (57) during the instrument calibration allows the use of the TotalQuant mode as a routine method, instead of the traditional quantitative method, aiming at a trace element environmental monitoring program

A summary of the resource.
A flow-injection system is proposed for the determination ofmetal-based additives in lubricating oils. The system, operating under computer control uses a motorised syringe for measuring and injecting the oil sample (200 μL) in a kerosene stream, where it is dispersed by means of a packed mixing reactor and carried to an atomic absorption spectrometer which is used as detector. Zinc was used as model analyte. Two different systems were evaluated, one for low concentrations (range 0–10 ppm) and the second capable of providing higher dilution rates for high concentrations (range 0.02%–0.2%w/w). The sampling frequency was about 30 samples/h. Calibration curves fitted a second-degree regression model (r2 = 0.996). Commercial samples with high and low zinc levels were analysed by the proposed method and the results were compared with those obtained with the standard ASTM method. The t test for mean values showed no significant differences at the 95% confidence level. Precision (RSD%) was better than 5% (2% typical) for the high concentrations system. The carryover between successive injections was found to be negligible.

The relative strengths of aromatic and aliphatic C―H⋅⋅⋅X hydrogen bonds in imidazolium ionic liquids were investigated through measurement of H/D isotope effects on the 19F nuclear shielding of deuterated isotopologues of 1‐n‐butyl‐3‐methylimidazolium hexafluorophosphate and tetrafluoroborate ([C4mim]PF6 and [C4mim]BF4). Δ19F(H,D) values ranging from 9.7 to 49.7 ppb were observed for [C4mim]PF6 isotopologues, while for the [C4mim]BF4 series these went from 26.2 to 83.8 ppb. Our findings indicate that the interactions between the fluorinated anions and protons on the C‐1′ and C‐1″ position of the N‐alkyl sidechains are comparable to, and in some cases stronger than, those involving protons on the aromatic ring, underscoring the role that these weak interionic forces have on the local ordering of imidazolium salts in the liquid state.

Several disciplines like Environmental Toxicology, Epidemiology, Public Health and Geology have been the basis of the development of Medical Geology in Uruguay during the last decade. The knowledge and performance in environmental and health issues have been improved by joining similar aims research teams and experts from different institutions to face environmental problems dealing with the population’s exposure to metals and metalloids and their health impacts. Some of the Uruguayan Medical Geology examples are reviewed focusing on their multidisciplinary approach: Lead pollution and exposed children, selenium in critically ill patients, copper deficiency in cattle and arsenic risk assessment in ground water. Future actions are also presented.

Assessment of the presence of estrogen receptors in breast cancer is crucial for treatment planning. With the objective to develop a potential agent for estrogen receptors imaging, we present the development and characterization of a 99mTc‐tricarbonyl–labelled estradiol derivative. Using ethinylestradiol as starting material, an estradiol derivative bearing a 1,4‐disubstituted 1,2,3‐triazole–containing tridentate ligand system was synthesized by “Click Chemistry” and fully characterized. Labelling with high yield and radiochemical purity was achieved through the formation of a 99mTc‐tricarbonyl complex. The radiolabelled compound was stable, exhibited moderate binding to plasma protein (approximately 33%) and lipophilicity in the adequate range (logP 1.3 ± 0.1 at pH 7.4). Studies in MCF7 showed promising uptake values (approximately 2%). However, more than 50% of the activity is quickly released from the cell. Biodistribution experiments in normal rats confirmed the expected “in vivo” stability of the radiotracer but showed very high gastrointestinal and liver activity, which is inconvenient for in vivo applications. Taking into consideration the well‐documented influence of the chelating system in the physicochemical and biological behaviour of technetium‐labelled small biomolecules, research will be continued using the same pharmacophore but different complexation modalities of technetium.

The aim of this work was to develop and evaluate a 99mTc‐labeled neuropeptide Y derivative with affinity toward Y1‐receptor. The selected amino acid sequence included nine amino acids derived from the C‐terminal portion of the NPY complemented with the addition of one cysteine‐mercaptoacetic acid moiety to bind the radiometal. Labeling was achieved through the preparation of a 3 + 1 nitrido complex. Physicochemical evaluation, cell uptake, internalization and externalization studies, and competitive assays were performed. Biodistribution experiments were carried out in normal and tumor‐bearing mice. A single product with radiochemical purity >90% and high stability was obtained. In vitro analysis showed specific cellular uptake, IC50 of 73.2 nM, and a high internalization rate (80%). Biodistribution studies showed low blood and renal uptake and combined hepatobiliary and urinary elimination. Preliminary studies in mice bearing induced breast tumors rendered promising uptake values.

Reduction of disulfide bonds and introduction of “de novo” thiol groups in cyclodextrin glucantransferase from Thermoanaerobacter sp. were assessed in order to perform reversible covalent immobilization onto thiol-reactive supports (thiolsulfinate-agarose). Only the thiolation process dramatically improved the immobilization yield, from 0 % for the native and reduced enzyme, up to nearly 90 % for the thiolated enzyme. The mild conditions of the immobilization process (pH 6.8–7.0 and 22 °C) allowed the achievement of 100 % coupling efficiencies when low loads were applied. Ionic strength was a critical parameter for the immobilization process; for high activity recoveries, 50 mM phosphate buffer supplemented with 0.15 M NaCl was required. The kinetic parameters, pH and thermal stabilities for the immobilized biocatalyst were similar to those for the native enzyme. For β-cyclization activity, optimal pH range and temperature were 4.0–5.4 and 85 °C. The possibility of reusing the support was demonstrated by the reversibility of enzyme–support binding.

In the agricultural field, chitosan (CS) is of great interest due to their broad antimicrobial activity and their ability to induce resistance mechanisms in plants against phytopathogens. Likewise, the potential of silver nanoparticles in preventing microbial infections has been demonstrated, including their broad antimicrobial activity against phytopathogens. In this study, the synthesis and characterization of chitosan-silver nanohybrids were performed. Firstly, mycosynthesis of biogenic silver nanoparticles (PchNPs) was carried out. Subsequently, the nanohybrid (CS-PchNHs) synthesis involved the addition of PchNPs, with continuous stirring, into a CS solution. For the first-time stable CS-Pch NHs were produced using biogenic silver nanoparticles from the fungus Phanerochaete chrysosporium, characterized by UV-visible spectroscopy (UV-Vis), Dynamic Light Scattering, (DLS), ζ-Potential, and X- ray diffraction (XRD). Additionally, CS-PchNHs exhibited high antifungal activity at low concentrations against four phytopathogens, suggesting that these nanosystems may be used as an alternative to the current hazardous antifungal agents in the integrated disease management.
