Coordination complexes may act as anticancer agents. Among others, the formation of the complex may facilitate the ligand uptake by the cell. Searching for new copper compounds with cytotoxic activity, the complex Cu-dipicolinate was studied as a neutral scaffold to form ternary complexes with diimines. A series of [Cu(dipicolinate)(diimine)] complexes (where diimine: Phenan throline, phen, 5-NO2 -phenanthroline, 4-methyl-phenanthroline, neocuproine, 3,4,7,8-tetramethyl phenanthroline, tmp, bathophenanthroline, bipyridine, dimethyl-bipyridine, as well as the ligand 2,2-dipyridil-amine, bam) were synthesized and characterized both in the solid state, including a new crystal structure of [Cu2 (dipicolinate)2 (tmp)2 ]·7H2O. Their chemistry in aqueous solution was explored by UV/vis spectroscopy, conductivity, cyclic voltammetry, and electron paramagnetic resonance studies. Their DNA binding was analyzed by electronic spectroscopy (determining Kb values), circular dichroism, and viscosity methods. The cytotoxicity of the complexes was assessed on human cancer cell lines MDA-MB-231, MCF-7 (breast, the first triple negative), A549 (lung epithelial) and A2780cis (ovarian, Cisplatin-resistant), and non-tumor cell lines MRC-5 (lung) and MCF-10A (breast). The major species are ternary, in solution and solid state. Complexes are highly cytotoxic as compared to Cisplatin. Complexes containing bam and phen are interesting candidates to study their in vivo activity in triple-negative breast cancer treatment. In order to seek for new compounds with antitumor activity, our research group is focused on copper complexes with mixed ligands. This work presents the synthesis and characterization of eight compounds with general formula [Cu(iminodiacetate)(diimine)(H2O)x], with x = 0 or 1, and their cytotoxic activities against tumor cells. The chosen diimine ligands were: 2,2́-dipyridil-amine (bam), 2,2́-bipyridine (bipy), 4,4́-dimethyl-2,2́-bipyridine (dmb), 1,10-phenanthroline (phen), 4-methyl-1,10-phenanthroline (4met-phen), 5-nitro-1,10-phenanthroline (5nitro-phen), neocuproine (neo) and bathophenanthroline (batho). Compounds were characterized in solid state using FT-IR, TGA, DSC and single crystal X-ray diffraction analyses by which five new crystal structures were determined. Common features of the coordination scheme include fac-NO + O isomerism of the iminodiacetate (ida) ligand, except for the complex with neo where the ida presents mer-NO2 isomerism. The diimine ligand completes the equatorial plane for the facial isomers, with different degrees of distortion in the octahedral or square-based pyramid geometry depending on the diimine ligand, whereas the neo is perpendicular to the equatorial plane in the latter. Studies in solution, using EPR, UV–Vis spectroscopy and conductimetry, showed that the copper coordination observed in solid state is retained except for the batho complex. DNA interaction studies evidenced that the complexes bind to this biomolecule, inducing different conformational changes. Biological experiments evidenced that all complexes induce cell death in the cell line MDA-MD-231 (metastasic breast cancer line). [Cu(ida)(neo)] and [Cu(ida)(batho)] presented the highest activity. Taken all these data together we propose that [Cu(ida)(neo)] and [Cu(ida)(batho)] are good candidates for further studies in vitro and in vivo. In thiscontext, the aim of the present work was to apply a combination of sensory and consumer methodologies tothe development of chocolate milk desserts enriched with resistant starch. Chocolate milk desserts containingmodified waxy maize starch were formulated with six different concentrations of two types of resistantstarch (which are part of insoluble dietary fiber). The desserts were evaluated by trained assessors usingQuantitative Descriptive Analysis. Moreover, consumers scored their overall liking and willingness to purchaseand answered an open-ended question. Resistant starch caused significant changes in the sensorycharacteristics of the desserts and a significant decrease in consumers’ overall liking and willingness topurchase. Consumer data was analyzed applying survival analysis on overall liking scores, considering therisk on consumers liking and willing to purchase the functional products less than their regular counterparts.The proposed methodologies proved to be useful to develop functional foods taking into account consumers’ perception, which could increase their success in the market. A new strategy for the preparation of activated supports containing oxidized lipoic acid (LA) is presented. An amino-agarose gel with low pKa (monoaminoethyl-N-aminoethyl-agarose) was highly substituted with a disulfide ligand (254 μmoles LA/g dried gel) which was subsequently oxidized to disulfide oxides. Oxidation carried out with H2O2 gave a support with a large binding capacity for low molecular weight thiols, as demonstrated with glutathione (GSH), achieving 105 μmoles bound GSH per g dried gel. GSH binding was completely reversible under reducing condition. Protein immobilization was assayed with K. lactis b- galactosidase. However, although this protein bound readily to the LA-modified support, there was no release of the protein after reduction. Two simple and straightforward sample preparation methods were developed for the multiresidue analysis of post-emergence herbicides in loam soil that are commonly used in rice crop cultivation. A number of strategic soil extraction and cleanup methods were evaluated. The instrumental analysis was performed by HPLC with a diode array detector. The best compromise between the recoveries (69–98%) and good repeatability (RSD <15%) was obtained using either methanol or methanol–ethyl acetate (70 + 30, v/v) as extraction solvents without any further cleanup. For the first time, residues of metsulfuron methyl, bensulfuron ethyl, bentazone, cyhalofop butyl, pyrazosulfuron ethyl, propanil, and clomazone were analyzed simultaneously. Quinclorac and bispyribac sodium were also assayed, but their recoveries were below 50%. Both methods had an LOD of 0.7 mg/kg and could accurately determine the residues at the 2 mg/kg level. These two methods could not be applied directly to other soil types as the recoveries strongly depended on the soil composition. The developed methodologies were successfully applied in monitoring 87 real-world soil samples, in which only propanil (6 to 12 mg/kg) and clomazone (15 to 20 mg/kg) residues could be detected. The development of 99mTc complexes for imaging 5-HT1A neural system receptors using the 3 + 1 mixed ligand approach is described. Six novel complexes (I-VI) were designed using two different strategies. In complexes I-IV the pharmacophore 1-(2-methoxyphenyl)piperazine was attached to a monodentate thiol used as co-ligand and combined with tridentate dianionic aminothiols (SNS and NNS). On the other hand, complexes V and VI were obtained using thiophenol and 4-methoxy-thiophenol as co-ligand and a tridentate ligand (SNS) with the pharmacophore bound to the nitrogen through an alkyl chain. All complexes were prepared at tracer level using 99mTc-glucoheptonate as precursor. Ligand and co-ligand concentration, reaction time and temperature were optimized to achieve high substitution yield and radiochemical purity. Structure was studied at carrier level through the corresponding rhenium complexes. Complexes I and II presented the expected ReOLK structure and a distorted trigonal bipyramidal geometry. The structure of the other four complexes has not been completely elucidated yet. Biodistribution studies of all the complexes demonstrated selective brain uptake and retention. Uptake of complex I in receptor-rich hippocampus was significantly higher than that of the cerebellum (P = 0.05) 1 h post-injection. Oxorhenium complexes I and II showed affinity for the 5-HT1A receptor binding sites, with IC50 values in the nanomo with IC50 values in the nanomolar range. The results demonstrate the potential of the mixed ligand approach for the design of 99mTc complexes with the ability to bind neuroreceptors. However, the goal of imaging 5-HT1A receptors with technetium requires further development of complexes with improved biological profiles. In the search for new anthelmintics able to overcome the resistance problem against all available drugs in livestock, the synthesis of novel valerolactam-benzimidazole hybrid compounds was reported. This allowed us to obtain these in vitro and in vivo bioactive compounds using Nippostrongylus brasiliensis rat model by integrating physiology-based assays and ex vivo diffusion studies. In order to further study those novel hybrid molecules, Haemonchus contortus (a sheep gastrointestinal nematode of interest) and Mesocestoides vogae tetrathyridia (a useful system to study the efficacy of anthelmintic drugs against cestoda) were used as parasite models to compare the ex vivo patterns of diffusion and biotransformation of benzimidazoles and their valerolactam-benzimidazole hybrid derivatives. On average, a nine-fold higher intraparasitic concentration of compounds was found in M. vogae compared with H.contortus, with similarities regarding the order of entry of compounds, highlighting febendazole (FEB) and its hybrid compound 10, while valerolactam compound 2 practically did not penetrate the parasites. Interestingly, sulphoxidation drug metabolism was observed and measured, revealing percentages of oxidation of 8.2% and 14.5% for albendazole (ABZ) and febendazole respectively in M. vogae, while this effect was more relevant in H. contortus parasite. More importantly, significant differences were observed between anthelmintic-susceptible adult parasites (Hc S) and those from sheep farms (Hc U). In fact, the percentages of oxidation of FEB and the hybrid compound 8 were higher in Hc U (25.5%, 54.1%, respectively) than in Hc S (8.8%, 38.2%). Interestingly, sulphoxidation of hybrid compound 10 was neither observed in M. vogae nor in H. contortus parasites, suggesting that increased drug metabolism (oxidation reactions) could not be used by these parasites as a defense mechanism against this novel drug. The preparation of oxyfluoroborate glasses and glass–ceramics doped with Yb3+ and Er3+ with improved luminescent properties is reported. Glasses with compositions (100 − x − y) (MO·2B2O3) + xPbF2 + y(YbF3:ErF3) (% in mol), where M is Sr, Ba, Ca, x = 10–40% and y = 0, 1 or 5% were prepared by the melt/quenching technique. Glass crystallization was studied using thermal analysis and X-ray diffraction techniques. Optical absorption and infrared up-conversion were studied on both glasses and glass–ceramics. The incorporation of a fluoride compound into the borate glass resulted to depend on the ionic radius of the glass modifier: as it increases, glasses become more stable against crystallization. On the other hand, alkali–fluoride compounds such as BaF2 and SrF2 can be crystallized from these systems by selecting an appropriate proportion between their components. Furthermore, the up-conversion response can be tuned by changing the glass modifiers (Sr, Ba and Ca) of the borate matrix, which also influence the type of fluoride crystallized compound. The strontium glass–ceramics have the highest luminescence response due to the crystallization of SrF2 compound in the system SrO–B2O3–PbF2. Meanwhile, the lowest luminescence signal was obtained for samples in the system BaO–B2O3–PbF2 where the Pb1.33Ba2.66B11FO20 phase crystallizes. The thiosemicarbazone moiety as a pharmacophore has been described for inhibition of the essential cysteine protease, cruzipain, of this parasite. Our recent study identified an amidinohydrazone containing benzofuroxan as a hit compound for cruzipain inhibition with trypanosomicidal activity. Structural modification of the amidinohydrazone, thio- and semicarbazone motifs, using benzofuroxan and including a benzimidazole 1,3-dioxide system as new core scaffolds are described. These changes allowed for the identification of new structural motifs with desired antitrypanosomal activity. The new amidinohydrazone, thio-, and semicarbazone derivatives had excellent anti-trypanosomal activity without improved cruzipain-inhibitory activity compared with the parent compounds. Adequate methods for skin diagnosis are necessary to determine the characteristics state of the skin of cosmetic users in order to properly formulate cosmetics products, and could provide a useful tool for supporting cosmetics efficacy claims. In the present work sensory methods of clinical evaluation and selfevaluation were developed for the diagnosis of the state of the facial skin of fifty nine volunteers. A Cosmetologist observed the facial skin of the volunteers and scored six attributes using a structured 9-point quality scale. Besides, volunteers were asked to evaluate the state of their facial skin by scoring the same attributes than the Cosmetologist, using the same quality scale. Both clinical evaluation and selfevaluation methods provided a simple an efficient tool to characterize the state of the volunteers’ skin. Clinical evaluation provided information regarding the objective state of the skin, whereas selfevaluation provided a measure of the perception that each volunteer has of her skin. Cluster analysis allowed the identification of groups of volunteers with similar state of their skin, according to clinical evaluation and selfevaluation. Considering clinical evaluation data, two groups of volunteers were identified, which differed in the type and frequency of the skin care treatment they used. Volunteers that frequently used antiaging and moisturizing products showed higher clinical evaluation scores, showing the positive effect of cosmetic treatments on the state of the skin. Regarding selfevaluation, three clusters were identified, that mainly differed in their age. Older volunteers tended to be stricter with themselves and gave lower scores to the state of their skin than younger volunteers, probably due to different expectations. Although clinical evaluation and selfevaluation data were not correlated, they provided complementary information. Triamcinolone acetonide (TA) is a powerful anti-inflammatory drug used in the treatment of inflammatory ocular disorders; however, its poor aqueous solubility and ocular anatomical barriers hinder optimal treatment. The aim of this work was to obtain triamcinolone acetonide nanocrystals (TA-NC) to improve ocular corticosteroid therapy. Self-dispersible TA-NC were prepared by the bead milling technique followed by spray-drying, exhaustively characterized and then evaluated in vivo in an ocular model of endotoxin-induced uveitis (EIU). Self-dispersible TA-NC presented an average particle size of 257 ± 30 nm, a narrow size distribution and a zeta potential of −25 ± 3 mV, which remained unchanged for 120 days under storage conditions at 25 ◦C. In addition, SEM studies of the TA-NC showed uniform and spherical morphology, and FTIR and XRDP analyses indicated no apparent chemical and crystallinity changes. The subconjunctival administration of TA-NC in albino male white rabbits showed no clinical signs of ocular damage. In vivo studies proved that treatment with self-dispersible TA-NC alleviated the inflammatory response in the anterior chamber and iris in EUI rabbit eyes. Dispersible TA-NC are a promising approach to obtaining a novel nanometric TA formulation for ocular disorders. The aim of the present study was to develop a mixed tropical fruit juice of cashew apple, acerola and melon with low added-sugar content considering sensory and nutritional aspects. Five formulations were developed varying the concentration of the different fruits. A total of 172 consumers rated their overall liking using a 9-point hedonic scale and described their sensory characteristics using a Check-all-that-apply question. In addition, the functional properties of the juices were evaluated by analyzing vitamin C, total phenolic compounds and antioxidant capacity. Results showed that the addition of sugar and fruit composition affected the sensory characteristics and consumer liking of the formulations. Samples without added sugar and with higher melon concentration showed lower liking scores (p<0.05). In addition, the highest concentration of acerola had a positive effect on nutritional characteristics. The formulation with 50% of fruit pulp (60% of cashew apple, 30% of acerola and 10% of melon), 47% of water, and 3% of added sugar achieved the best results. The effect of aroma on sweetness perception was investigated by adding identical to natural aromas of melon, apple and pineapple to the selected formulation. However, aroma did not significantly modify sweetness perception, evaluated using a rate-all-that-apply question. Methanol adsorption on PtCo(111) surface at low coverage is studied using Density Functional Theory (DFT) calculations without and with van der Waals corrections. We investigated the PtCo FCT alloy surface with a uniform distribution. The most favorable site for CH3OH adsorption is on top of a Co atom, with an adsorption energy of −0.92 eV. Methanol attachs to the surface by the O atom, with a distance of 2.24 Å. The molecule presents a small distortion after adsorption. The COCo bond angle is 142°. The CH bonds are strengthened whereas the CO and OH bonds are weakened. A charge transfer from C atom to O atom occurs upon adsorption, and then further transfer occurs to the Co atom on the surface. The calculated vibrational frequencies for adsorbed methanol present a red-shift displacement compared to gas-phase, confirming the adsorption process. Los estudios indican que los pacientes cuyos tumores presentan una amplificación o sobreexpresión de HER2 presentan una menor expectativa de vida que la de aquellos con tumores sin amplificaci6n 0 sobreexpresi6n de HER2. Trastuzumab (HERCEPTIN®) es un anticuerpo monoclonal humanizado que reconoce de forma selectiva el dominio extracelular de HER2 y actualmente es utilizado como agente quimioterapeutico al inhibir la proliferaci6n de las celulas tumorales humanas que sobreexpresan HER2. Basandose en la alta especificidad de Trastuzumab por HER2, el objetivo del presente trabajo es desarrollar el radiofarmaco 99mTccHYNIC- Trastuzumab como marcador molecular in vivo de la expresión de HER2 en cancer de mama. En el presente estudio se desarrollo y optimizo la marcación de Trastuzumab con 99mTc, para ello se derivatizó el anticuerpo con el ligando bifuncional HYNIC. Se desarrollaron metodos de control basados en sistemas cromatograficos tanto en ITLC como HPLC, tambien se desarrollo un sistema para estudiar la inmunoafinidad in vitro del anticuerpo marcado. A synthetic peptide (GU4) derived from an antigen B (AgB) subunit was serologically compared with crude antigen (HCFA); immunopurified AgB and antigen 5 (Ag5), and two other synthetic peptides, for diagnosis of human cystic hydatidosis. GU4 was derived from the sequence of AgB/2, the novel AgB subunit described by us. The other two peptides: 65 (AgB mimotope) and 89-122 (Ag5 mimotope), were described by others. Antigens B and 5 showed higher diagnostic sensitivity than corresponding peptides. All sera reacting with peptides 89-122 and GU4 also reacted with 65. The latter provided three to four times higher sensitivity than the former two peptides, but 30% lower specificity. The diagnostic efficiency of AgB (82%) was higher than those of Ag5 (74%) and HCFA (71%). Interestingly, 89-122 only reacted with hydatid sera, some of which did not react with AgB. Considering positive those reacting with 89-122 or AgB, sensitivity increases from 77% (with AgB) to 82% (combined), while specificity is the same as with AgB (86%). Our results suggest that hydatid serology may be improved by: a) combining several defined antigens (including synthetic peptides), b) design of new E. granulosus-specific mimotopes, which react with the false negative sera (16/90; 18%). La enfermedad de Huntington (EH), es un trastorno neurodegenerativo progresivo del sistema nervioso central, que afecta específicamente a algunas estructuras cerebrales. Es una patología genética de herencia autosómica dominante de penetrancia completa. Causada por una expansión de tripletes CAG, en el exón 1 del gen HTT que codifica la proteína Huntingtina. Es de expresividad variable con una edad de inicio en la década de los 40 años, y existe una fuerte correlación inversa entre la cantidad de repetidos CAG y la edad de inicio de los síntomas. Factores genéticos y factores ambientales influyen en esta variabilidad. En este trabajo se puso a punto el diagnóstico de Huntington, inexistente anteriormente en nuestro país, y se relacionó la variación en la edad de comienzo de los síntomas tanto con la cantidad de repetidos CAG, como con polimorfismos presentes en genes modificadores. Los polimorfismos estudiados fueron: la expansión CCG y Δ2642 en gen HTT, ApoE, UCHL1 (rs5030732), ADORA2A (rs5751876), PPARGC 1a (rs7665116), HAP 1 (rs4523977). Por tratarse de una enfermedad muy poco frecuente, las correlaciones que involucren estudios estadísticos no han sido posibles. Sólo se pueden 10 hacer deducciones caso a caso que permiten una más profunda comprensión de esta patología. El estudio en conjunto de los polimorfismos en los genes modificadores y las expansiones CAG, permitió entender mejor las bases genéticas de esta patología, aportándonos mayor información acerca de la variabilidad en la edad de aparición de los síntomas.
