A summary of the resource.
<p>Aims: To differentiate nine industrial wine strains of Saccharomyces cerevisiae using microsatellite (simple sequence repeats, SSR) markers.</p>
<p>Methods and Results: Six of the strains were indigenous yeasts currently used as high-density starter monocultures by the Uruguayan wine industry. Unequivocal differentiation of these six native strains and three commercial S. cerevisiae wine strains was achieved by PCR amplification and polymorphism analysis of loci containing microsatellite markers.</p>
<p>Conclusions: We recommend the use of this reproducible and simple molecular method to routinely discriminate wine yeast strains. Significance and Impact of the Study:  Microsatellites are superior to other methods for typing yeasts because the results can be exchanged as quantitative data. Knowledge of the frequencies of the alleles for different SSR markers will eventually lead to an accurate typing method to identify industrial wine yeast strains.</p>

A summary of the resource.
Diffusion Monte Carlo (DMC) and Reptation Monte Carlo (RMC) methods, have been applied to study some properties of the NaK molecule. Hartree-Fock (HF), Density Functional (DFT) and single and double configuration interaction (SDCI) wavefunctions with a valence quadruple zeta atomic natural orbital (VQZ/ANO) basis set were used as trial wavefunctions. Values for the potential energy curve, dissociation energy and dipole moment were calculated for all methods and compared with experimental results and previous theoretical derivations. Quantum Monte Carlo (QMC) calculations were shown to be useful methods to recover correlation in NaK, essential to obtain a reasonable description of the molecule. The equilibrium distance—interpolated from the potential energy curves—yield a value of 3.5 Å, in agreement with the experimental value. The dissociation energy, however, is not as good. In this case, a conventional CCSD(T) calculation with an extended aug-pc-4 basis set gives a much better agreement to experiment. On the contrary, the CCSD(T), other MO and DFT methods are not able to reproduce correctly the large dipole moment of this molecule. Even DMC methods with a simple HF trial wavefunction are able to give a better agreement to experiment. RMC methods are even better, and the value obtained with a B3LYP trial wavefunction is very close to the experimental one.

Trypsins from the pyloric caeca of menhaden (Brevoortia spp.) and croaker (Micropogonias furnieri) were used for hydrolysis of both myofibrillar and sarcoplasmic fractions from menhaden muscle. Digestion of muscle proteins was carried out at 37C for 20 min with the pH maintained at either 3, 4, 5, 6, 7, 8 or 9; or for 60 min at the pH's of 5, 7, 8 or 9 at 30C and 37C. The hydrolytic action was evaluated based on the concentration of peptides solubilized. Solubility after digestion of myofibrillar, sarcoplasmic fraction and hemoglobin was optimum at basic pH. The sarcoplasmic fraction was solubilized more than the myofibrillar fraction by both fish trypsins. Similar results were obtained with both crude pyloric extracts and purified trypsins from menhaden and white croaker. Thus, the preparation of the crude pyloric caeca may be preferentially used, because of its low cost as well as the simplicity of the preparative procedure.

With the aim to develop compounds able to target multiple metabolic pathways and, thus, to lower the chances of drug resistance, we investigated the anti-trypanosomal activity and selectivity of a series of symmetric diglycosyl diselenides and disulfides. Of 18 compounds tested the fully acetylated forms of di-β-D-glucopyranosyl and di-β-D-galactopyranosyl diselenides (13 and 15, respectively) displayed strong growth inhibition against the bloodstream stage of African trypanosomes (EC50 0.54 μM for 13 and 1.49 μM for 15) although with rather low selectivity (SI < 10 assayed with murine macrophages). Nonacetylated versions of the same sugar diselenides proved to be, however, much less efficient or completely inactive to suppress trypanosome growth. Significantly, the galactosyl (15), and to a minor extent the glucosyl (13), derivative inhibited glucose catabolism but not its uptake. Both compounds induced redox unbalance in the pathogen. In vitro NMR analysis indicated that diglycosyl diselenides react with glutathione, under physiological conditions, via formation of selenenylsulfide bonds. Our results suggest that non-specific cellular targets as well as actors of the glucose and the redox metabolism of the parasite may be affected. These molecules are therefore promising leads for the development of novel multitarget antitrypanosomal agents.

In this work, several immobilization strategies for Gluconobacter oxydans NBRC 14819 (Gox) were tested in the bioconversion of crude glycerol to dihydroxyacetone (DHA). Agar, agarose and polyacrylamide were evaluated as immobilization matrixes. Glutaraldehyde crosslinked versions of the agar and agarose preparations were also<br />tested. Agar immobilized Gox proved to be the best heterogeneous biocatalyst in the bioconversion of crude glycerol reaching a quantitative production of 50 g/L glycerol into DHA solely in water. Immobilization allowed<br />reutilization for at least eight cycles, reaching four times more DHA than the amount obtained by a single batch of free cells which cannot be reutilized. An increase in scale of 34 times had no impact on DHA productivity. The results obtained herein constitute a contribution to the microbiological production of DHA as they not only attain<br />unprecedented productivities for the reaction with immobilized biocatalysts but also proved that it is feasible to do it in a clean background of solely water that alleviates the cost of downstream processing.

Research into perceived wellbeing (WB) can improve understanding of consumer food choices and support development of strategies to promote healthier and more sustainable eating habits. In this research, using online studies, food-related WB was explored among New Zealand consumers. In a “quasi-replication” of Jaeger, Vidal, Chheang, and Ares (2022), Study 1 uncovered word associations to different WB-related terms (‘Sense of wellbeing,’ ‘Lack of wellbeing,’ ‘Feeling good,’ ‘Feeling bad/unhappy,’ ‘Satisfied with life,’ and ‘Dissatisfied with life’) with 912 participants in a between-subjects design. The results confirmed the multidimensional nature of WB, and the need to consider positive and negative aspects of food-related WB, as well as differences linked to physical, emotional, and spiritual WB. Drawing on Study 1, 13 characteristics of food-related WB were identified, and in Study 2 their importance for feeling a ‘Sense of wellbeing’ and ‘Satisfied with life’ was determined with 1206 participants using a between-subjects design. In a further extension, Study 2 also adopted a product-specific perspective and explored the associations of, and importance, for 16 different foods and beverages to foodrelated WB. Based on Best-Worst Scaling and penalty/lift analysis, the four most important characteristics overall were ‘Is good quality,’ ‘Is healthy,’ ‘Is fresh,’ and ‘Is tasty,’ with the nuance that healthiness contributed most to feeling a ‘Sense of wellbeing’ while good quality contributed most to feeling ‘Satisfied with life.’ The associations to individual foods and beverages underscored that food-related WB is a complex construct arising from an overall evaluation of the different effects of foods (including physical health, social and spiritual aspects of food consumption) and their short-term effects on food-related behaviour. Contextual and individual differences in perceptions of WB in relation to food deserve further investigation.

Prior to the side effects in the first Elan/ Wyeth AD vaccine trial, we proposed using amyloid-β (Aβ) derivatives as a safer approach. The route of administration may also affect vaccine safety. To assess the feasibility of oral immunization that promotes mucosal immunity, Tg2576 AD model mice were treated prophylactically three times over 6 weeks starting at 3−5 months of age with a Salmonella vaccine expressing K6Aβ1−30. At 22 −24 months of age, cortical Aβ plaque burden and total Aβ40/42 levels were reduced by 48−75% in the immunized mice compared to controls, which received unmodified Salmonella. Plaque clearance was not associated with increased microglial activation which may be explained by the long treatment period. Furthermore, cerebral microhemorrhages were not increased in the treated mice in contrast to several passive Aβ antibody studies. These results further support our findings with this immunogen delivered subcutaneously, and demonstrate its efficacy when given orally which may provide added benefits for human use. Keywords

La agricultura posee un rol preponderante en el desarrollo humano, resultando de importancia sustancial la investigación que promueva su desarrollo y comprenda en su campo de acción el control de enfermedades que puedan afectar su rendimento y productividad o amenazar la salud humana como por ejemplo, la contaminación microbiana o la presencia de micotoxinas. En este sentido, la industria citrícola cumple con estrictas prácticas de sanitización durante el cultivo y packing de las frutas frescas de forma de prevenir y controlar la contaminación microbiana que puede causar el deterioro de la fruta. Sumado al deteriodo de la calidad de los frutos que son afectados por pestes en el campo, las pérdidas durante el almacenamiento y transporte de los frutos cítricos, provocadas directamente por ataques de hongos patógenos en postcosecha, han sido estimadas en un 90 %, en aquellos casos en que no se aplican fungicidas. Es por ello, que los productos químicos utilizados en la producción se han convertido en una herramienta que permite la obtención de niveles de rendimiento y calidad. El estudio de la dinámica de los residuos de pesticidas resultantes en las distintas etapas de la cadena productiva, su distribución en los productos procesados y en los compartimentos ambientales es crítica para comprobar el cumplimiento de las buenas prácticas agrícolas, la sustentabilidad del sistema productivo global y la inocuidad de los alimentos y sub-productos que resulten. Para realizar este estudio, la instrumentación de alta sensibilidad junto a modernos tratamientos de muestra, permiten trabajar con metodologías analíticas con límites de cuantificación a nivel de trazas. El objetivo de esta tesis fue estudiar la dinámica de los residuos de pesticidas en la cadena citrícola. Los objetivos específicos, contemplando la cadena globalmente fueron: 1) Desarrollo de metodologías analíticas y estudio de los factores que influyen en la eficiencia de las determinaciones como ser el Efecto Matriz; 2) Estudio de la disipación de pesticidas de precosecha en el campo; 3) Adecuación de los tratamientos de pre y poscosecha en las condiciones de producción para cumplir con las disposiciones vigentes y asegurar la inocuidad de los productos; 4) Estudio de la eficacia de nuevas alternativas para la disminución del uso de RESUMEN 19 fungicidas de poscosecha; 5) Caracterización de los niveles de residuos de pesticidas en los efluentes de los packings nacionales y estudio de posibles vías para su remediación empleando procesos de oxidación avanzada. Durante todo el período de trabajo en estos estudios de posgrado 1. Se desarrollaron y validaron metodologías analíticas para la determinación de residuos de pesticidas en fruta fresca (naranja, mandarina y limón), en aceites esenciales y en jugo concentrado de limón. Estas metodologías permiten determinar la mayoría de los pesticidas seleccionados a niveles adecuados para cada matriz estudiada, lográndose en todos los casos, límites de cuantificación menores que los límites máximos de residuos establecidos para cada matriz. Las metodologías desarrolladas se emplearon para el análisis de muestras reales así como también para determinar el residuo remanente sobre muestras de ensayos realizados a campo y en poscosecha. 2. Se desarrolló una base de datos de alta resolución para el monitoreo de residuos de pesticidas en naranja con un sistema instrumental de alta resolución GC-ToF. Por otro lado, uno de los aspectos que influyen en la determinación analítica de residuos de pesticidas con espectrometría de masas utilizando ionización por electospray es el Efecto Matriz, esto es la influencia de los coextractivos de la matriz en estudio sobre la determinación. El Efecto Matriz se estudió durante el análisis de pesticidas utilizando una metodología acetato de etilo desarrollada (que fuera previamente validada) y trabajando con un sistema instrumental de alta resolución LC-ToF. Los resultados obtenidos indican que para una correcta cantificación de residuos de pesticidas, es necesario trabajar con curvas de calibración preparadas en la misma matriz que la muestra o en su defecto, utilizando una variedad de la misma especie. Se pudo determinar que la coelucion de analitos y metabolitos coextraídos de la matriz, es una de las causas más importantes del Efecto Matriz y que este efecto de la coelución puede minimizarse al trabajar con masas exactas de alta resolución. 2. Se desarrollaron experimentos a campo sobre variedades de mandarinas y naranja para establecer las curvas de disipación de 5 pesticidas utilizados habitualmente en instancias de precosecha y para los mismos se establecieron los tiempos de vida media bajo las condiciones del estudio. Los resultados mostraron que abamectina, spinosad, piraclostrobina, difenoconazol e imidacloprid se disipan en las condiciones de la producción nacional, alcanzando concentraciones menores que sus respectivos LMR al RESUMEN 20 momento de la cosecha. Se evaluó también cómo influye el almacenamiento en frío de la fruta durante la poscosecha de mandarinas Clementinas, sobre el nivel de residuos de pesticidas que fueron aplicados a nivel de campo. El nivel de residuo luego de 30 días de almacenamiento en frío no fue significativamente diferente para los activos estudiados en dicha variedad. 4. Se evaluó la utilización de NaHCO3 conjuntamente con imazalil como tratamiento de poscosecha en la línea de empaque buscando disminuir la carga de pesticida a ser utilizado y de esta forma influir sobre la generación de resistencia y disminución de su impacto ambiental. La incorporación de la sal no mostró diferencias significativas entre los tratamientos ni en el nivel de residuo de imazalil obtenido sobre la fruta. 5. Se ajustó la determinación de pesticidas de poscosecha en efluentes de packings y se analizaron muestras reales. En base a los compuestos detectados, se evaluó a escala de laboratorio la utilización de fotocatálisis heterogena utilizando TiO2 como catalizador como herramienta para remediar efluentes. Los resultados obtenidos muestran una degradación de más del 97 % de cada pesticida trabajando con el estándar puro a una concentración de 10 mg L-1. Los aportes de este trabajo sirven, por un lado para comprender mejor aspectos de la dinámica de pesticidas en el marco de las actividades productivas y tecnologías que la industria citrica realiza, que llevan a obtener los niveles de residuos observados. Se corrobora en estas condiciones, el cumplimiento de los límites máximos de residuos y por lo tanto, la inocuidad de la fruta producida. Por otro lado, se plantean estrategias de remediación de los residuos de pesticidas generados en los packing cítricos para disminuir el impacto ambiental de la producción citrícola sobre el ambiente acuatico

<p>A new crystal structure for complex [Cu2Cl2(bipy)2(μ-Cl)(μ-CH3O)] was obtained. X-ray diffraction data showed dinuclear complexes with metal-ions seated on two symmetrically related pentacoordinated environment, in which, a trigonal bipyramidal<br />(TBPY-5) coordination polyhedra was observed. While chloride was detected as monodentate and as a bridging-ligand,<br />the diimine unit and a bridging oxygen from methoxide occupied the remaining positions. The crystal structure accounted for dinuclear complexes interacting to each other via H-bonds involving C–H groups of bipy and monodentate chloride anions of an adjacent molecule. Theoretical studies were conducted by DFT methods employing B3LYP in combination with 6-31G(d). Larger basis sets (6-311G(d) and cc-pVDZ) were taken into account to assess the performance of B3LYP/6-31G(d) in dealing with equilibrium geometry and population analysis. This standard methodology showed to be a reasonable performer to reproduce geometric parameters keeping the computational cost very low.</p>

The reaction of K2ReCl6 with 1,2-bis(2,3-dimercaptobenzamido)ethane (H4-1), and 1,2-bis(2,3-dimercaptobenzamido) benzene (H4-2) in the presence of Na2CO3 in methanol affords dinuclear complexes of ReV. Experimental evidence supports the presence of self-assembled complexes with two {Re(S2C6H3)3} units connected in a triple-stranded fashion. Density Functional Theory (DFT) studies on geometry and electronic properties were conducted employing the hybrid B3LYP and PBE1PBE functionals. The helical (DD and KK) and meso-helical (DK) isomers were considered. For the helicate complexes included in this study, differences in the stability of the isomers were observed originating in different steric and strain interactions between the three ligand strands. The geometries at the minimum exhibit a distorted trigonal-prismatic coordination environment at the metal centers. Natural bond orbitals (NBO) analysis indicates the presence of Re–S bonds which are strongly polarized toward the non-metal. Time-Dependent DFT (TD-DFT) calculations were performed for a further understanding of the optical spectra. The calculations show the occupied 5d orbitals of the rhenium lying beneath occupied sulfur-based MOs. The general features of the electronic spectra in the visible region are reasonably reproduced by the calculations. The analysis of molecular orbitals also allows the assignment of the origin for all experimentally detected absorption bands. In the high-energy region of the spectrum the absorptions are attributed to ligand-to-metal–ligand charge transfer (LMLCT), in which sulfur-based orbitals and unoccupied orbitals at the rhenium atom and the benzene-o-dithiolato groups are involved. Also in the blue region, shoulders originating from LMLCT are observed

<p>En los últimos años las biotransformaciones se han vuelto parte importante de la química sintética dadas las grandes ventajas que se le conocen, no solo por las condiciones suaves de reacción requeridas y la gran versatilidad que presentan; sino también por su alta quimio, regio- y estereoselectividad. Esto permite realizar transformaciones específicas sobre los sustratos y obtener productos que muchas veces no son accesibles por métodos clásicos, y principalmente permite obtener compuestos enantioméricamente puros de gran interés a nivel industrial. Entre las principales transformaciones en química orgánica se encuentran las reacciones de oxidación, lo cual ha impulsado el estudio de las oxidorreductasas como biocatalizadores de interés, y es dentro de este grupo que se encuentran las dioxigenasas tipo Rieske con las que se trabajó en esta tesis. Estas enzimas catalizan el primer paso para la degradación de compuestos aromáticos en bacterias, cuando son utilizados como fuente de carbono y energía. En 1968 el grupo del Dr. Gibson describió la actividad dioxigenasa en una cepa mutante de Pseudomonas putida y a partir de ese momento comenzaron los estudios de esta vasta familia de enzimas, a la cual se le reconoció un gran potencial en química sintética siendo utilizadas hasta el día de hoy para la producción de materiales de partida quirales. De los diferentes miembros de dicha familia de enzimas, el sistema Tolueno Dioxigenasa (TDO) es uno de los más utilizados a escala preparativa, pero no el más estudiado en cuanto a estructura del sitio activo y posible alteración de su regio- o estereoselectividad. En esta tesis se abordó el estudio de tales aspectos, seleccionando por alineamiento de secuencias aquellos residuos del sitio activo identificados como claves en otras enzimas tipo Rieske, como Naftaleno – y Bifenilo Dioxigenasas (NDO y BPDO). El desarrollo de cuatro variantes de TDO (Q215A, I324F, T365N y F366V) permitió evaluar la influencia de estos residuos en las propiedades de la enzima, identificando algunos importantes para la quimio- y estereoselectividad de la reacción de dihidroxilación. A su vez, se desarrolló el primer 10 modelo computacional del sitio activo de TDO, lo cual constituye una herramienta de suma importancia, tanto para la comprensión de los resultados obtenidos como para el desarrollo de trabajos futuros en el área. Por otra parte, se reportan aquí nuevas actividades para las dioxigenasas tipo Rieske.</p>

The kinetic isotope effects, KIEs, for hydrogen abstraction from the isotopologs CH4, 13CH4, CH3D, and CD4 by chlorine atoms have been studied by the dual-level direct dynamics approach with the MORATE computer program. Low-level calculations of the potential energy surface were carried out at the NDDO-SRP level (in particular the AM1-SRP level), using two different sets of specific reaction parameters labeled SRP4 and SRP13. High-level structural and energetic properties of the reactants, saddle point, and products were obtained at the MP2-SAC and MP2 levels using the 6-311G(2d,d,p) basis set and were used to interpolate corrections to the low-level calculations. The dual-level calculations were carried out using the ICL-Eckart improved interpolated corrections algorithm. Tunneling was included by the microcanonical optimized multidimensional tunneling (μOMT) method, and we find that large-curvature tunneling paths usually provide the dominant contribution, with significant participation of excited vibrational states. Both rectilinear and curvilinear coordinates were applied to the unsubstituted reaction. The 12C/13C KIEs calculated at the MP2-SAC///SRP4 level using MP2 frequencies are in very close agreement with the experimental ones, with values of 1.07 and 1.06 at 243 and 297 K, respectively, as compared to experimental values of 1.07 at both temperatures. For the reaction CH3D + Cl, the calculated H/D KIEs are equal to 1.55 and 1.45 at 223 and 296 K, while the measured values are equal to 1.59 and 1.50, respectively. The H/D KIE for the reaction CD4 + Cl is calculated to decrease from 11 to 4.7 as the temperature increases from 300 K to 450 K, whereas the experimental value decreases from 12 to 3.9. We also make comparison with previous results for the unsubstituted reaction CH4 + Cl.

The symbiosis of the Fourier transform spectrometer with the computer permits the development of novel procedures for handling spectroscopic data. Two such procedures are here described. The first one produces directly the spectrum of the intermolecular interaction perturbation in solutions, while the second obtains the spectrum of pressure broadening coefficients of a gas sample. The interpretation of these techniques are discussed in the light of experimental results and shown to be critically dependent on a constant, well defined slit function and on nearly perfect spectral and intensity registration.<br /><br />

<p>Almost all human cancers display dysregulated expression and/or function of one or more receptor tyrosine kinases (RTKs). The strong causative association between altered RTK function and cancer progression has been translated into novel therapeutic strategies that target these cell surface receptors in cancer. Yet, the full spectrum of RTKs that may alter the oncogenic process is not completely understood. Accumulating evidence suggests that a unique set of RTKs known as the discoidin domain receptors (DDRs) play a key role in cancer progression by regulating the interactions of tumor cells with their surrounding collagen matrix. The DDRs are the only RTKs that specifically bind to and are activated by collagen. DDRs control cell and tissue homeostasis by acting as collagen sensors, transducing signals that regulate cell polarity, tissue morphogenesis, and cell differentiation. In cancer, DDRs are hijacked by tumor cells to disrupt normal cell–matrix communication and initiate pro-migratory and pro-invasive programs. Importantly, several cancer types exhibit DDR mutations, which are thought to alter receptor function and contribute to cancer progression. Other evidence suggests that the actions of DDRs in cancer are complex, either promoting or suppressing tumor cell behavior in a DDR type/isoform specific- and context-dependent manner. Thus, there is still a considerable gap in our knowledge of DDR actions in cancer tissues. This review summarizes and discusses the current knowledge on DDR expression and function in cancer. It is hoped that this effort will encourage more research into these poorly understood but unique RTKs, which have the potential of becoming novel therapeutic targets in cancer.</p>

Conceptual 2D group III nitrides and oxides (e.g., 2D InN and 2D InO) in heterostructures with graphene have been realized by metal–organic chemical vapor deposition (MOCVD). MOCVD is expected to bring forth the same impact in the advancement of 2D semiconductor materials as in the fabrication of estab lished semiconductor materials and device heterostructures. MOCVD employs metal–organic precursors such as trimethyl-indium, -gallium, and -aluminum, with (strong) metal–carbon bonds. Mechanisms that regulate MOCVD processes at the atomic scale are largely unknown. Here, we employ density functional molecular dynamics – accounting for van der Waals interactions – to identify the reaction pathways responsible for dissociation of the trimethylindium (TMIn) precursor in the gas phase as well as on top-layer and zero-layer graphene. The simulations reveal how collisions with hydrogen molecules, intramolecular or surface-mediated proton transfer, and direct TMIn/graphene reactions assist TMIn transformations, which ultimately enables delivery of In monomers or InH and CH3In admolecules, on graphene. This work provides knowledge for understanding the nucleation and intercalation mechanisms at the atomic scale and for carrying out epitaxial growth of 2D materials and graphene heterostructures.

In this study, we report a strategy using dynamic combinatorial chemistry for targeting the thioredoxin (Trx)-reductase catalytic site on Trx glutathione reductase (TGR), a pyridine nucleotide thiol-disulfide oxido-reductase. We chose Echinococcus granulosus TGR since it is a bottleneck enzyme of platyhelminth parasites and a validated pharmacological target. A dynamic combinatorial library (DCL) was constructed based on thiol-disulfide reversible exchange. We demonstrate the use of 5-thio-2-nitrobenzoic acid (TNB) as a non-covalent anchor fragment in a DCL templated by E. granulosus TGR. The heterodimer of TNB and bisthiazolidine (2af) was identified, upon library analysis by HPLC (IC[Formula: see text] = 24 [Formula: see text]M). Furthermore, 14 analogs were synthetically prepared and evaluated against TGR. This allowed the study of a structure-activity relationship and the identification of a disulfide TNB-tricyclic bisthiazolidine (2aj) as the best enzyme inhibitor in these series, with an IC[Formula: see text] = 14 [Formula: see text]M. Thus, our results validate the use of DCL for targeting thiol-disulfide oxido-reductases.

The parasitic kinetoplastid diseases Leishmaniasis, Chagas disease and Human African Trypanosomiasis constitute serious threats for populations throughout the (sub-)tropics. Most available drugs to treat these diseases possess inadequate properties and candidates to fill the drug pipeline are urgently needed. Paullone-N5 -acetamides inhibit trypanothione synthe tase (TryS), an essential kinetoplastid enzyme, and exhibit antiparasitic activity in the low micromolar range, but lack the desired selectivity against mammalian cells (selectivity index (SI):<10). With the aim to identify the paullones’ moieties responsible for TryS inhibition and bioactivity, we applied molecular simplification and ring disconnection approaches. The new indolylacetamides lost activity against the expected molecular target (TryS) compared to the reference paullone MOL2008 (Leishmania infantum TryS IC50 :150 nM; Trypanosoma brucei bloodstream form EC50: 4.3 μM and SI: 2.4). However, several of them retained potency (T. b. brucei EC50: 2.4–12.0 μM) and improved selectivity (SI: 5 to >25).

<p>We report the design, the synthesis and the biological evaluation of the analgesic and anti-inflammatory activities of furoxanyl N-acylhydrazones (furoxanyl-NAH) by applying molecular hybridization approach. Hybrid compounds with IL-8-release inhibition capabilities were identified. Among them, furoxanyl- NAH, 17, and benzofuroxanyl-derivative, 24, together with furoxanyl-NAH derivative, 31, without IL-8 inhibition displayed both orally analgesic and anti-inflammatory activities. These hybrid derivatives do not have additional LOX- or COX-inhibition activities. For instance, LOX-inhibition by furoxanyl-NAH derivative, 42, emerged as a structural lead to develop new inhibitors. The lack of mutagenicity of the active derivatives 17, 31, and 42, allow us to propose them as candidates for further clinical studies. These results confirmed the success in the exploitation of hybridization strategy for identification of novel N-acylhydrazones (NAH) with optimized activities</p>
