The present report prepared by a Consultants' Meeting convened by the International Atomic Energy Agency in May, 1974, deals with general and specific problems associated with the utilization of accelerators in nuclear medicine. The report is intended to serve as a guide for potential entrants in the field. The different types of accelerators are surveyed with indications of both positive and negative features. In addition, some of the attendant problems of radioisotope and radiopharmaceutical production are addressed. The report includes a set of conclusions and recommendations as well as a selected annotated bibliography.<br /><br />

Previous research has shownthat consumers' sensory and hedonic perceptions could be greatly influenced by the messages highlighted on the front of the packaging, particularly nutrition and health claims for reduced-calorie or functional foods. In food products designed for hunger control, mentioning satiating effects or meal replacement could potentially influence the expected satiety or other perceptions, including those associated with a filling effect. This study investigated the effect of packaging information on consumer perceptions of eleven commercial chocolate-flavoured satiating and meal replacement bars. Projective mapping (Napping®) with a descriptive step was used to profile the expectations elicited by the eleven bars, based on their extrinsic characteristics (without tasting them), with two different groups of consumers in two different scenarios: mapping the samples' packaging and mapping cards with an identical design showing the most important information displayed on the packaging (product name, nutrition information panel and nutrition and health messages or claims). The terms and comments obtained in the two scenarios were analysed and perceptual spaces were generated from the sample location and attributes obtained in the Napping® exercises, using multi factor analysis (MFA). The results showed that when evaluating the real packaging, the consumers mainly focused on the meal replacement- or satiation-related messages and on the images to build up their perception, attaching less or no importance to the nutritional information that was also displayed on the box. When examining the packaging, sensory expectations – which probably emerged from the images of the bars – were also a very important factor in placing the samples in the perceptual space.When looking only at the cards, the consumers' grouping and attribute generation focused in great detail on the nutritional information. The calorie content also became a much more important factor in their perceptions of the bars. Interestingly for the product category analysed, messages about “satiating” and “meal replacement” effects were clearly distinct and negatively correlated in the consumers' minds.

The report of an AIEA meeting held in April 1976 reviews the work carried out by the participants of an IAEA-Coordinated Research Programme on Rapid Methods for the Quality Control of Radiopharmaceuticals. The applicability of the radiometric sterility testing and the limulus test for pyrogens is examined. The current status, advantages and limitations of these techniques are presented as well as the conclusions and recommendations drawn up by the participants. The IAEA coordinated work confirmed that both techniques are more rapid than the corresponding standard ones and are suitable for most radiopharmaceuticals.

En el presente trabajo se realizaron estudios para confirmar la existencia de lipoxigenasas activas en los hígados de merluza, enzimas que pueden ser las promotoras del deterioro del aceite contenido en los hígados. Cuando una solución conteniendo una mezcla de ácidos grasos obtenidos a partir del aceite de girasol se incubó con el agregado de un extracto acuoso obtenido a partir de los hígados de merluza, se verificó un gradual y continuo incremento en la concentración de hidroperóxidos en la mezcla. La velocidad de oxidación de los ácidos grasos en las incubaciones resultó directamente proporcional a la cantidad de extracto agregado, pudiendo ser eficientemente disminuida por el agregado de antioxidantes. Los microorganismos influyen de manera significativa en el proceso corrosivo y generan condiciones que afectan la velocidad y/o el mecanismo de deterioro. Su presencia se manifiesta por la formación de bio-películas: conglomerados de bacterias y polímeros extracelulares. Dichas bio-películas afectan la durabilidad del material, la velocidad de flujo y la transferencia de calor. En el presente trabajo se evalúa el crecimiento de microorganismos heterótrofos aerobios, heterótrofos anaerobios y bacterias sulfato-reductoras sobre latón aluminio (UNS C68700), latón almirantazgo (UNS C443) y acero inoxidable AISI 316. Asimismo, se estudia la influencia del crecimiento de la bio-película sobre el comportamiento corrosivo mediante técnicas electroquímicas: curvas de polarización y espectroscopia de impedancia electroquímica. Las exposiciones se realizan en la Bahía de Montevideo, estuario del Río de la Plata (Uruguay). A partir de los resultados obtenidos se establecen hipótesis acerca del mecanismo asociado, a la vez que se discuten criterios de selección de materiales en las condiciones particulares de exposición. Aunque el acero inoxidable AISI 316 muestra una velocidad media de corrosión inferior a los otros materiales ensayados, presenta una tendencia a la generación de fenómenos de deterioro localizado.

Then it describes the ways in which modelling, applications, and problem solving are approached in several Latin American countries. Based on the experience of these workshops, recommendations are made with respect to promoting modelling in the curricula.

The enzyme penicillin G acylase (PGA) is currently employed at an industrial scale in the hydrolysis of penicillin and cephalosporin G. Here, we describe the preparation of a new immobilized preparation of the enzyme that yields derivatives that are very thermostable and resistant in the presence of organic solvents. The stabilization is obtained via a double strategy. First, using a new epoxy support (Sepabeads), a high degree of multipoint covalent attachment is achieved. A threestep methodology is proposed: (1) immobilization on the support at high ionic strength, (2) promotion of an intense multipoint covalent attachment by incubation for a long time at alkaline pH, and (3) blocking hydrophylization of the support surface. These derivatives are more rigid and therefore more resistant to inactivation by both temperature or organic solvents. Second, the contact of the enzyme with the organic solvent molecules is reduced by generating a highly hydrophilic microenvironment using polymers (polyethylenimine, aldehyde-dextran, and sulfate-dextran). The exact protocol for the production of this environment has been found to be critical to achieve the desired results (final derivatives remain active in 90% of organic solvents). This derivative could be used in the hydrolysis of antibiotics even in media fully saturated with organic solvents, making simpler the current processes of production of 6-aminopenicillanic acid (6-APA) or 7-aminodeacetoxycephalosporanic acid (7-ADCA).

A summary of the resource.
The production, under environmentally benign conditions, of efficient and more cost-effective anti-infective agents (available to the whole mankind) is one of most ambitious dreams of the industrial medicinal chemistry. Semi-synthetic b-lactam antibiotics are very effective anti-infective agents. They are very stable and can be used via oral delivery. They exhibit a very wide spectrum of anti-bacterial activity and minimal side effects after being massively used for a very long time. In this way, we can assume that semi-synthetic b-lactam antibiotics are going to be one of the key anti-infective agents in the next years. The condensation of natural or modified antibiotic nuclei with different acyl donor chains is one of the key steps for the industrial synthesis of these anti-infective agents. Up to now, these condensations are mainly carried out through classical chemical methods and this implies a number of economical, ecological and technological drawbacks (high energy requirements, many protection and deprotection steps, utilization of toxic methylene chloride as solvent, etc). Enzyme biocatalysts may be very useful to catalyse these selective condensations under very mild experimental and environmental conditions. In fact, the possibility of using enzymes to carry out such biotransformations, at laboratory scale, has been discussed and demonstrated a long time ago. However, industrial synthesis of b-lactamic antibiotics is still carried out via unfavorable chemical routes. In fact, enzymes have not been produced in the nature to act in industrial reactors; so they are usually very unstable, inhibited by substrates and products and they may not have ideal catalytic properties for industrial uses (high reaction rates, required selectivity, ability to reach quantitative synthetic yields, stable enough to run a number of reaction cycles, etc). These limitations of enzyme biocatalysis become even more significant mainly when the enzyme is going to be used with non-natural substrates, catalysing non-natural processes and working under non-conventional conditions. However, in the last ten years, a great number of papers have reported substantial improvement of these enzymatic synthetic approaches (new enzymes, better enzyme derivatives, improved reaction designs and so on) and it seems that a massive industrial implementation of enzymes in antibiotic synthesis is approaching. In this review, we would like to make a critical discussion of these very interesting advances in the application of enzyme biocatalysts for the industrial synthesis of semi-synthetic antibiotics.

This paper described that glyoxyl activated supports are only able to quantitatively immobilize proteins at alkaline pH values and using highly activated supports (otherwise, immobilization is negligible). Furthermore, the immobilization rate depends on the surface density of glyoxyl groups and not on the total concentration of groups in the suspension. Moreover, the temperature presented a dramatic effect on enzyme immobilization rate and different enzymes become immobilized at very different rates. Finally, it was not possible to detect immobilization of mono aminated compounds at neither alkaline nor neutral pH values. In fact, these supports may be used to purify enzymes due to these great differences in immobilization rate (e.g., glutamate racemase). These special features are not shared with other supports usually utilized for covalent immobilization of enzymes (e.g., glutaraldehyde, cyanogen bromide supports) that are able to immobilize proteins by just one very stable bond and suggesting an unique mechanism for glyoxyl agarose supports. In fact, all these results suggested that each individual amino-glyoxyl bond is very weak. Therefore, the first immobilization of proteins on supports activated with glyoxyl groups, in absence of reducing reagents, needs the simultaneous establishment of, at least, two attachments between the protein and the support. This mechanism promotes that proteins become immobilized by the area/s where the highest lysine residues density is located and explain the high enzyme stabilization usually achieved by using this immobilization technique.

Se sintetizó y caracterizó siete nuevos complejos de vanadio(V) junto a una serie de otros compuestos ya conocidos. Estos compuestos noveles se obtuvieron por modificación de las técnicas originales. En general, la caracterización fisicoquímica y estructural de los compuestos se realizó mediante un conjunto de técnicas: análisis elemental, espectroscopía electrónica UV/Vis y vibracional, difracción de rayos X en polvo microcristalino y monocristal, medida de propiedades magnéticas y voltametría cíclica. En particular, se puso a punto una técnica espectrofotométrica de dosificación de vanadio de uso general, y se realizó un estudio vibracional profundo de los compuestos mediante espectroscopía IR y Raman, lo que constituye el principal aporte de este Trabajo de Tesis a la Química Inorgánica Básica. Además, se realizó la evaluación biológica de los compuestos sintetizados en diferentes líneas celulares y se estudió en detalle los efectos, a nivel celular, de dos compuestos seleccionados. En este Trabajo de Tesis se trabajó con tres grupos de compuestos: derivados de vanadio(IV) y vanadio(V) de la tropolona, peroxocompuestos de vanadio(V) y compuestos heterolépticos de vanadio(V) hidroxilamido-aminoácido. Para el caso de los derivados de la tropolona [VIVO(trop)2] y [VVO(trop)2Cl] (trop= ligando tropolonato), se desarrolló un método alternativo de síntesis del complejo [VVO(trop)2Cl]; fue analizado su comportamiento electroquímico mediante voltametría cíclica y las propiedades magnéticas del complejo [VIVO(trop)2]. Además, al igual que para las otras series, se estudió en profundidad su espectro vibracional mediante espectroscopía IR y Raman. A efectos de facilitar la asignación de las bandas del espectro vibracional se sintetizaron los complejos [M(trop)2] (donde M = Co, Cu, Zn). Para los complejos de vanadio(V) de ligando peroxo, K3[VO(O2)2CO3]∙H2O y K2[VO(O2)NTA]∙2H2O, se abordó el estudio de sus propiedades biológicas en las líneas celulares osteoblasto-símiles MC3T3-E1 (preosteoblasto normal) y UMR196 (células derivadas de osteosarcoma de rata). Se evaluó la capacidad antiproliferativa de los compuestos en ambas líneas celulares. También, se estudió la acción antiproliferativa del complejo K2[VO(O2)NTA]∙2H2O en células derivadas de adenocarcinoma de colon humano (Caco-2) y macrófagos de tumor murino RAW264.7. Por ser el ligando hidroxilamido (NH2O) isoelectrónico del peroxo, para algunos complejos de fórmula general [VO(NH2O)2(aa)]·nH2O (donde aa representa el ligando aminoácido), ha sido interesante evaluar sus propiedades biológicas en cultivos celulares, utilizando las líneas celulares anteriormente mencionadas. En este sentido se ha ensayado inhibición de la fosfatasa alcalina intestinal y proliferación, alteraciones de morfología, acción citotóxica, acción genotóxica y generación de estrés oxidativo en las líneas celulares osteoblasto-símiles. Dos de los compuestos fueron evaluados en un modelo in vivo de huevos de pez cebra

The Tn determinant (GalNAc-O-Ser=Thr), one of the most specific human carcinoma–associated structures, can be identified using both monoclonal antibodies and lectins. In this work we have explored, for the first time, the presence of Tn-binding proteins in aqueous extracts of plants collected in Uruguay. A total of 21 extracts from plants belonging to 12 Phanerogam families were screened by an ELISA assay (competitive inhibition of anti-Tn antibody binding) and by hemagglutination (HAG) activity. Five of the extracts displayed binding to Tn residues, although only those extracts prepared from fruit and leaf from Myrsine coriacea (Sw.) R. Br. ex Roem & Schult caused total inhibition of the binding activity of anti-Tn MAb 83D4. Considering that Myrsine coriacea extracts did not display HAG activity against several types of normal red blood cells, we characterized the material with Tn-binding activity from these extracts. We purified this material from Myrsine coriacea leaf extracts using perchloric acid treatment followed by affinity chromatography and reverse-phase high performance liquid chromatography (HPLC), conserving its Tnbinding activity. We found a high-molecular-weight glycoconjugate that has an apparent molecular mass of approximately 670 kDa, as judged by SDS-PAGE.

A summary of the resource.
The capacity of yeast to modify anthocyanin concentration during red wine fermentation was studied with the objective of classifying yeast strains according to their capacity for color removal during vinification. A model red grape juice medium was used to better understand yeast-anthocyanin interactions. An anthocyanin extract of Vitis vinifera cv. Tannat was prepared from grape skins and diluted in a white must in order to obtain a red grape juice without solids. Anthocyanin removal was confirmed to be higher for compounds with higher polarity. Acylated anthocyanins (acetyl and p-coumaryl compounds) were differentially removed, and the percentage of removal for each compound was determined. Results showed no correlation between color intensity and total anthocyanin concentration after fermentation, possibly because of the presence of anthocyanin derivatives formed during the process. HPLC-MS analysis allowed identification of anthocyanin derivatives, while the quantification of several derivatives was performed by HPLC-DAD. The sum of these derivatives showed a direct correlation with the color intensity obtained with each strain, thus explaining the color variability observed.

In this paper we report on the magnetic properties of pure bulk ferromagnetic graphite, obtained by a chemical route previously described. This magnetic graphite has been obtained by a vapor reaction consisting of a controlled etching on the graphite structure. By magnetic force microscopy we have verified that its magnetic properties are related to the topographic defects introduced in the pristine material. Also, the magnetic properties have been verified through magnetization measurements as a function of temperature and applied magnetic field. At low temperatures s2 Kd the saturation magnetization reaches a value of 0.58 emu/g, leading to a defect concentration of 1250 ppm. The system is highly irreversible due to the inhomogeneity of the distribution of defects in the material. Two transition temperatures are detected, Tc1=115s5d K and Tc2 =315s5d K. These transitions could be associated to the weak coupling between ferromagnetic regions related to defects and to the ferromagnetism inside the defect regions.

A summary of the resource.
As a contribution to the development of novel vanadium complexes with pharmacologically interesting moieties, new dioxovanadium( V) semicarbazone complexes with the formula cis-VO2L, where L = 5-bromosalicylaldehyde semicarbazone and 2-hydroxynaphtalen- 1-carboxaldehyde semicarbazone have been synthesized. and characterized by 1H and 13C NMR, Raman and FTIR spectroscopies. Results were compared with those previously reported for other three analogous complexes of this series. The five complexes were tested in three different human tumor cell lines for bioactivity as potential anti-tumor agents, showing selective cytotoxicity on TK-10 cell line. Results showed that structural modifications on the semicarbazone moiety could have a significant effect on the anti-tumor activity of the vanadium complexes. In addition, the electrochemical behavior of all the complexes was studied. No apparent correlation could be demonstrated between reduction potentials of the complexes and their anti-tumor activities. The molecular structure of the novel [VVO2(5-bromosalicylaldehyde semicarbazone)] complex was solved by X-ray diffraction methods. The vanadium atom shows a distorted square pyramidal coordination sphere. The (VO2)+ cation is coordinated to a nearly planar (L)� anion acting as a tridentate ligand through both oxygen and one nitrogen atoms.

In order to gain inside the mechanism of action of a series of benzo[1,2-c]1,2,5-oxadiazole N-oxide derivatives with in vitro anti-trypanosomal activity, electrochemical and biological studies were performed. Cyclic voltammetry and electron spin resonance spectroscopy were used to predict the bio-reduction processes. Effects on the parasitic respiration were studied.

In order to assess the utility of mtDNA typing for forensic purposes in Uruguay, a population sample of 120 maternally unrelated individuals were amplified and directly sequenced for the HV1 and for the HV2 segments in the control region of the human mtDNA, following previous international recommendations (1). Blood and saliva samples were taken from 120 unrelated individuals and deposited into FTA cards (FTA card, Whatman WB 12-0205). The DNA was extracted in a 1.2 mm punch according to the manufacturer recommendations. DNA amplification and sequencing was performed according to previously published paper (2). All statistical analysis were calculated using the Arlequin Software ver. 2.000 (3). Results are shown in Table 1. In this population sample, 107 different mtDNA sequences were observed out of 120 individuals. Of these, 97 sequences were observed once (80.8%), 8 sequences were observed twice (13.3%), one sequence was observed three times (2.5%), and another four times (3.3%). The variation of mtDNA HV1 and HV2 regions was confined to 146 positions, of which 99 was observed in the HV1 and 47 in the HV2. Sequence variation consisted of 120 transitions, 21 transversions, and 11 indels. The genetic diversity value for combined HV1 and HV2 regions was 0.998 (±0.002) in this Uruguayan population sample, and the nucleotide diversity was 0.068 (±0.035).
