The development of the biodiesel industry with the concomitant production of glycerol as the major byproduct has had a negative impact on the glycerol market. The application of circular economy concepts has led to progress in production processes that use this residue as starting material. Propylene glycol has been identified as a potential destiny of glycerol surplus; however, the possibility of increasing this valorization from the existence of a stereogenic center has been scarcely explored. Using metabolic engineering in E. coli, a set of biocatalysts capable of biotransforming glycerol to (R)-1,2-propanediol has been designed, built, and evaluated. The simultaneous expression of a methylglyoxal reductase (Gre2p) and an aldehyde reductase (Ypr1p) both from S. cerevisiae origin, along with the overexpression of E. coli methylglyoxal synthase (mgsA), resulted in an E. coli strain capable of producing enantiomerically pure (e.e.> 99%) (R)-1,2-propanediol from glycerol, in a concentration of 209 ± 9mg.L-1. The developed biocatalysts presented a satisfactory performance when crude glycerin obtained from biodiesel production process was used in place of glycerol.

Biodiesel generated by transesterification of triglycerides from renewable sources is a clean form of energy that is currently used in many countries in blends with petrodiesel. It is mainly produced from food-grade vegetable oils obtained from oleaginous crops. High prices of these oils have made the sustainability of biodiesel production questionable. The use of nonedible feedstocks, such as intracellular triglycerides accumulated by oleaginous yeasts, appears as a feasible alternative. However, it has been demonstrated that an economically sustainable production of yeast oil could only be possible if lowcost media based on industrial subproducts, or wastes are used. In this work, we propose intracellular lipids production by a previously selected oleaginous yeast strain in a medium composed only by sugar cane vinasse and crude glycerol. Different culture strategies were studied. The highest biomass and lipid yields were obtained when the yeast R. graminis S1/2R was cultivated in batch without control of dissolved oxygen. The fatty acid methyl esters obtained under these conditions met the specification of international biodiesel standards.

The use of biodiesel has generated a diversification of the energy matrix, which has led to a lower dependence on fossil fuels and a concomitant reduction of the adverse effects on the environment generated by their use. Currently, biodiesel is mainly produced by transesterification with short-chain alcohols of oils from different sources, in particular from oil crops, such as soybean and canola. Since the use of crops for the generation of fuels competes with their use as food, it seems appropriate to explore new oil sources for the production of biodiesel, such as the intracellular triglycerides accumulated by oleaginous yeasts. In this case, to achieve a competitive process, yeast production should be carried out using low-cost fermentable substrates such as industrial wastes or by-products. In this work we optimized the production of intracellular lipids by a previously selected oleaginous yeast strain in a fed-batch culture in presence of crude glycerol as a source of carbon and energy. In the optimized conditions an important improvement of lipid concentration was achieved, obtaining 12.2 g L−1 of fatty acid methyl esters (FAMEs) and 26.6 g L−1 of dry biomass in 360 h of culture.

Background Microbial oils produced by diverse microorganisms are being considered as alternative sources of triglycerides for biodiesel production. However, the standalone production of biodiesel from microorganisms is not currently economically feasible. In case of yeasts, the use of low-value nutrient sources in microbial production and the implementation of cost-efficient downstream processes could reduce costs and make microbial lipids competitive with other commodity-type oils in biodiesel production. Industrial biodiesel synthesis from oleaginous seeds is currently based on a multistep process. However, a simple process called in situ transesterification (ISTE), which takes place within the biomass without a previous lipid extraction step, is receiving increasing interest. In this work, the optimal conditions for an ISTE process to obtain biodiesel from previously selected oleaginous yeast (Rhodotorula graminis S1/S2) were defined using the response surface methodology (RSM). Results Using the RSM approach, the optimal conditions for the maximum yield with minimum reaction time included a methanol-to-biomass ratio of 60:1, 0.4 M H2SO4, and incubation at 70°C for 3 h. The optimized in situ process yield was significantly higher (123%) than that obtained with a two-step method in which fatty acids from saponifiable lipids were first extracted and then esterified with methanol. The composition of the fatty acid methyl ester mixture obtained from R. graminis S1/S2 by ISTE met Uruguayan standards for biodiesel. Conclusion The characteristics achieved by the optimized method make microbial oil a potential alternative for biodiesel production from yeast at an industrial scale. This work investigates and compares the reaction performance of soybean oil transesterification under supercritical methanol and ethanol, in a continuous catalyst-free process, as a cleaner alternative to conventional chemically catalyzed process. Reactions were performed in a tubular reactor, at 20 MPa, with oil to alcohol ratio of 1:40, varying the temperature in the range from 250 �C to 350 �C, and at two levels of water concentrations, 0 and 10 wt%. Although both processes proceeded with a relatively high reaction rate, conversion achieved by methanolysis was higher than that obtained by ethanolysis. Water positively affected both process: higher ester content and triacylglycerols depletion occurred when 10 wt% water was used compared with anhydrous conditions. Temperature increase favored the conversion of soybean oil to the corresponding methyl or ethyl esters, although temperatures above 300 �C increased the fatty acid degradation degree, a phenomenon responsible for the low ester contents obtained at the highest temperatures and lowest flow rates studied.

In this work different samples of Brazilian macauba oil obtained from mechanical pressing were characterized and production of esters of fatty acids using a catalyst-free continuous process under supercritical alcohols was assessed. Analysis of oil samples showed that the major fatty acid on pulp oil was oleic acid (mean value 62.8%), the amount of free fatty acid (FFA) was very high (37.4–65.4%), samples contained glycerides (7.4–16.5% TAG, 14.2–16.8% DAG and 1.0–3.4% MAG) and moisture was around 1.0%. Oil was processed in a continuous reactor using supercritical methanol or ethanol and the effects of temperature (573, 598, 623 and 648 K), pressure (10, 15 and 20 MPa), oil to alcohol molar ratio (1:20, 1:30 and 1:40), water concentration (0, 5 and 10 wt% added) and the flow rate of reaction mixture (1.0, 1.5, 2.0, 2.5 and 3.0 mL/min) on process efficiency were evaluated. The highest ester content achieved in reactions with supercritical methanol was 78.5% (648 K, 15 MPa, 1:30 oil:methanol molar ratio, 5 wt% water and 2.5 mL/min flow rate), while with supercritical ethanol was 69.6% (598 K, 15 MPa, 1:30 oil:ethanol molar ratio, 5 wt% water and 2.0 mL/min flow rate). The extent of the reaction was explored using a novel parameter, convertibility, which corresponds to the maximum ester content attainable from the feedstock. According to the convertibility of macauba pulp oil, the highest ester content corresponded to efficiencies of 98.0% and 86.9%, respectively. Results demonstrate that macauba oil might be a potential alternative for biodiesel production, though purification steps should be taken into account to achieve biodiesel specifications.

Phase-equilibrium diagrams of biodiesel synthesis substrate (oil/alcohol/alkyl esters) and product (alkyl esters/glycerol/alcohol) mixtures containing methanol, ethanol and propanol, respectively, were used for the identification of those composition ratios leading to mixture homogeneity. Based on the diagrams, complete conversion of the substrate oil contained in a sunflower oil/ ethanol/ethyl ester mixture at a weight ratio of 7.6:11.4:81.0 resulted in the generation of an amount of glycerol capable of remaining in solution in the product mixture. In contrast, complete conversion using a mixture of a lower alcohol and ethyl ester content (19.0:6.0:75.0) leads to the separation of phases. A continuous reactor charged with Lypozyme TL-IM and fed with the former mixture showed constant conversion and productivity with time, whereas process efficiency was found to decrease drastically when the latter mixture was fed. When a sunflower oil/isopropanol/isopropyl ester mixture at a weight ratio of 35:35:30 was fed in the reactor charged with Novozym 435, the output glycerol concentration was equivalent to that theoretically expected. In contrast, feeding a sunflower oil/ethanol/ethyl ester mixture of equal proportions as above, the glycerol concentration was lower than expected, suggesting the occurrence of glycerol adsorption on the enzyme support.

The cost of starting materials for the production of biodiesel is typically 75 % of the final retail price. Oils previously used for frying, waste frying oils (WFO), are a very suitable resource. Repetitive use of oil for frying foods involves high temperature, moisture and aeration for extended periods. The most important deterioration processes triggered by these conditions are hydrolysis and oxidation. In this study, 24 WFO samples of different origins were analyzed and classified as potential starting materials for biodiesel production using three quality parameters representing the main factors that affect the conversion of WFO. These parameters were: acid value, content of polar compounds and content of polymers, which varied in the ranges from 0.2 to 7.6, 14.9 to 43.2 and 0.9 to 15.2 %, respectively. Ester content obtained using conventional transesterification (TE) for WFO conversion decreased with increased levels of WFO deterioration determined by any of the three parameters noted above. TE products obtained had ester content between 81.4 and 95.7 %. Total ester content of a WFO sample with relatively low %AV could be increased to 96.5 % using a two-stage base catalysis TE. Finally, conversion of WFO samples resulted in ester contents of 89.0 and 91.3 %, respectively, when transesterified by conventional TE. After blending up to 10 % with refined oil, the ester content achieved was near 96.5 %. Thus, the blending represents an alternative for obtaining a product with suitable ester content.

A summary of the resource.
This work reports the production of fatty acid methyl esters (FAMEs) and fatty acid ethyl esters (FAEEs) by means of waste fried oil (WFO) transesterification using methanol and ethanol at supercritical conditions, in a continuous catalyst-free process. Transesterification experiments were performed from 573 to 623 K, at 10, 15, and 20 MPa, with oil/alcohol molar ratios of 1:20, 1:30, and 1:40 and water addition to alcohol of 0, 5, and 10 wt %. The extent of the reaction was explored using a novel parameter, convertibility, which corresponds to the maximum ester content attainable from the feedstock (92.1%). The highest FAME and FAEE contents achieved were 81.7 and 82.2%, respectively. Results show that transesterification of WFO in methanol was more efficient than that in ethanol, the temperature had the strongest influence, and the addition of water considerably improved the ester yield.

Disponer de un método de transesterificación de aceites eficiente en presencia de agua resultaría del mayor interés con vistas a la utilización de alcohol rectificado en lugar del metanol utilizado tradicionalmente para dicho fin. En este trabajo, se estudió el efecto sobre la eficiencia de la transesterificación en continuo en etanol supercrítico de los diferentes parámetros operativos, entre ellos el contenido de agua del alcohol. Los resultados indican que el proceso de conversión en biodiesel de los aceites de soja, girasol, girasol alto oleico y ricino puede realizars con relativa alta eficiencia si eligen convenientemente los parámetros operativos. Sin embargo, se encontró que el material lipídico sometido a las condiciones extremas de temperatura y presión requeridas sufrió un grado de deterioro significativo, produciendo la reducción del porcentaje de los ácidos grasos insaturados, fenómeno que aparece como la principal limitante del contenido de éster del producto.
