Cannabis use during pregnancy has recently become an important concern due to its increasing trend and association with neurodevelopmental alterations of exposed children. Although inhalation is the primary route of cannabis consumption in humans, few studies replicated this route in preclinical models of prenatal exposure. This study aimed to analyze the effects of chronic prenatal exposure to vaporized cannabis (PEVC) on hippocampal neurodevelopment, functionality, and learning and memory processes. Using a commercially available high-potency cannabis strain (THC 14.7%), pregnant rats were exposed to cannabis vapor from gestational day 8 to 21. A combination of primary hippocampal cultures, electrophysiological studies in slices, and behavioral tests was employed to assess the impact of PEVC. We found that PEVC induced presynaptic remodeling of hippocampal neurons in offspring under both basal and activity-dependent conditions by increasing synaptic vGlut1 abundance and the total recycling pool of synaptic vesicles. Chronic PEVC also induced a downregulation of CB1R at glutamatergic and GABAergic synapses. Additionally, an increase in axonal recruitment and synaptic efficacy was observed at hippocampal CA1 synapses of juvenile offspring. Moreover, long-lasting cognitive impairments were identified during adolescence, including deficits in spatial memory in male and female offspring. In summary, these findings demonstrate that exposure to high-potency cannabis vapor during pregnancy leads to significant changes in synaptic remodeling and efficacy, axonal recruitment, and long-term cognitive consequences in offspring.

While Cannabis sativa L. varieties have been traditionally characterized by their major cannabinoid profile, it is now well established that other plant metabolites can also have physiological effects, including minor cannabinoids, terpenes, and flavonoids. Given the multiple applications of cannabis in the medical field, it is therefore critical to characterize it according to its chemical composition (i.e., its metabolome) and not only its botanical traits. With this in mind, the cannabinoid and metabolomic profiles from inflorescences of two C. sativa varieties with either high D9 -tetrahydrocannabinolic acid (THCA) or high cannabidiolic acid (CBDA) contents harvested at different times were studied. According to results from HPLC and NMR-based untargeted metabolomic analyses of organic and aqueous plant material extracts, we show that in addition to expected variations according to cannabinoid profiles, it is possible to distinguish between harvests of the same variety. In particular, it was possible to correlate variations in the metabolome with presence of powdery mildew, leading to the identification of molecular markers associated with this fungal infection in C. sativa.

Abstract

We compared chemical profiles and in vitro biological activity of extracts obtained through maceration with ethanol or supercritical fluid extraction with CO2 (scCO2), from two different cannabis inflorescences chemotypes, one with high total tetrahydrocannabinol (THC) and other with high total cannabidiol (CBD) content. scCO2 was performed to obtain extracts with highest total THC and CBD recovery using ethanol as a co-solvent. In addition, a short maceration with ethanol was performed for both chemotypes. Cannabinoid and terpene composition of the extracts were determined using chromatographic and spectroscopic methods, and their cytotoxicity and cell proliferation effects were evaluated in vitro using HT-29 colon adenocarcinoma cell line. In vitro biological activity confirmed that cannabis extracts from both chemotypes have anti-proliferative activity but the scCO2 extract from high total THC chemotype was more cytotoxic. These results underscore the importance of chemotype and extraction method selection in the preparation of cannabis extracts for different therapeutic applications.

Abstract

Choosing an appropriate treatment for chronic pain remains problematic, and despite the available medication for its treatment, still, many patients complain about pain and appeal to the use of cannabis derivatives for pain control. However, few data have been provided to clinicians about the pharmacokinetic drug-drug interactions of cannabinoids with other concomitant administered medications. Therefore, the aim of this brief review is to assess the interactions between cannabinoids and pain medication through drug transporters (ATP-binding cassette superfamily members) and/or metabolizing enzymes (cytochromes P450 and glucuronyl transferases).
