BACKGROUND: The global beef market demands the meat industry to ensure product quality and safety in markets that are often very distant. The present study aimed to evaluate the effects of chilled (CH, 120 d) and chilled-then-frozen (CHF, 28 d + 92 d) storage conditions of beef vacuum packaged (VP) and vacuum packaged with antimicrobial (VPAM) on meat quality, oxidative status and microbial loads. Treatments resulted from the combination of storage condition and packaging type: VP + CH, VP + CHF, VPAM + CH and VPAM + CHF. RESULTS: Warner–Bratzler shear force values decreased in all treatments after 28 d of chilling. Except for VP + CH, L* values (lightness) of meat color did not differ in each treatment as the storage time increased. Meat from VP + CH had greater a* values than CHF treatments on day 120 of storage. A consumer panel did not detect differences in tenderness, flavor and overall liking between VP and VPAM beef, but they preferred CHF steaks rather than CH beef. TBARS values did not differ between VP and VPAM and between CH and CHF at any time during the storage period. At the end of storage time, all treatments except VP + CHF presented a greater concentration of thiols than at 48 h post-mortem. On day 120 of storage, VP + CH had greater cat alase enzyme activity than CHF treatments while VP + CH and VP + CHF showed a greater superoxide dismutase activity than VPAM + CHF. Storage condition (CH or CHF) had a greater impact on microbial counts than the type of packaging. CONCLUSION: Freezing meat after an ageing period represents a suitable strategy to extend beef storage life without a detri mental impact on its quality.

A summary of the resource.
Copper (Cu), iron (Fe) and zinc (Zn) bioaccessibility was studied in beef, chicken, chickpeas, lentils, quinoa, soybeans, and tuna fish using an in vitro assay. Samples were cooked as usually consumed locally. Analytical determinations were performed by atomic absorption spectrometry. The samples that presented the highest bioaccessible fraction (BF) were soybeans (~87 %) (Cu), chickpeas (~78 %) (Fe), and quinoa (~92 %) (Zn), respectively. Zinc was the mineral that presented the highest BF value of all, being above 66 % in all food samples. The effect of garlic and onion on the BF was also evaluated, being the highest effects observed for quinoa with a percent increase of 26.9 % (Fe) promoted by garlic and 19.7 % (Zn) promoted by onion. The contribution of Cu, Fe and Zn to the recommended dietary allowance (RDA) for adults over 19 years was esti mated, considering a 200 g portion. Soybeans and lentils contributed between 56.7 % and 78.9 % (men and women) to Cu RDA, chickpeas, lentils, and soybeans contributed an average of 34.6 % (men) and 15.4 % (women) to Fe RDA, while beef contributed an average of 65.5 % (men) and 90.1 % (women) to Zn RDA. This information shows that beef is a major source of Zn to the diet.

A summary of the resource.
A telephone survey (n = 601) was conducted in Uruguay to explore participants’ mental associations with meat and the underlying reasons changes in the quantity and type of meat consumed. Participants mentioned mostly positive associations with meat’s culinary and cultural aspects and its nutritional value as source of high-quality protein. Other associations were related to meat production, specific types of meat and cuts, meat consumption and pleasure, while price was perceived negatively. Animal welfare concerns emerged as a relevant association, but minor references were made to the effects of meat production on the environment. Changes in the amount and type of meat consumed (higher pork, chicken and cheaper cuts and beef) in the last year were mainly due to price followed by health reasons. Results emphasize the strong meat culture among Uruguayans and reveal that changes in the amount and type of meat consumed last year relate to external (price) or egoistic (health) rather than altruistic motives (ethical or environmental concerns).

<p>Cu and Zn bioaccessibility was studied in two different beef cuts (rib plate and top sirloin), through the development of an in vitro assay. Samples were cooked at two degrees of doneness (medium and well-done) as usually consumed locally, to establish the contribution to the recommended daily allowance (RDA). Simulated gastrointestinal conditions were optimized and trueness of the method was evaluated by performing mass balances. Analytical determinations were performed by atomic absorption spectrometry and microwave-induced plasma optical emission spectrometry. No significant differences were found between the studied cuts nor between the degrees of doneness evaluated. Mean bioaccessible fraction (BF) of Zn was 83%, for both cuts and both degrees of doneness. In the case of Cu, rib plate presented a mean BF of 20%, while for top sirloin was of 38%. However, these differences found between cuts and degrees of doneness were not statistically significant. The contribution of Cu and Zn to the RDA for adults over 19 years was estimated, as an example, considering a 200 g portion. Results showed that the RDA of Zn would be completely covered, while the contribution to the RDA of Cu would be up to 33%.</p>

En el presente trabajo se evaluó la influencia del grado de cocción en la retención de los micronutrientes cobre (Cu), hierro (Fe), hierro hemo (Fe-Hemo) y cinc (Zn) en tres cortes de carne vacuna de consumo en Uruguay, diferentes estructuralmente (asado, cuadril y bola de lomo) provenientes de 40 animales del territorio nacional. Se estudió la variación de estos elementos según el corte (evaluando el diferente aporte nutricional) y según el punto de cocción (tomando como medida del grado de avance en el proceso de cocción, la pérdida de masa de la muestra luego del tratamiento térmico respecto a la misma cruda).<br />Las muestras de carne vacuna (provistas por el Instituto Nacional de Carnes, INAC), se recibieron envasadas al vacío con 36 horas de maduración sanitaria. A cada una de ellas luego de retirarse el tejido óseo y adiposo visible se la dividió en tres porciones, una de las cuales fue analizada cruda y las otras dos fueron sometidas al mismo proceso de cocción (que consistió en un sellado en una plancha a 300ºC hasta dorar cada una de las caras de las piezas y se culminó el proceso en horno a 200ºC, siendo ambos equipos eléctricos).<br />Se trabajó en dos puntos (o grados) de cocción en cada muestra, que fueron establecidos evaluando la pérdida de masa respecto a la misma cruda, para lo cual se monitoreó el proceso mediante la utilización de una termocupla de pincho, que se introdujo en el centro de los cortes. Cabe mencionar que durante las etapas del proceso térmico no se agregó ningún aditivo a las porciones de carne analizadas.<br />El tratamiento de las muestras consistió en una molienda y homogeneización (tanto de las muestras crudas como las cocidas) con un molino de cuchillas, un posterior secado de las mismas hasta peso constante, mediante el uso de una estufa a 105ºC entre 15 y 18 horas (AOAC 950.46).<br />Para el análisis de elementos inorgánicos en matrices complejas como lo es la carne, es necesario realizar un tratamiento previo de la muestra para la extracción simultánea y cuantitativa de los minerales. En esta etapa de mineralización se requiere eliminar la totalidad de la material orgánica, para lo cual se realizó una digestión asistida con microondas, adicionando 10 mL de HNO3 4,2 mol L-1 a 0,5 g de carne (molida y seca) con la siguiente programación: potencia 400-1800 W; rampa de 15 minutos hasta alcanzar los 200ºC y mantenimiento a dicha temperatura durante 15 minutos. Una vez finalizada la digestión se realizaron las diluciones adecuadas con agua tipo I (ASTM).<br />Las determinaciones de los elementos cobre (Cu), hierro (Fe) y cinc (Zn) se realizaron mediante espectrometría de absorción atómica de llama (Flame Atomic Absorption Spectrometry, FAAS) para los primeros cuatro muestreos (n=20) y mediante espectrometría de emisión óptica con plasma inducido por microondas (MIP-OES) en las muestras restantes (n=20). Los métodos analíticos fueron validados y resultaron adecuados para el fin definido.<br />Los niveles de Cu, Fe y Zn encontrados en la carne vacuna cruda uruguaya son concordantes con los reportados a nivel nacional, los cuales informan niveles de Cu en el rango de (0,019 – 0,12) mg / 100 g de carne vacuna cruda, niveles de Zn entre (2,0 -7,27) mg / 100 g, de Fe entre (1,42 - 4,82) mg / 100 g y de Fe Hemo de (1,4 - 3,3) mg / 100 g (todos los valores están expresados en base fresca).<br />La tendencia observada en las muestras analizadas durante todo el estudio fue de un mayor nivel de oligoelementos en el corte bola de lomo, lo cual le brindaría un mayor contenido nutricional en cuanto a los minerales determinados. El corte cuadril presentó un nivel significativamente menor de Zn respecto a los otros dos cortes, pero a pesar de ello se determinó que este alimento aún luego del proceso de cocción con calor seco es una buena fuente de Cu y posee alto contenido de Fe Hemo, Fe y Zn.<br />La mayor parte del hierro presente en este alimento está bajo su forma más biodisponible (Fe Hemo), lo que hace fundamental su presencia en la dieta humana para satisfacer las necesidades de los individuos y promover un desarrollo humano saludable. Para la determinación del mismo se optimizó un procedimiento basado en el método empírico de Hornsey (1956) ampliamente referenciado, el cual es recomendado utilizar por la Unión Europea como forma de armonizar criterios. Si bien a nivel nacional e internacional existen trabajos que reportan el contenido de hierro hemo en carne vacuna cruda, muy pocas lo hacen sobre la influencia de la cocción en sus niveles. Hasta el momento a nivel nacional no se han encontrado reportes sobre los porcentajes de retención de hierro hemo en carne vacuna luego del proceso de cocción, por lo cual este trabajo aporta nueva información y actualizada sobre las variaciones del nivel nutricional en las carnes vacunas uruguayas al ser sometidas a este proceso de cocción en particular.<br />La carne vacuna es una fuente importante de micronutrientes pero se sabe poco acerca de cómo y en qué medida afectan los procesos de cocción su valor nutricional. Si bien el contenido de minerales analizado varió con la cocción utilizada en este trabajo, las pérdidas por lixiviación no fueron significativas hasta una pérdida de peso aproximada de 15% (para el punto poco cocido), a partir del cual este porcentaje aumenta. La evaluación de la influencia del proceso de cocción en el contenido de nutrientes se realizó mediante el cálculo del porcentaje de retención mediante la utilización de la ecuación descrita por Bognár y Piekarski (2000). Estos porcentajes de retención se pueden utilizar para calcular el contenido nutricional de este alimento de forma rápida y sencilla.

<p>La calidad de la carne vacuna está definida por la cantidad de nutrientes que aporta al consumirla, entre otros aspectos. En este sentido, la determinación de elementos esenciales como zinc (Zn), cobre (Cu), molibdeno (Mo) y selenio (Se), entre otros, proporciona información relevante para dar un valor agregado al alimento. Por otro lado, es necesario garantizar su inocuidad, ya que contaminantes inorgánicos como cadmio (Cd) y plomo (Pb), entre otros, pueden estar presentes y provocar daños a la salud humana. Por lo general, la determinación de metales y semimetales en matrices complejas consiste en mineralizaciones utilizando ácidos minerales concentrados en<br />caliente o mediante calcinación. Ambos procesos son tediosos y además poco amigables con el ambiente. En esta tesis se exploró una nueva estrategia de preparación de muestras de carne vacuna, para dos cortes de gran consumo en el país, como son el asado y el cuadril, bajo la hipótesis de que el uso de hidróxido de tetrametilamonio (TMAH) sería eficiente para la extracción cuantitativa de los elementos en estudio en una sola etapa. La optimización del método propuesto se realizó mediante un diseño experimental multivariado de tipo central compuesto, de dos variables a cinco niveles, utilizando un material de referencia certificado de músculo bovino. Las variables estudiadas consideradas críticas la metolodogía fueron: la concentración de TMAH y la temperatura de extracción. Las determinaciones de Zn y Cu se realizaron mediante espectrometría de absorción atómica de llama (FAAS). Por su parte, las determinaciones de Cd, Pb y Mo se<br />realizaron mediante espectrometría de absorción atómica con atomización electrotérmica (ETAAS).</p>
<p>Por otra parte, si bien el consumo de carne vacuna puede constituir una fuente importante de los elementos estudiados, es importante lograr establecer la cantidad de nutrientes que se encuentran potencialmente accesibles para ser absorbidos luego del proceso digestivo. Por este motivo, también se estudió la bioaccesibilidad de Cu y Zn en asado y cuadril, dado que dentro de los elementos seleccionados estos son los que se encuentran en mayor proporción, mediante el desarrollo de un ensayo in vitro, simulando los procesos que ocurren típicamente en el sistema digestivo humano: en el estómago y el intestino delgado. Estos ensayos se realizaron sobre una<br />serie de muestras de carne cocida, en dos puntos de cocción diferentes, con el fin de estudiar su aporte a la ingesta diaria recomendada (IDR) de dichos elementos en la forma en que puede ser consumido el alimento. Se optimizaron las condiciones gastrointestinales simuladas y se evaluó la<br />veracidad del método mediante la realización de los balances de masa correspondientes. Las determinaciones se Zn se realizaron mediante FAAS, mientras que las de Cu se realizaron mediante ETAAS.</p>
<p>En cuanto al desarrollo del método propuesto, éste consistió en el agregado de 0,25 g de muestra seca, 1,50 mL de TMAH 25% m/v en metanol y 5,00 mL de agua ultrapura, y posterior<br />calentamiento en baño termostatizado a 85 ºC durante 60 min. Los parámetros de desempeño obtenidos resultaron adecuados para los fines propuestos. Los límite de detección (LD)/límites de cuantificación (LC) expresados en mg kg-1 (base seca) fueron: 0,016/0,052 – 0,013/0,044 – 0,015/0,048 – 0,008/0,027 – 0,06/0,2 para Zn, Cu, Mo, Cd y Pb, respectivamente. La veracidad expresada como porcentaje de recuperación R(%) se encontró entre 81 y 106%. La precisión<br />expresada como desviación estándar relativa RSD(%) fue menor al 10% en todos los casos. Luego de la validación, se analizaron muestras de carne vacuna cruda. Las muestras de asado y cuadril fueron proporcionadas por el Instituto Nacional de Carnes (INAC), gracias a un proyecto de trabajo en conjunto con el sector productivo (CISC - VUSP). Con el método aquí desarrollada, se obtuvieron resultados estadísticamente equivalentes a los obtenidos mediante un método de digestión clásico asistido por microondas, para Zn, Cu y Mo. Ninguna de las muestras estudiadas presentó niveles cuantificables de Cd y Pb, por lo que se pudo garantizar la inocuidad de estas desde el punto de vista de estos contaminantes, encontrándose muy por debajo de los límites máximos establecidos por la normativa nacional.</p>
<p>El método desarrollado resultó adecuado para los fines propuestos y puede proponerse como una alternativa a los métodos estándar, siendo además simple y con la generación de muy baja cantidad de residuos peligrosos, alineándose de esta forma con los principios de la Química Analítica Verde. Sin embargo, no fue posible la determinación de Se utilizando este tratamiento alcalino. En este caso, se realizó una digestión ácida asistida por microondas y la posterior determinación mediante espectrometría de masas con plasma acoplado inductivamente (ICP-MS).</p>
<p>En lo que refiere a las fracciones bioaccesibles de Cu y Zn, no se encontraron diferencias significativas entre ambos cortes, ni entre los puntos de cocción evaluados. En el caso de Zn, para<br />asado y cuadril, poco y muy cocido, la fracción bioaccesible fue del 83%. En el caso de Cu, el asado presentó una fracción del 20% , mientras que el cuadril presentó una fracción del 38%.</p>
<p>A modo de ejemplo, considerando una porción de 200 g de carne cocida, se aportaría el total de la IDR de Zn requerida para un adulto, mientras que se aportaría un 33% de la IDR de Cu para un adulto. Estos datos constituyen información que aporta al conocimiento de la calidad nutricional de la carne uruguaya como alimento en forma cocida.</p>

Se determinó el contenido de selenio (Se) antes y después del proceso de cocción en tres cortes de carne vacuna (asado, cuadril y bola de lomo) provenientes de 20 animales. Los distintos cortes fueron sometidos a dos puntos de cocción. Cada corte fue dividido en varias porciones y se realizó la cocción en plancha y horno. El tratamiento de las muestras consistió en una digestión asistida con microondas y posterior determinación de Se mediante espectrometría de emisión atómica con plasma inducido por microondas acoplado a generación de hidruros (HG-MIP OES). El método analítico propuesto fue validado, resultó adecuado para el fin definido y presentó ventajas respecto a los métodos habitualmente utilizados en los laboratorios de análisis. Los puntos de cocción se establecieron evaluando la pérdida de peso (desde 8 hasta 35%). Los resultados indican que la carne vacuna aporta entre 20 y 26% del requerimiento diario de Se de un adulto. Este porcentaje se mantiene sin cambios significativos aun luego de una pérdida de peso de hasta un 35%, por lo que se concluye que la cocción no influye en el aporte de selenio a la dieta.

Este trabajo describe la optimización de un método espectrofotométrico clásico (“método Hornsey”) para determinar hierro hemo en carnes. Este método extrae hierro hemo de la carne mediante agitación con acetona ácida, maceración en la oscuridad y determinación espectrofotométrica a 640nm sobre el filtrado. Es ampliamente utilizado, pero su descripción original no detalla la influencia de algunas variables. En numerosos trabajos se determina hierro hemo según el “método Hornsey” utilizando diferentes estrategias de agitación y tiempos de incubación diversos. Estas variables se optimizaron mediante un diseño multivariado de tipo central compuesto. Con el método optimizado se analizaron distintos cortes de carne vacuna para evaluar su valor nutricional en cuanto a niveles de hierro biodisponible. A partir de una optimización racional, se obtuvieron las condiciones experimentales adecuadas para realizar un método analítico, considerado de referencia, de forma sistemática y protocolizada en los laboratorios para evaluar el contenido de hierro hemo en carnes.
