La alta incidencia de floraciones de cianobacterias productoras de microcistinas en el país y la región representa un riesgo muy elevado para humanos y animales. A fin de estudiar el impacto y la presencia de las microcistinas (MCs) en animales, es importante disponer de métodos simples de bajo costo. Como primera aproximación a estos objetivos en peces, se estudiaron Astraloheros facetus (Castañetas) expuestas a una floración de Microcystis spp (MCs 60 y 600 μgMCs/L) en un bioensayo subcrónico (18 días). Si bien no hubo mortalidad, la histopatología mostró infiltración grasa en el hígado, más relevante en los peces expuestos a la mayor concentración. Para analizar MCs en pescados se optimizaron dos métodos inmunoquímicos sensibles basados en un anticuerpo recombinante de llama (nanobody) de alta especificidad: ELISA y MALDI-TOF cuantitativo, utilizando partículas magnéticas funcionalizadas. Los métodos fueron recientemente desarrollados localmente. La excelente correlación ELISA/MALDI-TOF (rSpearman = 0,988, p< 10-7) resalta el potencial de este ELISA como herramienta simple y costo-efectiva para minimizar las muestras a analizar por métodos de referencia. Las concentraciones de MCs en las Castañetas fueron relevantes, acordes con bioensayos en otras especies y peces de la naturaleza. Esto destaca la importancia de analizar MCs en pescado para consumo

Freshwater harmful algal blooms produce a broad array of bioactive compounds, with variable polarity. Acute exposure to cyanotoxins can impact the liver, nervous system, gastrointestinal tract, skin, and immune function. Increasing evidence suggests chronic effects from low-level exposures of cyanotoxins and other associated bioactive metabolites of cyanobacterial origin. These sundry compounds persist in drinking and recreational waters and challenge resource managers in detection and removal. A systematic approach to assess the developmental toxicity of cyanobacterial metabolite standards was employed utilizing a robust and high throughput developmental Danio rerio embryo platform that incorporated a neurobehavioral endpoint, photomotor response. Subsequently, we applied the platform to cyanobacterial bloom surface water samples taken from temperate recreational beaches and tropical lake subsistence drinking water sources as a model approach. Dechorionated Danio rerio embryos were statically immersed beginning at four to six hours post fertilization at environmentally relevant concentrations, and then assessed at 24 h and 5 days for morbidity, morphological changes, and photomotor response. At least one assessed endpoint deviated significantly for exposed embryos for 22 out of 25 metabolites examined. Notably, the alkaloid lyngbyatoxin–a resulted in profound, dose-dependent morbidity and mortality beginning at 5 μg/L. In addition, hydrophobic components of extracts from beach monitoring resulted in potent morbidity and mortality despite only trace cyanotoxins detected. The hydrophilic extracts with several order of magnitude higher concentrations of microcystins resulted in no morbidity or mortality. Developmental photomotor response was consistently altered in environmental bloom samples, independent of the presence or concentration of toxins detected in extracts. While limited with respect to more polar compounds, this novel screening approach complements specific fingerprinting of acutely toxic metabolites with robust assessment of developmental toxicity, critical for chronic exposure scenarios.

Owing to their reproducibility, stability, and cost-effective production, the recombinant variable domains of heavy-chain-only antibodies (VHHs) are becoming a salient option as immunoassay reagents. Recently, there have been several reports describing their application to the detection of small molecules (haptens). However, lacking the heavy-light chain interface of conventional antibodies, VHHs are not particularly apt to bind small analytes and failures are not uncommon. Here we describe the construction of a VHH phage display library against the cyanobacterial hepatotoxin microcystin LR and its selection using competitive panning and two novel panning strategies. The outcome of each strategy was evaluated by a large-scale screening using in vivo biotinylated nanobodies. The three methods selected for different nonoverlapping subsets of VHHs, allowing one to optimize the immunodetection of the toxin. The best results were obtained by promoting the isolation of VHHs with the slowest koff (off-rate selection). Among these, the biotinylated nanobody A2.3 performed in ELISA with excellent recovery and high sensitivity, IC50 = 0.28 μg/L, with a limit of detection that is well below the most rigorous guidelines for the toxin. While it may be case-specific, these results highlight the importance of exploring different panning strategies to optimize the selection of antihapten nanobodies.

Blooms of Nostocales (Cyanobacteria) are thought to be invading subtropical and temperate water bodies. According to nutrient stoichiometry and physiological differences between cyanobacterial groups, the replacement of non-heterocystous species by Nostocales is favored when dissolved inorganic nitrogen decreases. However, some studies have shown different trends. We used laboratory bioassays to evaluate the concomitant effects of light and nutrient enrichment on phytoplankton assemblages dominated by non-heterocystous filamentous cyanobacteria. Three nutrient conditions (no addition, addition of phosphate, and addition of nitrate and phosphate) and two light intensities (40 and 80 µmol photon m−2 s−1) were assayed. Nostocales replaced or co-dominated with non-heterocystous species in all treatments by the end of the study. The shift in community composition towards Nostocales dominance led to an increase in species richness, which suggests that species with different eco-physiological traits may have differential impacts on diversity. The highest saxitoxin concentrations were measured in no addition treatments, which could link production to nutritional stress. Nostocales featured high phenotypic plasticity in terms of changes in average trichome volume and growth rates. These results help to have a better understanding of the conditions under which Nostocales predominance can occur.

The harmful bloom-forming cyanobacterium Cylindrospermopsis raciborskii grows in freshwaters over a wide range of light conditions. This species has increased its global distribution recently. The influence of ultraviolet radiation (UVR) on the fitness and toxin production of C. raciborskii has not previously been explored. We performed short-term experiments with three C. raciborskii strains (MVCC19, LB2897, and CYP011 K), and we compared their responses with other bloom-forming species (Microcystis sp.1 and Plankthotrix agardhii) to determine the impact of UV-B radiation on pigments, biomass, and morphological traits. In addition, we analyzed the effect of UV-B on the saxitoxin content and sxtU gene expression in the strain MVCC19. C. raciborskii strains were stressed differentially by UV-B exposure as evidenced by changes in growth, morphology, and heterocytes number. A significant increase in saxitoxin concentration and sxtU gene expression under UV-B suggests that toxin production in C. raciborskii can be a response to UV-B stress. In comparison, Microcystis sp.1 was more tolerant, while P. agardhii was severely impacted by UV-B, indicating also different sensitivities among cyanobacteria to UVR. Our results underscore the influence of UVR on C. raciborskii and the differences between strains which showed phenotypic plasticity, which potentially could affect its distribution in freshwaters.

Cyanobacterial 16S ribosomal RNA gene diversity was examined in a benthic mat on Fildes Peninsula of King George Island (62809054.400S, 58857020.900W), maritime Antarctica. Environmental DNA was isolated from the mat, a clone library of PCR-amplified 16S rRNA gene fragments was prepared, and amplified ribosomal DNA restriction analysis (ARDRA) was done to assign clones to seven groups. Low cyanobacterial diversity in the mat was suggested in that 83% of the clones were represented by one ARDRA group. DNA sequences from this group had high similarity with 16S rRNA genes of Tychonema bourrellyi and T. bornetii isolates, whose geographic origins were southern Norway and Northern Ireland. Cyanobacterial morphotypes corresponding to Tychonema have not been reported in Antarctica, however, this morphotype was previously found at Ward Hunt Lake (838N), and in western Europe (528N). DNA sequences of three of the ARDRA groups had highest similarity with 16S rDNA sequences of the Tychonema group accounting for 9.4% of the clones. Sequences of the remaining three groups (7.6%) had highest similarity with 16S rRNA genes of uncultured cyanobacteria clones from benthic mats of Lake Fryxell and fresh meltwater on the McMurdo Ice Shelf.

In recent years, the international demand for commodities has prompted enormous growth in agriculture in most South American countries. Due to intensive use of fertilizers, cyanobacterial blooms have become a recurrent phenomenon throughout the continent, but their potential health risk remains largely unknown due to the lack of analytical capacity. In this paper we report the main results and conclusions of more than five years of systematic monitoring of cyanobacterial blooms in 20 beaches of Montevideo, Uruguay, on the Rio de la Plata, the fifth largest basin in the world. A locally developed microcystin ELISA was used to establish a sustainable monitoring program that revealed seasonal peaks of extremely high toxicity, more than one-thousand-fold greater than the WHO limit for recreational water. Comparison with cyanobacterial cell counts and chlorophyll-a determination, two commonly used parameters for indirect estimation of toxicity, showed that such indicators can be highly misleading. On the other hand, the accumulated experience led to the definition of a simple criterion for visual classification of blooms, that can be used by trained lifeguards and technicians to take rapid on-site decisions on beach management. The simple and low cost approach is broadly applicable to risk assessment and risk management in developing countries
