<dcterms:abstract><![CDATA[Thermophilic oleate-degrading bacteria were enriched in mineral medium in association withMethanobacterium thermoautotrophicum . The enrichment culture also produced methane from linear saturated fatty acids (two to 18 carbon atoms), and fermented crotonate to acetate and butyrate, with a stoichiometry similar to that reported forSyntrophomonaswolfei . Two thermophilic, anaerobic members of the domain Bacteria were isolated and phylogenetically characterised as Coprothermobacter proteolyticus and Anaerobaculum thermoterrenum. Amplified ribosomal DNA restriction analysis of the members of the domain Bacteria in the enrichment culture showed that there were three dominant restriction patterns, two of which corresponded to those of the isolated non-syntrophic organisms. The sequence of the third pattern clustered with a group of bacteria that degrade C4 and higher fatty acids in syntrophic association. However, it diverged considerably from the closest thermophilic relatives, Syntrophothermus lipocalidus and Thermosyntropha lipolytica, indicating the presence of a new thermophilic long-chain fatty acid-degrading bacterium in the enrichment culture.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Sterols (e.g. cholesterol) present in wool scouring effluent represent the most recalcitrant fraction in anaerobic treatment. This study was conducted to examine the feasibility of removal of this organic load through a denitrifying post-treatment stage. A stable cholesterol-denitrifying enrichment (CHOL-1) was obtained from sludge of a bench-scale upflow sludge bed (USB) denitrifying reactor integrated to a carbon and nitrogen removal system for sanitary landfill leachate. According to the amounts of cholesterol degraded and of nitrite and nitrogen gas formed, the capacity for complete cholesterol oxidation under anaerobic conditions by CHOL-1 can be assumed. Nitrite accumulation observed at a low C/N ratio outlines the importance of determining the optimal C/N ratio for adequate denitrifying reactor performance. The enrichment was partly identified with molecular analysis of cloned 16S rDNA sequences revealing the presence of two groups of bacteria belonging to the b subclass of the Proteobacteria. According to analysis of sequences, it can be inferred that a yet uncultivated new bacterium is the one responsible for cholesterol oxidation. Results of this study suggest that sludge from a denitrifying reactor treating leachate is potentially useful in a combined anaerobic-anoxic system for degradation of cholesterol that remains after methanogenic treatment.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[In this study, the microbial community characteristics in continuous lab-scale anaerobic reactors were correlated to reactor functionality using the microbial resource management (MRM) approach. Two molecular techniques, denaturing gradient gel electrophoresis (DGGE) and terminal-restriction fragment length polymorphism (TRFLP), were applied to analyze the bacterial and archaeal communities, and the results obtained have been compared. Clustering analyses showed a similar discrimination of samples with DGGE and T-RFLP data, with a clear separation between the meso- and thermophilic communities. Both techniques indicate that bacterial and mesophilic communities were richer and more even than archaeal and thermophilic communities, respectively. Remarkably, the community composition was highly dynamic for both Bacteria and Archaea, with a rate of change between 30% and 75% per 18 days, also in stable performing periods. A hypothesis to explain the latter in the context of the converging metabolism in anaerobic processes is proposed. Finally, a more even and diverse bacterial community was found to be statistically representative for a wellfunctioning reactor as evidenced by a low Ripley index and high biogas production.]]></dcterms:abstract>
