                <text>Introduction Quillaja brasiliensis (St. A. ‐Hil. &amp;amp; Tul) Mart (Quillajaceae) is a species native to South America, which is rich in saponins. Saponins are used in different industries, so there is a constant demand for this type of compound. Based on the wide range of applications for the saponins found in this species, notably as immunoadjuvants, we conducted a comprehensive study of this tree and its saponins. Objective The purpose of this work is to complete the characterisation of the immunoadjuvant saponin fraction from Q. brasiliensis leaves and further study the saponin fraction obtained from Q. brasiliensis bark. Methodology Saponin fractions were studied using mass spectrometry in combination with classical methods of monosaccharide and methylation analysis. We performed direct infusion and liquid chromatography/electrospray ionisation ion trap multiple‐stage mass spectrometry (DI‐ESI‐IT‐MSn and LC‐ESI‐IT‐MS2). Results Seventy‐five saponins, 21 from leaves and 54 from bark, were tentatively identified according to their molecular mass, fragmentation pattern and chromatographic behaviour. This work represents the first investigation of saponins from the bark of Q. brasiliensis and some of them presented new structural motifs not previously reported in the genus Quillaja. Conclusion The efficiency and selectivity of the data dependent LC‐MS2 method allowed the rapid profiling of saponins from Q. brasiliensis. The results of the monosaccharide and methylation analysis performed in saponins from Q. brasiliensis fractions and Q. saponaria Molina (Quillajaceae) fraction gives further support to the structures proposed according to the mass spectral data, validating the strategy used in the present work.</text>

                <text>With the aim of monitoring multiclass semi-polar pesticide residues in freshwater fatty fish, two QuEChERS approaches (so-called acetate buffered and unbuffered versions) were evaluated for the determination of 77 pesticide residues. Compounds were selected according to the dominant rainfed agriculture activities in South America. Unbuffered QuEChERS was finally chosen for validation purposes owing that it provided the best results in terms of recovery yields. Method performance was evaluated in two instrumental systems, liquid chromatography &amp;ndash; tandem mass spectrometry (LC&amp;ndash;MS/MS) in Scheduled MRM&amp;trade; algorithm available on hybrid quadrupole &amp;ndash; linear ion trap (QLIT) instrument, and gas chromatography &amp;ndash; mass spectrometry (GC&amp;ndash;MS) under selected ion monitoring (SIM) mode. Spiking experiments were carried out to determine the trueness, precision, linearity, limit of quantification of the method as well as matrix effect. The Unbuffered QuECHERS method described here: &amp;bull; Was validated for the analysis of 67 pesticide residues in fish muscle tissue. &amp;bull; Presented quantification limits in the range 1&amp;ndash;15&amp;thinsp;&amp;mu;g&amp;thinsp;kg&amp;minus;1 for the vast majority of the studied compounds. &amp;bull; Enable environmental monitoring of pesticide residues in fish due to their low LOQs.</text>

                <text>This paper describes a new, improved systematic qualitative analysis of yucca saponins in commercial products by combined use of high-pressure liquid chromatography (HPLC) coupled to an evaporative light scattering detector (ELSD) and matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS). Three groups with four saponins in each were completely resolved by HPLC-ELSD under C18 reversed-phase conditions using a linear gradient composed of methanol and water. The selectivity of the described HPLC-ELSD method is given by the sapogenin differences at C12 (carbonyl group) and at C2 (hydroxyl group), followed by the presence or absence of an exomethylene group at C25, and the composition and length of the oligosaccharidic chain at C3 in the sapogenins. The saponins were identified by chemical and spectroscopic methods including MALDI-TOF MS1 and MS2, high resolution MS, and one and bi-dimensional nuclear magnetic resonance (NMR) experiments. Among the twelve saponins identified, eleven were previously reported, and one is reported for the first time in Yucca schidigera. An identification flowchart procedure based on the analysis of the sodium adducts of intact saponins [M+Na]+ and the oligosaccharidic chain ions obtained in the MALDI-TOF MS1 and the MS2 spectrums, was developed for the analysis of Y. schidigera saponins. Two intact isomeric saponins were differentiated by this method, which could also be applied to the structural assignment of other steroid and triterpenic saponins. Using the four-major saponins as reference standards, a C18 reversed-phase HPLC-ELSD method was validated for their specific analysis in commercial extracts of Y. schidigera. Finally, the applicability of the HPLC-ELSD method for the relative quantification of yucca saponins is discussed.</text>

                <text>Mycobacterium tuberculosis, agente etiol&amp;oacute;gico de la tuberculosis, representa un importante problema de salud p&amp;uacute;blica a nivel global. El genoma de M. tuberculosis codifica 11 Ser/Thr quinasas &amp;ldquo;de tipo eucariota&amp;rdquo; denominadas Pkns (PknA a PknL), ocho de las cuales se encontraron expresadas durante la infecci&amp;oacute;n, seg&amp;uacute;n datos que surgen de an&amp;aacute;lisis prote&amp;oacute;micos y transcript&amp;oacute;micos. En particular una de ellas, PknG, cumple un rol tanto en el metabolismo de las micobacterias como en la patog&amp;eacute;nesis, y ha cobrado gran relevancia como una de las mol&amp;eacute;culas centrales para la sobrevida de la bacteria dentro del hospedero. Desde el punto de vista estructural esta es una quinasa singular, ya que adem&amp;aacute;s del dominio catal&amp;iacute;tico posee otros dominios de funci&amp;oacute;n a&amp;uacute;n poco caracterizada (rubredoxina en el N-terminal y un motivo repetido tetratricop&amp;eacute;ptido en la regi&amp;oacute;n C-terminal). Si bien la importancia de esta quinasa ha sido reconocida hace ya m&amp;aacute;s de una d&amp;eacute;cada, al d&amp;iacute;a de hoy se desconocen los mecanismos moleculares subyacentes a su acci&amp;oacute;n, los sustratos fosforilables tanto en la bacteria como en el hospedero, los mecanismos de activaci&amp;oacute;n/inhibici&amp;oacute;n de la misma y las se&amp;ntilde;ales que los desencadenan. En el presente trabajo nos propusimos contribuir a la elucidaci&amp;oacute;n del rol fisiol&amp;oacute;gico de PknG y su regulaci&amp;oacute;n en las micobacterias. Para ello caracterizamos el papel de los distintos dominios de PknG sobre su actividad enzim&amp;aacute;tica, e identificamos nuevos mediadores y sustratos en las v&amp;iacute;as de se&amp;ntilde;alizaci&amp;oacute;n mediadas por esta quinasa. Adem&amp;aacute;s reportamos un nuevo inhibidor de PknG que act&amp;uacute;a sobre un dominio extracatal&amp;iacute;tico de la quinasa y que representa un nuevo mecanismo para inhibir este importante factor de virulencia. Nuestros resultados indican que el dominio rubredoxina cumple un papel en la regulaci&amp;oacute;n de la actividad de esta quinasa. Estudios interact&amp;oacute;micos nos permiten adem&amp;aacute;s postular la participaci&amp;oacute;n de PknG en la regulaci&amp;oacute;n del metabolismo bacteriano y la s&amp;iacute;ntesis de la pared celular. En particular, esta enzima estar&amp;iacute;a encargada de la fina regulaci&amp;oacute;n tendiente a mantener el balance entre la s&amp;iacute;ntesis y la degradaci&amp;oacute;n de intermediarios nitrogenados en la bacteria.</text>
