<text>Benzenoid-derived metabolites act as precursors for a wide variety of products involved in essential metabolic roles in eukaryotic cells. They are synthesised in plants and some fungi through the phenylalanine ammonia lyase (PAL) or tyrosine ammonia lyase (TAL) pathways. Ascomycete yeasts and animals both lack the capacity for PAL/TAL pathways, and metabolic reactions leading to benzenoid synthesis in these organisms have remained incompletely known for decades. Here we show genomic, transcriptomic and metabolomic evidence that yeasts use a mandelate pathway to synthesise benzenoids, with some similarities to pathways used by bacteria. We conducted feeding experiments using a synthetic fermentation medium that contained either 13C-phenylalanine or 13C-tyrosine and, using methylbenzoylphosphonate (MBP) to inhibit benzoylformate decarboxylase, we were able to accumulate intracellular intermediates in the yeast Hanseniaspora vineae. To further confirm this pathway, we tested three mutants in separate fermentation experiments with deletions in the key genes putatively proposed to form benzenoids (Saccharomyces cerevisiae aro10Δ, dld1Δ and dld2Δ). Our results elucidate the mechanism of benzenoid synthesis in yeast through phenylpyruvate linked with the mandelate pathway to produce benzyl alcohol and 4- hydroxybenzaldehyde from the aromatic amino acids, phenylalanine and tyrosine, as well as sugars. These results provide an explanation for the origin of the benzoquinone ring, 4-hydroxybenzoate, and suggest Aro10p has benzoylformate and 4-hydroxybenzoylformate decarboxylase functions in yeast. Keywords: phenylpyruvate pathway, yeast, benzyl alcohol, Hanseniaspora vineae, 4- hydroxybenzoic acid, benzoylformate decarboxylase, ARO10, coenzyme Q.</text>

<text>We investigated the hosting of planar and curved π systems by ExCage6+. The M06‐2X/6‐311G* method and including basis set superposition error (BSSE) corrections showed good agreement with the experimental free energy changes in solution. The mean absolute deviation (MAD) with respect to experiment was 1.3 kcal/mol. The M06‐2X/6‐31G* method exhibited a MAD of 2.1 kcal/mol, so it may be useful to investigate large systems. The good agreement between the M06‐2X/6‐311G*+BSSE results and the M06‐2X/6‐31G* ones was due to a fortuitous error cancelation between basis set incompleteness and BSSE. The interaction energies decreased linearly with the number of CC double bonds present in the PAH, but the shape of the PAH is an important factor in determining the binding strength. Finally, we studied how effective is ExCage6+ in reducing the inversion barriers of buckybowls. For corannulene, sumanene and dibenzo[a,g]corannulene they are reduced by 2.0, 2.7, and 2.0 kcal/mol, respectively. Although these values indicate an induced fit catalysis by ExCage6+, they are far from those values calculated inside bilayer graphene. Therefore, much work is necessary to replicate the reduction of inversion barriers observed for graphene.</text>

These dienediols were obtained from the biotransformation of monosubstituted arenes using bacterial dioxygenases (toluene and benzoate dioxygenases). Ethyl glyoxylate and its N-tosyl imine were used as dienophiles to afford the corresponding hetero Diels–Alder bicyclic adducts with excellent regio- and stereoselectivities. Quantum chemical calculations at the B3LYP/6-31+G(d,p) level of theory were performed to rationalize the observed selectivities especially the stereochemical aspects of the cycloadditions. The synthetic importance of these adducts is highlighted for the preparation of enantiopure 2,2,3,4,5,6-hexasubstituted piperidine and tetrahydropyran from toluene</text>
