Abstract
A summary of the resource.
The high protein content of Arthrospira platensis makes this cyanobacterium a great component of food. Thus, the protein obtained should guarantee the nutrient characteristic, in this case, as bioactivity and techno-functional properties. Membrane separation processes (MSPs) have been highlighted by their capacity to separate proteins and keep their properties. Therefore, MSP was used to concentrate and separate the proteins fromA. platensis, which were characterized by their physicochemical and techno-functional properties. Different ultrafiltration (UF) membranes and operational conditions (10- and 50-kDa pore size, conventional and tangential flow, hollow fiber type and pressures of 1.0, 1.5, and 2.0 bar) were tested and associated with diafiltration (DF). The maximized condition of UF/DF (hollow fiber, 50 kDa, tangential flow at 1.5 bar, associated with one DF) was up scaled (pilot scale) and allowed concentrate and permeate fractions with 81.75% and 61.20% of protein, respectively, to be obtained. A protein concentrate with 142 mg L−1 of phycocyanin, 90% of antioxidant activity, with proteins more soluble at high pH (50% soluble protein at pH 7.0) and molecular weight among 5–15 and 100 kDa, was obtained. Protein thermal and emulsifying properties were maintained by the MSP, and the concentrate fraction presented results similar as A. platensis biomass, which confirms the advantage of this method in guaranteeing the protein quality.

Efficiency of in-situ alkaline transesterification of dry biomass from a crop of Spirulina sp., using ultrasound-assisted and/or the addition of different co-solvents to the reaction medium was studied. The starting biomass had a total lipid content of 7.0% (Folch method), comprised mainly by the following fatty acids: 16:0 (44.2%), 18:3 (n–6) (23.3%), and 18: 2 (n–6) (11.1%). When the transesterification reaction was carried out without ultrasound, it only extracted 12.8% of lipids (0.9% of the biomass) after 2 h of reaction, achieving only a 63.6% ester content. When the process was carried out under the same conditions except for the ultrasound-assisted (80 W), the weight yield remained mostly unchanged, but the ester content increased to 97.6%. Additionally, when chloroform was added to methanol as a co-solvent in a 2:1 ratio it was possible to extract 43% of lipids. However, the ester content was reduced to 69.7%. Moreover, with maximum ultrasound power (180 W) a high ester content (96.9%) and a relatively high extraction yield (26%) were obtained, even without the co-solvent. These results show that the ultrasound significant increased the yield of the in-situ alkaline transesterification of Spirulina sp. biomass, promoting a higher percent recovery of lipids as well as ester content from the starting material.

En una primera etapa de la presente tesis se estudi6 el contenido en acidos grasos de diferentes cepas y especies de Spirulina con el fin de tener un perfil de composici6n. Luego se estudiaron diferentes condiciones de cultivo de una misma cepa de Spirulina platensis alterada lisa . Las variaciones en lascondiciones de cultivo estudiadas con el fin de obtener una sobreproducci6n de acido gama linolenico, fueron de indole fisico (fotoperiodo, temperatura y forma de colecta) y de tipo quimico ( diferentes fuentes de mtr6geno, agregado de vitaminas,etc) . Los cultivos se realizaron en un reactor cilindrico y en un reactor "air lift" con el fin de comparar la productividad del acido de interes. EI control de los cultivos se realiz6 mediante parametros de control estandares (clorofila a, peso seco, absorbancia a 540, etc.) y parametros de control electroquimicos (02 disuelto, pH, conductividad y potencial redox). En relaci6n a la colecta, la selecci6n de microalgas de gran tamafio de filamento y el muestreo en la fase terminal favorece en la producci6n de acido gama linolemco. Se encontr6 que condiciones de cultivo en las que se reduce la temperatura y se utilizan fotoperiodos con mayor tiempo de oscuridad mejoran la producci6n de acido gama linolemco. Por otro lado en cuanto a la producci6n de biomasa estas variaciones tienen un efecto nocivo puesto que la reducen .. La modificaci6n quimica que actu6 en favor del aumento del contemdode acido gama linolenico fue el el agregado de un complemento vitamimco .(Dayamineral). En cuanto a los reactores empleados se vio que la producci6n en un reactor "Air lift" es ampliamente superiorque uno cilindrico. En cuanto al control de los cultivos se encontro que la medida de los panimetros electroquimicos es facil de automatizar y da una amplia informacion sobre el estado de los cultivos.
