This research explores the hydrochar production from grape pomace via hydrothermal carbonization and the determination of the kinetic parameters of its combustion process. The hydrochar was obtained by hydrothermal carbonization at 220 ◦C for 240 min with self-generated pressure. The process indicated a mass yield of 65.21 %, and the hydrochar exhibited an HHV of 26.60 MJ/kg, showing its potential as an efficient biofuel. Thermokinetic analysis of hydrochar combustion was conducted using multiple complementary methods: isoconversional analysis, mathematical deconvolution, combined kinetic analysis of each step, and optimization of parameters through nonlinear regression. The findings indicate that the overall process starts at ~152 ◦C and involves three apparent overlapping steps. Grape pomaces have a wide and diverse antioxidant phenolics composition. Six Uruguayan red grape pomaces were evaluated in their phenolics composition, antioxidant capacity, and anti inflammatory properties. Not only radical scavenging methods as DPPH· and ABTS·+ were employed but also ORAC and FRAP analyses were applied to assess the antioxidant potency of the extracts. The antioxidant reactivity of all extracts against hydroxyl radicals was assessed with ESR. The phenol profile of the most bioactive extract was analyzed by HPLC-MS, and a set of 57 structures were determined. To investigate the potential anti-inflammatory activity of the extracts, Nuclear Factor kappa-B (NF-κB) modulation was evaluated in the human colon cancer reporter cell line (HT-29-NF-κB-hrGFP). Our results suggest that Tannat grapes pomaces have higher phenolic content and antioxidant capacity compared to Cabernet Franc. These extracts inhibited TNF-alpha mediated NF-κB activation and IL-8 production when added to reporter cells. A molecular docking study was carried out to rationalize the experimental results allowing us to propose the proactive interaction between the NF-κB, the grape extracts phenols, and their putative anti-inflammatory bioactivity. The present findings show that red grape pomace constitutes a sustainable source of phenolic compounds, which may be valuable for pharmaceutical, cosmetic, and food industry applications.

In the present work the feasibility of Tannat grape skin (TGS) as a functional ingredient in the formulation of two snacks (yogurt and biscuits) was studied. The research provided novel information on the effects of the food matrix and digestion process, under simulated human oral gastrointestinal conditions, in the bioaccessibility of TGS bioactive compounds composing of the snacks with health promoting properties (antioxidant, anti-inflammatory, and antidiabetic). TGS polyphenolic profile was analyzed by ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) finding mainly flavonoids, phenolic acids, and anthocyanins, which may exert antioxidant, anti-inflammatory, and carbohydrase inhibition capacities. TGS digest showed antioxidant and antidiabetic potential compared to the undigested sample (p < 0.05). Yogurt and biscuits with TGS were developed with the nutrition claims “no-added sugars” and “source of fiber” and were digested in vitro to evaluate the bioaccessibility of compounds with health promoting properties after food processing and digestion. After in vitro simulation of digestion, bioactive properties were enhanced for control and TGS snacks which may be attributed to the formation/release of compounds with health-promoting properties. Biscuits showed significant increase in ABTS antioxidant capacity and yogurt showed increased �-glucosidase inhibition capacity by the addition of TGS (p < 0.05). Polyphenols from TGS and bioactive peptides from snacks which may be released during digestion might be responsible for the observed bioactivities. Consumer’s acceptance of TGS yogurt and biscuits showed scores of 6.3 and 5.1 (scale 1–9), respectively, showing TGS yogurt had higher overall acceptance. Sensory profile assessed by check-all-that-apply + just-about-right (CATA+JAR) showed most of the attributes were evaluated as “just about right”, supporting good food quality. The developed yogurt presented adequate shelf-life parameters for 28 days. TGS yogurt with higher acceptability showed reduced ROS formation (p < 0.05) induced by tert-butyl hydroperoxide (1 mM) in CCD-18Co colon cells and RAW264.7 macrophages when pre-treated with concentrations 500–1000 and 100–500 �g/mL of the digests, respectively. Moreover, TGS yogurt digest pre-treatment reduced nitric oxide (NO) production (p < 0.05) in lipopolysaccharide (LPS)-induced RAW264.7 macrophages, showing anti-inflammatory potential. Bioactive peptides generated during lactic fermentation and digestion process may be contributors to intracellular effects. In conclusion, yogurt and biscuits with Tannat grape skin addition were obtained with nutrition claims “no-added sugars” and “source of fiber” with the potential to modulate key biochemical events associated with diabetes pathogenesis.

A summary of the resource.
<p>The reduction of the environmental impact represents an important challenge<br />for the winemaking industry. Red grape by-products are composed of several<br />health-promoting components such as polyphenols and dietary fiber, which may<br />be employed as sustainable food ingredients. Although the bioactive properties<br />of red grape pomace, one of the most abundant byproducts of the winemaking<br />industry worldwide, have been extensively documented by in vitro studies,<br />there is a lack of information on the bioaccessibility, bioavailability and food<br />applications that will subsequently affect the contribution of grape byproduct<br />bioactive compounds to these properties. Therefore, the present chapter aimed<br />to present some of the latest grape byproducts in vitro bioactive properties,<br />as well as novel and updated knowledge related to its bioaccessibility,<br />bioavailability and food applications.</p>

El propóleos y el orujo de uvas son productos reconocidos por contener antioxidantes. Un extracto elaborado en base a ellos podría ser de gran aplicabilidad en las industrias alimenticia (alimentos funcionales, nutracéutico), farmacéutica (fitofármacos), y/o de aditivos preservantes de la oxidación. El trabajo de investigación presentado en esta Tesis de Doctorado fue basado en la necesidad de generar conocimiento en cuanto a las características fisicoquímicas, farmacológicas y biológicas de esos dos productos naturales uruguayos. Con el mismo se aportó nuevo conocimiento científico en la aplicación de metodología in silico al estudio de mezclas complejas de fenoles en propóleos y orujos de uvas que se complementó con las de laboratorio húmedo o in vitro. Los fenoles, moléculas antioxidantes detectadas en los propóleos y orujos de uvas, atrapan radicales libres modulando el estrés oxidativo celular. Presentan en su estructura grupos aromáticos hidroxilados e intervienen en los mecanismos redox de vegetales y animales, promoviendo un descenso de la cantidad de especies reactivas de oxigeno (ROS). En los propóleos se han identificado flavonoides (como flavonas y flavanonas), ácidos aromáticos y sus ésteres, aldehídos aromáticos, cumarinas y triglicéridos fenólicos. Los orujos de uvas contienen estilbenos, antocianinas, taninos, catequinas, flavanoles, flavonoles y flavonas. El perfil estructural es complementario de tal manera, que un “cóctel polifenólico” elaborado a partir de propóleos y orujos de uvas uruguayos, contendrá un amplio rango de estructuras fenólicas que sirvan para desarrollar un extracto patronizado en cuanto a sus capacidades como antioxidante y antiinflamatorio. Dado que es característica en los productos naturales la alta variabilidad en sus contenidos de fenoles, cualquier potencial aplicabilidad industrial implica la necesidad de analizarlos cualitativa y cuantitativamente así como su funcionalidad. Para lograrlo, se realizaron muestreos, se optimizaron los procesos extractivos, se analizaron los contenidos fenólicos totales, la capacidad antioxidante por los métodos DPPH y ABTS, se realizó la dilucidación de sus huellas dactilares de protón por resonancia magnética nuclear (RMN 1H), se estudió la reactividad de los grupos OH y la energía de disociación del enlace O-H de los polifenoles como formadores del radical libre mediante química cuántica DFT (M06-2X/6-311+G(3df,2p) y G4.
