<p>In the search for a more effective chemotherapy for the treatment of Human African Trypanosomiasis, a disease caused by the parasite Trypanosoma brucei, the development of ferrocenyl compounds has arisen as a promising strategy. In this work, five new Pd–Fe heterobimetallic [PdII(L)(dppf)](PF6) compounds, including 8-hydroxyquinolyl derivatives HL1–HL5 as bioactive ligands and dppf = 1,1’-bis(diphenylphosphino)<br />ferrocene as the organometallic co-ligand, were synthesized and fully characterized in the solid state and in solution. Molecular structures of three compounds were solved by single crystal X-ray diffraction methods. The compounds displayed submicromolar or micromolar IC50 values against bloodstream<br />T. brucei (IC50: 0.33–1.2 μM), and good selectivity towards the pathogen (SI: 4–102) with respect to mammalian macrophages (cell line J774). The new Pd complexes proved to be 2-fold to 45-fold more potent than the drug nifurtimox but most of them are less active than their Pt analogues. Potential molecular<br />targets were studied. The complexes interact with DNA but they do not alter the intracellular thiolredox homeostasis of the parasite. In order to understand and predict the main structural determinants on the anti-T. brucei activity, a search of quantitative structure–activity relationships (QSAR) was performed including all the [M(L)(dppf)](PF6) complexes, where M = Pd(II) or Pt(II), currently and previously<br />developed by us. The correlation obtained shows the relevance of the electronic effects, the lipophilicity and the type of metal. According to the QSAR study, compounds with electron-withdrawing ligands, higher lipophilicity and harboring Pt would result in higher T. brucei cytotoxicity. From the whole series of<br />[M(L)(dppf)](PF6) compounds developed, where M = Pt(II) or Pd(II) and HL = 8-hydroxyquinolyl derivatives, Pt-dppf-L4 (IC50 = 0.14 μM, SI = 48) was selected to perform an exploratory pre-clinical study in infected mice. This hit compound lacks acute toxicity when applied to animals in the dose/regimen described and exerts an anti-proliferative effect on parasites, which extends animal survival but is not curative.</p>

Cyclophilins (CyPs) are enzymes involved in protein folding. In Trypanosoma cruzi (T. cruzi), the most abundantly expressed CyP is the isoform TcCyP19. It has been shown that TcCyP19 is inhibited by the immunosuppressive drug cyclosporin A (CsA) and analogs, which also proved to have potent trypanosomicidal activity in vitro. In this work, we continue and expand a previous study on the molecular interactions of CsA, and a set of analogs modeled in complexes with TcCyP19. The modeled complexes were used to evaluate binding free energies by molecular dynamics (MD), applying the Linear Interaction Energy (LIE) method. In addition, putative binding sites were identified by molecular docking. In our analysis, the binding free energy calculations did not correlate with experimental data. The heterogeneity of the non-bonded energies and the variation in the pattern of hydrogen bonds suggest that the systems may not be suitable for the application of the LIE method. Further, the docking calculations identified two other putative binding sites with comparable scoring energies to the active site, a fact that may also explain the lack of correlation found. Kinetic experiments are needed to confirm or reject the multiple binding sites hypothesis. In the meantime, MD simulations at the alternative sites, employing other methods to compute binding free energies, might be successful at finding good correlations with the experimental data.

Current knowledge on the role of oxygen radicals in the development of chemotherapy for Chagas' disease is reviewed. This includes a) a brief description of the disease and its prevalence in the endemic area; b) applicability and toxicity of nifurtimox and benznidazole; c) the production of oxygen radicals by nitrocompounds, specially by nifurtimox; d) the action of nifurtimox and benzidazole on DNA, RNA and protein synthesis and degradation in Trypanosoma cruzi, including nuclear DNA and kinetoplast DNA cleavage and repair; e) nifurtimox reduction by lipoamide dehydrogenase; J) the role of T cruzi trypanothione system; g) the chemical and biological consequences of replacing the tetrahydrothiazine group of nifurtimox by unsaturated nitrogen heterocycles; h) the electronic properties of the new compounds, in relation to their capability for nitroanion radical production and trypanocidal action in vitro (QSAR) and in vivo; i) evidence for the possible role of oxygen radicals in the action of nifurtimox on T. cruzi and the human host. It is concluded that the chemotherapy of Chagas'disease remains an unsolved problem and, therefore, the search for new drugs must continue.

The review presents: a) a brief description of the disease; b) a summary of the most important metabolic targets so far identified in Trypanosome cruzi (T. cruzi) along with corresponding inhibitor compounds; c) the current state of knowledge on the trypanothione reductase system of trypanosomatids with reference to oxidative stress defenses; d) detailed discussions on T. cruzi trypanothione reductase inhibitors such as nitrofuranes, naphthoquinones and phenothiazines. As yet, the chemotherapy of Chagas' disease remains an unsolved problem. Further search for new drugs must continue by means of nucleating existing chemotherapy efforts.

El presente trabajo de tesis se enmarca en una línea de investigación vinculada a la enfermedad de Chagas y desarrollada por muchos años en el actual Centro de Bioinformática Estructural del DETEMA, de la Facultad de Química, Universidad de la República. Esta línea, iniciada con motivo de la tesis doctoral de la patrocinante, la Dra. Paulino, ha dado múltiples trabajos publicados, entre ellos “Modeling and study of cyclosporyn A and related compounds in complexes with Trypanosoma cruzi cyclophilin”, como producto de la primer parte del trabajo presentado en esta tesis. También la línea de investigación ha originado otras tesis de Maestría y Doctorado, entre ellas la de uno de los copatrocinantes, el Dr. Iribarne. Por otro lado no es ajena la mencionada línea al Laboratorio de Inmunotecnología dirigido por el otro copatrocinante, MsC. Battistoni. Mucho se ha investigado con el objetivo de obtener una cura para el Mal de Chagas. Esta Tesis es un aporte más, centrándose en el estudio de la molécula de ciclosporina (CsA), y compuestos estructuralmente relacionados. Estos compuestos han mostrado actividad inhibitoria sobre Trypanosoma cruzi. Si bien la CsA es una droga con actividad inmunosupresora, algunos de los compuestos estructuralmente relacionados activos contra T. cruzi no presentan efectos inmunosupresores. Se ha propuesto a CsA como compuesto líder para el desarrollo de nuevas drogas útiles en la farmacoterapias de la enfermedad de Chagas. Se conocen con cierto grado de detalle las bases bioquímicas de las actividades inmunosupresoras e inhibitorias sobre protozoarios de CsA. En ambas están involucradas una familia de proteínas, las ciclofilinas. Se han publicado datos de las actividades inhibitorias de CsA y compuestos relacionados sobre ciclofilinas presentes en T. cruzi, en particular sobre la ciclofilina mas expresada en T. cruzi, la TcCyP19. Las bases moleculares de la diferencia de actividad inmunosupresora y de inhibición sobre T. cruzi de CsA y de una serie análogos no han sido dilucidadas. Esta tesis pretende, mediante el uso de diversas herramientas de modelado molecular, docking y de mecánica molecular, estudiar el comportamiento de los complejos binarios de TcCyP19 con una serie de compuestos análogos a CsA para obtener información a nivel molecular que aporte a la comprensión de las diferencias encontradas. En forma complementaria, se estudian complejos de CsA y dos de sus análogos con una ciclofilina humana, así como el comportamiento de una proteína mutante de la ciclofilina humana con baja afinidad por CsA. Una vez elaborados los modelos de los sistemas a estudiar se analizaron los posibles sitios de unión de estos compuestos con las ciclofilina en estudio, así como los modos de unión a los mismos. Utilizando simulaciones de dinámica molecular, se obtuvo un muestreo del espacio de fases de los sistemas en estudio que permitió analizar estructuralmente y energéticamente dichos sistemas. Varios términos energéticos se calcularon, que incluyeron energías de interacción ligando-proteína y cálculos de energías libres de asociación por medio del método LIE (Linear Interacction Energy). Se discuten algunos aspectos metodológicos, como la necesidad de hacer varias simulaciones de dinámica molecular para obtener un muestreo mas apropiado del comportamiento del sistema y el cumplimiento de las premisas para usar el método LIE. Los resultados obtenidos muestran que se trata de sistemas con comportamiento muy similar, para los cuales los métodos de cálculo usados no son lo suficientemente sensibles para diferenciar energías libres de unión. Se demostró la posible existencia de lugares de unión de Modelado y estudio de complejos de Ciclosporina A y compuestos relacionados ….. 5 los compuestos estudiados diferentes al sitio catalítico, cuya relevancia deberá ser estudiada in vitro. Todo lo aprendido con estos sistemas permitirá abordar con mejores perspectivas ulteriores etapas del estudio de esta familia de compuestos que involucran complejos ternarios y sus potenciales actividades inmunosupresoras en el huésped o inhibitorias del crecimiento de T. cruzi.
