    <dcterms:abstract><![CDATA[BACKGROUND: Piezodorus guildinii (Westwood, Hemiptera: Pentatomidae) causes economic losses in soybean crops by diminishing productivity, impacting grain size, oil content, and germination power. RNA interference (RNAi), a conserved gene regulation process initiated by double-stranded RNA (dsRNA), may be used as a safe technique in integrated pest management strategies. Conjugating dsRNA with cationic polymers, which protect dsRNA from degradation has shown to enhance this response. This study aims to assess the impact of dsRNA administration on the viability of P. guildinii and evaluate dsRNAchitosan nanocomplexes as a pest control strategy. RESULTS: Ten target genes were selected, dsRNAs were designed and synthesized. In injection assays, significant mortality exceeding 76% was observed, while mortality from ingestion also showed significant differences, reaching 49%. The dsRNA targeting Srp54k gene was selected and a chitosan-dsRNA nanocomplex was synthesized. These nanocomplexes demonstrated enhanced stability under RNase treatment in vitro, and, in in vivo assays, a cumulative corrected mortality rate of 66% was reported. Consistent with these findings, RT-qPCR analysis revealed that gene expression levels in treated insects were significantly reduced compared to control at 24, 48, and 72 h post treatment for targeted genes. CONCLUSION: In this work, we evaluated the effect of dsRNA administration by injection and ingestion on the viability of P. guildinii and determined its molecular silencing effects. Additionally, we synthesized and characterized chitosan-dsRNA nanoparticles, which demonstrated improved efficacy in terms of stability. These findings are promising for the development of new integrated management strategies for stink bugs control.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The immunoregulatory cation channel TMEM176B plays a dual role in tumor immunity. On the one hand, TMEM176B promotes antigen cross-presentation to CD8+ T cells by regulating phagosomal pH in dendritic cells (DCs). On the other hand, it inhibits NLRP3 inflammasome activation through ionic mechanisms in DCs, monocytes and macrophages. We speculated that formulating BayK8644 in PEGylated chitosan nanoparticles (NP-PEG-BayK8644) should slowly release the compound and by that mean avoid cross-presentation inhibition (which happens with a fast 30 min kinetics) while still triggering inflammasome activation. Chitosan nanocarriers were successfully obtained, exhibiting a particle size within the range of 200 nm; they had a high positive surface charge and a 99 % encapsulation efficiency. In in vitro studies, NP-PEG-BayK8644 did not inhibit antigen cross-presentation by DCs, unlike the free compound. The NP-PEG-BayK8644 activated the inflammasome in a Tmem176b-dependent manner in DCs. We administered either empty (eNP-PEG) or NP-PEGBayK8644 to mice with established tumors. NP-PEG-BayK8644 significantly controlled tumor growth and improved mice survival compared to both eNP-PEG and free BayK8644 in melanoma and lymphoma models. This effect was associated with enhanced inflammasome activation by DCs in the tumor-draining lymph node and infiltration of the tumor by CD8+ T cells. Thus, encapsulation of BayK8644 in chitosan NPs improves the anti-tumoral properties of the compound by avoiding inhibition of antigen cross-presentation.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[In the agricultural field, chitosan (CS) is of great interest due to their broad antimicrobial activity and their ability to induce resistance mechanisms in plants against phytopathogens. Likewise, the potential of silver nanoparticles in preventing microbial infections has been demonstrated, including their broad antimicrobial activity against phytopathogens. In this study, the synthesis and characterization of chitosan-silver nanohybrids were performed. Firstly, mycosynthesis of biogenic silver nanoparticles (PchNPs) was carried out. Subsequently, the nanohybrid (CS-PchNHs) synthesis involved the addition of PchNPs, with continuous stirring, into a CS solution. For the first-time stable CS-Pch NHs were produced using biogenic silver nanoparticles from the fungus Phanerochaete chrysosporium, characterized by UV-visible spectroscopy (UV-Vis), Dynamic Light Scattering, (DLS), &zeta;-Potential, and X- ray diffraction (XRD). Additionally, CS-PchNHs exhibited high antifungal activity at low concentrations against four phytopathogens, suggesting that these nanosystems may be used as an alternative to the current hazardous antifungal agents in the integrated disease management.]]></dcterms:abstract>

    <dcterms:abstract><![CDATA[The objective of this work was to investigate the effect of the addition of carboxymethyl chitosan on the structural properties and antilisterial activity of nisin-incorporated chitosan films. Chitosan and carboxymethyl chitosan solutions were prepared with different mass ratios and bacteriocin nisin was added (0, 1000 and 6000 IU/ml). Filmogenic solutions were cast, dried and their physico-chemical and antimicrobial properties were investigated. For the same chitosan/carboxymethyl chitosan mass ratio, the addition of NIS at 6000 IU/ml led to changes in the macro- and microstructure, as well as in physico-chemical properties of films. On the other hand, carboxymethyl chitosan had a plasticizing effect and enhanced the distribution of the bacteriocin within the biopolymer matrix. Moreover, nisin-incorporated blend films of chitosan and carboxymethyl chitosan were more effective against Listeria monocytogenes than their pure chitosan counterparts. This study showed that different formulations of nisin-incorporated composite films of chitosan and carboxymethyl chitosan may provide options for developing bioactive packaging to improve food safety.]]></dcterms:abstract>
