Abstract
A summary of the resource.
Several novel semicarbazones derivatives were prepared from 5-nitro-2-furaldehyde or 5-nitrothiophene-2-carboxaldehyde, and tested in vitro as potential anti-trypanosomal agents. The compounds were prepared in good to excellent yields in 2–3 steps from readily available starting materials. Some derivatives were found to be active against Trypanosoma cruzi with an activity similar to that of Nifurtimox.

Abstract
A summary of the resource.
The synthesis and characterization of the first two Re complexes with semicarbazone ligands is presented. Selected ligands are 5-Nitro-2-furaldehyde semicarbazone (Nitrofurazone) (L1) and its derivative 3-(5-Nitrofuryl)acroleine semicarbazone (L2). Complexes of general formula [ReVOCl2(PPh3)L], where L � L1 and L2, were prepared in good yields and high purity by reaction of [ReVOCl3(PPh3)2] with L in ethanol or methanol solutions. The complexes formula and molecular structures were supported by elemental analyses and electronic, FTIR, 1H, 13C and 31P NMR spectroscopies. In addition, the crystal and molecular structure of [ReVOCl2(PPh3)L2] was determined by X-ray diffraction methods. [ReOCl2(PPh3)(3-(5-Nitrofuryl)acroleine semicarbazone)] crystallizes in the space group P-1 with a � 11.2334(2), b � 11.3040(2), c � 12.5040(2) A ° , α � 81.861(1), β � 63.555(1), γ � 83.626(1)°, and Z � 2. The Re(V) ion is in a distorted octahedral environment, equatorially coordinated to a deprotonated semicarbazone molecule acting as a bidentate ligand through its carbonylic oxygen and azomethynic nitrogen atoms, to an oxo ligand and a chlorine atom. The six-fold coordination is completed by another chlorine atom and a triphenylphosphine ligand at the axial positions.

ortho-Hydroxybenzaldehyde semicarbazone (salicylaldehyde semicarbazone) undergoes chemo-selective hydrolysis of the iminic carbon nitrogen double bond through its reaction with [RuCl2(dmso)4] in ethanol in the presence of water, yielding free salicylaldehyde and semicarbazide that remains coordinated to the ruthenium ion as a bidentate N,O-donor to afford [RuCl2(dmso)2(semicarbazide)] Æ 2H2O complex. The ruthenium–semicarbazide complex has been characterized by 1H NMR and FTIR spectroscopies and X-ray diffraction methods. Related semicarbazones, derived from p-hydroxybenzaldehyde and benzaldehyde, were not hydrolyzed under the same conditions, suggesting a significant role of the structural o-hydroxy motive in the reaction. Theoretical studies were performed in order to gain further insight on the mechanism of reaction. Results support the hypothesis that the ortho-hydroxy moiety, in the keto tautomeric form, participates in the chemo-selective hydrolysis promoted by [RuCl2(dmso)4].

Expanding our previous work on salicylaldehyde semicarbazone metal complexes as prospective anti-trypanosomal agents, five new fac-ReI(CO)3-containing complexes with ligands of this semicarbazone series were synthesized and characterized. An atypical coordination mode of these potentially tridentate ligands through only the carbonylic oxygen and the azomethine nitrogen (the so-called N,O fashion) was demonstrated by IR spectroscopy and supported by theoretical calculations. Three of the compounds showed moderate in vitro anti-Trypanosoma cruzi activity and increased activity with respect to the corresponding free ligands. The brominated ligands, 5-bromo-2-hydroxybenzaldehyde semicarbazone (L2) and 5-bromo-2-hydroxy-3-methoxybenzaldehyde semicarbazone (L5), led to the most active rhenium(I) complexes. These compounds are among the few reported examples of rhenium complexes bearing in vitro activity against T. cruzi.

A summary of the resource.
Metal complexes that establish interactions with DNA are being studied not only because of their potential use as therapeutic agents but also as tools for biochemistry and molecular biology. Searching for drugs with anti-trypanosome activity, we previously synthesized a series of ruthenium mixed ligand dimethyl sulfoxide complexes of the type [RuIICl2(DMSO)2L], where L is 5-nitrofurylsemicarbazone derivatives and DMSO is dimethyl sulfoxide. Though they present the ability to bind DNA, no activity against parasites in cell culture was observed. Considering their potential application as molecular tools we further analyzed the interactions with DNA through an electrophoretic approach. Non covalent withdrawal of superhelicity and a rapid nicking activity upon covalent interaction was observed. Inhibition of both effects was observed in the presence of distamycin suggesting the involvement of the DNA minor groove in the interaction with the nitrofurylsemicarbazone ruthenium complexes. In addition cleavage inhibition by dimethyl sulfoxide suggests an oxidative mechanism of action.

Vanadium compounds display important pharmacological actions in vivo and in vitro systems. Semicarbazones are versatile ligands with therapeutic effects. Herein, we report the effects of VVO2(salicylaldehydesemicarbazone) (V(V)-Salsem) on two osteoblast cell lines in culture (MC3T3-E1 and UMR106). V(V)-Salsem inhibited cell proliferation in a dose response manner. At 100 lM, the complex caused an inhibition of ca. 48% and 38% for the normal and the tumoral osteoblasts, respectively (p < 0.001). This inhibition could be partially reversed to 35% and 28% by NAC (N-acetylcysteine) and a mixture of vitamins E and C. Changes in cell proliferation correlated with morphological alterations and the disruption of actin cytoskeleton fibers. The complex also enhanced the level of ROS (reactive oxygen species) up to ca. 100% over basal in both cell lines. Activation of ERK signalling cascade was also observed. These events led to apoptosis (up to 44% in MC3T3-E1 and 33% in UMR106 cells). Scavengers of ROS and inhibitors of ERK cascade allowed to elucidate the mechanisms involved in the cytotoxicity. In conclusion, V(V)-Salsem displayed cytotoxic effects on osteoblasts in culture through the production of free radicals and the activation of ERK cascade. In the search for gallium bioactive compounds five Ga(III) complexes, [GaIII(L–H)2](NO3), with tridentate salicylaldehyde semicarbazone derivatives as ligands (L) have been synthesized and characterized in the solid state and in solution by different techniques. The crystal structure of [GaIII(L4–H)2](NO3)�2H2O, where L4 is 3-ethoxysalicylaldehyde semicarbazone, was solved by X-ray diffraction methods. The gallium( III) ion is in a distorted octahedral environment, coordinated to two nearly planar and mutually perpendicular 3-ethoxysalicylaldehyde semicarbazonato anions acting as tridentate ligands through their phenol and carbonyl oxygen atoms and their azomethine nitrogen atom. Their biological potential has been explored by evaluating their activity on Mycobacterium tuberculosis, causative agent of tuberculosis, and their cytotoxicity on tumor cell lines. Three different human tumor cell lines were selected that show different degrees of resistance to metallodrugs: ovarian A2780 (low resistance), breast MCF7 (medium resistance) and prostate PC3 (high resistance) cells. Although the complexes have not shown activity on M. tuberculosis, complexation with gallium has led to the enhancement of the cytotoxic potencies of the organic compounds. Those complexes that contain a bromide substituent at the phenolate ring have shown the highest cytotoxicities. In particular, [GaIII(L2–H)2](NO3), where L2 is 5-bromosalicylaldehyde semicarbazone,�has shown a remarkable cytotoxicity on A2780 tumor cell line with an IC50 value of the same order than cisplatin (IC50 Ga–L2 = 2.4 ± 0.3 lM; IC50 cisplatin = 2.0 ± 0.1 lM, 72 h incubation at 37 �C). Interestingly, this complex has also shown moderate cytotoxicity against MCF7 and PC3 cells (IC50 MCF7 = 30 ± 6; IC50 PC3 = 18 ± 3 lM). Therefore, this gallium compound could be considered a promising wide spectrum potential anti-tumor agent.
