<dcterms:abstract><![CDATA[Rice is an important source of methane (CH4) and other crops may be sources of nitrous oxide (N2O), both of which are powerful greenhouse gases. In Uruguay, irrigated rice rotates with perennial pastures and allows high productivity and low environmental impact. A long-term experiment with contrasting rice rotation intensification alternatives, including rice–soybean and continuous rice, was recently carried out in an Argialboll located in a temperate region of South America. To know if rotation systems influence soil microbial activity involved in CH4 and N2O emissions, the abundance and potential rate for gas production or consumption of microbial populations were measured during the rice crop season. CH4 was only emitted when rice was flooded and N2O emission was not detected. All rotational soils showed the highest rate for methanogenesis at tillering (30 days after rice emergence), while for methanotrophy, the maximum rate was reached at flowering. The abundance of related genes also followed a seasonal pattern with highest densities of mcrA genes being observed at rice flowering whereas pmoA genes were more abundant in dry soils after rice harvest, regardless of the rotation system. Differences were foundmainly at tillering when soils with two consecutive summers under rice showed higher amounts ofmcrA and pmoA gene copies. The potential denitrification rate was highest at the tillering stage, but the abundance of nirK and nirS genes was highest in winter. Regarding ammoniumoxidation, bacterial amoA abundance was higher in winter while the archaeal amoA gene was similar throughout the year. A strong influence of the rice growth stage was registered for most of the parameters measured in rice paddy soils in this no-till rice intensification experiment. However, differences among rotations begin to be observed mainly at tillering when the abundance of populations of the methane and nitrous oxide cycles seemed to respond to the rice intensification.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Input of plant material may strongly change decomposition rates of soil organic matter (SOM), i.e. causing priming effect (PE), but the underlying mechanisms are largely unknown. We found that rice straw addition in anoxic Fuyang (F) rice field soil stimulated CH4 production from SOM at the expense of CO2, whereas in Uruguay (U) soil it suppressed SOM degradation to CO2 plus CH4 (negative PE). Reciprocal inoculation experiments with non-sterile and sterile soils showed that the soils always displayed the effect of rice straw characteristic for the live microbial community rather than for the soil physicochemical properties. Pyrosequencing of 16S rRNA genes showed that bacterial communities in these soil samples were separated into two clusters (F and U). Symbiobacterium was abundant or dominant in microbiota from U soil, but negligible in those from F soil. Network analysis indicated that the bacterial populations involved in SOM decomposition were different between soils of F and U clusters; moreover, they were more tightly connected to methanogens in U than in F clusters. Ultimately, our results suggested that the PE of rice straw is mediated by the composition and activity of soil microbial community.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Intermittent drainage is one of the most promising approaches to mitigate methane (CH 4 ) emission from paddy fi elds. Irrigated rice fi elds in Uruguay are temporarily established on soils used as cattle pastures. We studied soil from the pasture-rice rotation (UR) as well as soil from a permanent cattle pasture (UT) hypothesizing that activity and structure of the bacterial and archaeal communities involved in pro- duction of CH 4 change systematically with intermittent drainage. Methane production started after 7 days and 16 days of anoxic incubation in UR and UT soil, respectively. Then, production rates of CH 4 were higher in UT than UR soil. Intermittent drainage signi fi cantly decreased the rates of CH 4 production. Analysis of d 13 C indicated that CH 4 was mainly produced from acetate both in UR (73 e 98%) and UT (51 e 80%) soil. Intermittent drainage did not change the pathway of CH 4 production. Quantitative PCR showed that methanogenic archaeal gene copy numbers (16S rRNA, mcrA ) were much lower in UT than UR soil, but increased upon incubation under anoxic conditions. Terminal restriction fragment length polymorphism (T-RFLP) and pyrosequencing of bacterial and archaeal 16S rRNA genes showed that the communities were clearly different between UR and UT soil. The bacterial community consisted of 9 phyla with relative abundance of > 1% in both soils. Whereas the archaeal community in UR soil was dominated by Methanocellales and Methanosarcinaceae , that in UT soil was dominated by Crenarchaeota . Anoxic incubation affected the composition of the bacterial and archaeal communities in UT soil, but not so much in UR soil. In UT soil, the relative abundance of Clostridiales increased to 19%, and the archaeal community changed to dominance by Methanosarcinaceae and Methanobacteriales . Subsequent drainage and re- fl ooding, however, had comparatively little effect on the composition, although it decreased the rates of CH 4 production in both soils. Difference in previous soil management and in the structures of the microbial communities apparently only affected their dynamics and functioning after the fi rst fl ooding but not upon subsequent drainage and re- fl oodin]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Two simple and straightforward sample preparation methods were developed for the multiresidue analysis of post-emergence herbicides in loam soil that are commonly used in rice crop cultivation. A number of strategic soil extraction and cleanup methods were evaluated. The instrumental analysis was performed by HPLC with a diode array detector. The best compromise between the recoveries (69–98%) and good repeatability (RSD <15%) was obtained using either methanol or methanol–ethyl acetate (70 + 30, v/v) as extraction solvents without any further cleanup. For the first time, residues of metsulfuron methyl, bensulfuron ethyl, bentazone, cyhalofop butyl, pyrazosulfuron ethyl, propanil, and clomazone were analyzed simultaneously. Quinclorac and bispyribac sodium were also assayed, but their recoveries were below 50%. Both methods had an LOD of 0.7 mg/kg and could accurately determine the residues at the 2 mg/kg level. These two methods could not be applied directly to other soil types as the recoveries strongly depended on the soil composition. The developed methodologies were successfully applied in monitoring 87 real-world soil samples, in which only propanil (6 to 12 mg/kg) and clomazone (15 to 20 mg/kg) residues could be detected.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[Polychlorinated dibenzo-p-dioxins (PCDD) and dibenzofurans (PCDF) are persistent, toxic, and bioaccumulate in the environment. Due to their high analytical costs, these compounds are hardly regulated and mostly not monitored in the Third World. To overcome this, bioassays have been proposed as low-cost alternative methods. Two of the most established bioanalytical tools, the dioxin antibody-based enzyme-linked immunosorbent assay ELISA and the micro-EROD bioassay are evaluated and compared to high resolution gas chromatography and high resolution mass spectrometry (HRGC/HRMS) analytical methodology. The methods were tested using thirteen soils and sediment samples selected from diverse sites in Montevideo, Uruguay. The WHO₂₀₀₅ total toxic equivalent (WHO₂₀₀₅-TEQ) of soils ranged from 2.4 to 2212 (ng WHO₂₀₀₅-TEQ/kg dry sample) and from 0.14 to 9.4 (ng WHO₂₀₀₅-TEQ/kg dry sample) in sediments. This study shows significant contamination related to dioxin-like compounds, particularly in sites where uncontrolled burnings were carried out. ELISA and micro-EROD bioassay correlated well with HRGC/HRMS, R Spearman 0.773 and 0.913, respectively and were highly correlated to each other, R Spearman 0.879. Preliminary threshold values of bioassay toxic equivalents of 330 (ng/kg dry sample) for the micro-EROD bioassay and 220 (ng/kg dry sample) for ELISA are proposed.]]></dcterms:abstract>

<dcterms:abstract><![CDATA[The origins of the biological complexity and the factors that regulate the development of community composition, diversity and richness in soil remain largely unknown. To gain a better understanding of how bacterial communities change during soil ecosystem development, their composition and diversity in soils that developed over c. 77 000 years of intermittent aeolian deposition were studied. 16S rRNA gene clone libraries and fatty acid methyl ester (FAME) analyses were used to assess the diversity and composition of the communities. The bacterial community composition changed with soil age, and the overall diversity, richness and evenness of the communities increased as the soil habitat matured. When analysed using a multivariate Bray–Curtis ordination technique, the distribution of ribotypes showed an orderly pattern of bacterial community development that was clearly associated with soil and ecosystem development. Similarly, changes in the composition of the FAMEs across the chronosequence were associated with biomarkers for fungi, actinomycetes and Gram-positive bacteria. The development of the soil ecosystem promoted the development of distinctive microbial communities that were reminiscent of successional processes often evoked to describe change during the development of plant communities in terrestrial ecosystems.]]></dcterms:abstract>
