Saponin-based adjuvants are promising adjuvants that enhance both humoral and T-cell-mediated immunity. One of the most used natural products as vaccine adjuvants are Quillaja saponaria bark saponins and its fraction named Quil A®. Despite that, its use has been restricted for human use due to safety issues. As an alternative, our group has been studying the congener species Quillaja brasiliensis saponins and its performance as vaccine adjuvants, which have shown to trigger humoral and cellular immune responses comparable to Quil A® but with milder side effects. Here, we studied a semi purified aqueous extract (AE) and a previously little characterized saponin-enriched fraction (QB-80) from Q. brasiliensis as vaccine adjuvants and an inactivated virus (bovine viral diarrhea virus, BVDV) antigen co-formulated in experimental vaccines in mice model. For the first time, we show the spectra pattern of the Q. brasiliensis saponins by MALDI-TOF, a novel and cost-effective method that could be used to characterize different batches during saponins production. Both AE and QB-80 exhibited noteworthy chemical similarities to Quil A®. In addition, the haemolytic activity and toxicity were assessed, showing that both AE and QB-80 were less toxic than Quil A®. When subcutaneously inoculated in mice, both fractions promoted long-term strong antibody responses encompassing specific IgG1 and IgG2a, enhanced the avidity of IgG antibodies, induced a robust DTH reaction and significantly increased IFN-ɣ production in T CD4+ and T CD8+ cells. Furthermore, we have proven herein that AE has the potential to promote dose-sparing, substantially reducing the dose of antigen required for the BVDV vaccines and still eliciting a mixed Th1/Th2 strong immune response. Based on these results, and considering that AE is a raw extract, easier and cheaper to produce than commercially available saponins, this product can be considered as candidate to be escalated from experimental to industrial uses.

Commercially available saponins are extracted from Quillaja saponaria barks, being Quil A® the most widely used. Nanoparticulate immunostimulating complexes (ISCOMs or ISCOMATRIX) formulated with these, are able to stimulate strong humoral and cellular immune responses. Recently, we formulated novel ISCOMs replacing QuilA® by QB-90 (IQB-90), a Quillaja brasiliensis leaf-extracted saponin fraction, and reported that IQB-90 improved antigen uptake, and induced systemic and mucosal antibody production, and T-cell responses. However, its mechanism of action remains unclear. In this study we provide a deeper insight into the immune stimulatory properties of QB-90 and ISCOMATRIX-like based on this fraction (IMXQB-90). We show herein that, when used as a viral vaccine adjuvant, QB-90 promotes an “immunocompetent environment”. In addition, QB-90 and IMXQB-90 induce immune-cells recruitment at draining-lymph nodes and spleen. Subsequently, we prove that QB-90 or IMXQB-90 stimulated dendritic cells secret IL-1β by mechanisms involving Caspase-1/11 and MyD88 pathways, implying canonical inflammasome activation. Finally, both formulations induce a change in the expression of cytokines and chemokines coding genes, many of which are up-regulated. Findings reported here provide important insights into the molecular and cellular mechanisms underlying the adjuvant activity of Q. brasiliensis leaf-saponins and its respective nanoparticles.

Echinococcus granulosus is the parasite responsible for cystic echinococcosis (CE), an important worldwide‐distributed zoonosis. New effective vaccines against CE could potentially have great economic and health benefits. Here, we describe an innovative vaccine design scheme starting from an antigenic fraction enriched in tegumental antigens from the protoscolex stage (termed PSEx) already known to induce protection against CE. We first used mass spectrometry to characterize the protein composition of PSEx followed by Gene Ontology analysis to study the potential Biological Processes, Molecular Functions, and Cellular Localizations of the identified proteins. Following, antigenicity predictions and determination of conservancy degree against other organisms were determined. Thus, nine novel proteins were identified as potential vaccine candidates. Furthermore, linear B cell epitopes free of posttranslational modifications were predicted in the whole PSEx proteome through colocalization of in silico predicted epitopes within peptide fragments identified by matrix‐assisted laser desorption/ionization‐TOF/TOF. Resulting peptides were termed “clean linear B cell epitopes,” and through BLASTp scanning against all nonhelminth proteins, those with 100% identity against any other protein were discarded. Then, the secondary structure was predicted for peptides and their corresponding proteins. Peptides with highly similar secondary structure respect to their parental protein were selected, and those potentially toxic and/or allergenic were discarded. Finally, the selected clean linear B cell epitopes were mapped within their corresponding 3D‐modeled protein to analyze their possible antibody accessibilities, resulting in 14 putative peptide vaccine candidates. We propose nine novel proteins and 14 peptides to be further tested as vaccine candidates against CE.

Actualmente la mayoría de las vacunas son diseñadas con subunidades las cuales poseen una inmunogenicidad limitada, por lo que es necesario el uso de adyuvantes en su formulación. A pesar de los avances en inmunología, el empleo de compuestos de aluminio como adyuvantes sigue siendo la opción más aceptada. Éstos presentan desventajas, lo que ha impulsado el desarrollo de nuevos adyuvantes, que es en la actualidad un campo de gran actividad científica. En esta área se enmarca el presente proyecto de tesis. Las suspensiones FCC-S06 y HAP-S08, sintetizadas a partir de calcita (carbonato de calcio), suministradas por una empresa extranjera fueron evaluadas como posible adyuvante. La FCCS06 es una suspensión de micropartículas funcionalizadas con grupos fosfato. Obteniéndose, según la empresa, micropartículas donde se incorpora hidroxiapatita en la superficie, resultando en un incremento considerable en la superficie específica. La HAP-SO8 fue sintetizada utilizando otro proceso de síntesis con el objetivo de obtener una suspensión de partículas con un diámetro promedio inferior al mínimo obtenido utilizando la síntesis estándar de FCCs. La empresa informó que la suspensión resultante teóricamente está compuesta por partículas que son 100% hidroxiapatita. Como está demostrada la eficacia de la hidroxiapatita en la adsorción de una gran variedad de proteínas, es biocompatible y se ha utilizado como adyuvante de inmunización se eligieron ambas suspensiones como candidatas para su evaluación como adyuvantes. En el presente trabajo se confirmó la hipótesis de que la suspensión FCCS06 es biocompatible ya que no se observó toxicidad en los estudios in vitro ni in vivo, siendo menos tóxica que el adyuvante de referencia. Está formada por un core de carbonato de calcio con una sal de la familia de los fosfatos de calcio precipitada en su superficie, cristalina, de morfología esferoide con estructuras en forma de placas laminares que sobresalen de la superficie formando poros intrapartículas, lo que aumenta considerablemente la superficie específica. Las formulaciones con FCC-S06 tienen la capacidad de activar al sistema inmune 9 innato, estimulando la activación y maduración de células presentadoras de antígenos, la presentación antigénica, el reclutamiento de componentes del sistema inmune al sitio de inoculación y la secreción de citoquinas proinflamatorias. Por lo anterior, se podría considerar como un posible candidato adyuvante. Lo antes mencionado modula la respuesta adaptativa hacia un perfil Th17 y Th1. En cuanto a la suspensión HAP-S08 no se pudo identificar la sal de la familia de los fosfatos de calcio que la compone. Se determinó que está formada por partículas amorfas sin poros intrapartículas. Los resultados permiten asegurar que no es tóxica ni in vitro ni in vivo, y sugieren que las formulaciones con HAPS08 inducen in vitro la activación y maduración de células presentadoras de antígenos, y en concordancia con el perfil de citoquinas secretado por estas células, estimulan la activación de una respuesta inmune adaptativa polarizada hacia un perfil Th17. Sin embargo, poseen una gran limitante, son muy inestables y sedimentan en periodos cortos de tiempo siendo difícil su re-dispersión. Lo anterior hace que su aplicación en el diseño de vacunas no sea recomendable. Por lo cual para seguir avanzando en el proceso de desarrollo de un nuevo adyuvante es necesario realizar un nuevo proceso de síntesis que no se base en la precipitación. <em>Quillaja brasiliensis</em> (A. St.-Hil. & Tul.) Mart. is currently one of the only two species of the only genus in the Quillajaceae D. Don (Dicotyledoneae) family. Recently, remarkable developments have been achieved regarding its domestication,<br />propagation, and biochemical study. The first results on the structural study of the complex mixture of saponins produced by different organs of the tree have been published. Analogously to the related species Q. saponaria Molina, its saponins, either alone or in colloidal formulations, have been proved to be very effective as adjuvants in experimental vaccines, being able to elicit an early humoral and cellular response against the co-administered antigens. Quillaja saponins have an intrinsic capacity to interact with membrane lipids that self-assembles in nanoparticles (immu nostimulating complexes or ISCOM-matrices) with outstanding immunoadjuvant activity and low toxicity profle. However, the expensive and laborious purifcation processes applied to purify Quillaja saponins used to assemble ISCOM-matrices show an important drawback in the large-scale use of this vaccine adjuvant. Thus, in this study, we describe a protocol to appropriately formulate ISCOM-matrices using the raw aqueous extract (AE) of Quillaja lancifolia leaves. In the presence of lipids, AE was able to self-assemble in nanostructures that resembles immunostimulating complexes (ISCOM). These nega tively charged nanoparticles of approximately 40 nm were characterized by transmission electron microscopy and dynamic light scattering. In addition, well-known saponins with remarkable immunoadjuvant activity, as QS-21, were detected into nanoparticles. Thus, the easier, robust, cheaper, and environmentally friendly method developed here may be an alternative to the classical methods for ISCOM-matrices production that use high-purifed saponins.

Vaccines are among the most important advances in medicine throughout the human history. However, conventional vaccines exhibit several drawbacks in terms of design and production costs. Peptide-based vaccines are attractive alternatives, since they can be designed mainly in silico, can be produced cheaply and safely, and are able to induce immune responses exclusively towards protective epitopes. Yet, a proper peptide design is needed, not only to generate peptide-specific immune responses, but also for them to recognize the native protein in the occurrence of a natural infection. Herein, we propose a rational workflow for developing peptide-based vaccines including novel steps that assure the cross-recognition of native proteins. In this regard, we increased the probability of generating efficient antibodies through the selection of linear B-cell epitopes free of posttranslational modifications followed by analyzing the 3D-structure similarity between the peptide in-solution vs. within its parental native protein. As a proof of concept, this workflow was applied to a set of seven previously suggested potential protective antigens against the infection by Echinococcus granulosus sensu lato. Finally, two peptides were obtained showing the capacity to induce specific antibodies able to exert anti-parasite activities in different in vitro settings, as well as to provide significant protection in the murine model of secondary echinococcosis.

Cystic echinococcosis, a zoonosis caused by cestodes belonging to the Echinococcus granulosus sensu lato (s.l.) genetic complex, affects humans and diverse livestock species. Although a veterinary vaccine exhibiting high levels of antibody-mediated protection has successfully reached the market, the large genetic diversity among parasite isolates and their particular host preferences, makes still necessary the search for novel vaccine candidates. Glutathione transferases (GSTs) constitute attractive targets for immunoprophylaxis due to their outstanding relevance in helminth detoxification processes, against both exogenous and endogenous stressors. Among the six GSTs known to be expressed in E. granulosus s.l., EgGST1 (Mu-class), EgGST2 (Sigma-class), and EgGST3 (a still non-classifiable isoenzyme), show the highest proteomic expression. Therefore, their recombinant forms -rEgGST1, rEgGST2 and rEgGST3- were herein analyzed regarding their potential to induce long-term antiparasite protection in mice. Only immunization with rEgGST1 induced long-lasting protection; and accordingly, rEgGST1-specific antibodies enhanced the parasite killing through both the classical activation of the host complement system and the antibody-dependent cellular cytotoxicity by macrophages. These results support further testing of rEgGST1 as a vaccine candidate in diverse hosts due to the broad expression of EgGST1 in different parasite stages and tissues.

Increasing the effectiveness of vaccines against respiratory viruses is particularly relevant for the elderly, since they are prone to develop serious infections due to comorbidities and the senescence of the immune system. The addition of saponin-based adjuvants is an interesting strategy to increase the effectiveness of vaccines. We have previously shown that ISCOM matrices from Q. brasiliensis (IMXQB) are a safe and potent adjuvant. In this study, we evaluated the use of IMXQB as an adjuvant for the seasonal trivalent influenza vaccine (TIV) in an aged mice model. Herein, we show that subcutaneous injection of the adjuvanted vaccine promoted higher titers of IgM, IgG (and isotypes), and serum hemagglutination inhibition titers (HAI). Notably, aged mice immunized by intranasal route also produced higher IgG (and isotypes) and IgA titers up to 120 days after priming, as well as demonstrating an improvement in the HAI antibodies against the TIV. Further, experimental infected aged mice treated once with sera from adult naïve mice previously immunized with TIV-IMXQB subcutaneously successfully controlled the infection. Overall, TIV-IMXQB improved the immunogenicity compared to TIV by enhancing systemic and mucosal immunity in old mice conferring a faster recovery after the H1N1pdm09-like virus challenge. Thus, IMXQB nanoparticles may be a promising platform for next-generation viral vaccines.
