Ph and temperature effects on the Km values of muscle lactate dehydrogenase isozyme LDH-A4 from fishes of the family Sciaenidae (Perciformes)

 1. Lactate dehydrogenase isosyme LDH-A4 from the skeletal muscle of the two stenothermal species, Cynoscion striatus and Micropogonias furnieri, showed an optimal activity at pH 6.8 at high temperature (30°C), decreasing abruptly at higher pH values. Meanwhile, in the eurythermal species Ronxador stearnsii, the LDH-A4 activity showed a plateau until pH 7.2, decreasing moderately at higher pH. Such differences were not pronounced at lower pyruvate concentrations of the order of ten or less.

 2. At low temperature (20°C) LDH-A4 for one stenothermal species showed a plateau of activity with predominance until pH 7.6, decreasing less abruptly until pH 8.3, while the opposite behaviour was shown by the eurythermal species.

3. Q10 values (20–30°C) at three different pyruvate concentrations were determined, showing the values expected at low pH, which reduced with increasing pH.

4. The relationship between the apparent KM and temperature at five different pH values was determined.
