Abstract

Nitro-fatty acids(NO2-FA)areelectrophilicsignalingmediatorsformedintissuesduringinflammation, which areabletoinducepleiotropiccytoprotectiveandantioxidantpathwaysincludingupregulationof Nuclearfactorerythroid2-relatedfactor2(Nrf2)responsivegenes.AmyotrophicLateralSclerosis(ALS)is a fatalneurodegenerativediseasecharacterizedbythelossofmotorneuronsassociatedtoanin- flammatoryprocessthatusuallyaggravatesthediseaseprogression.InALSanimalmodels,theactivation of thetranscriptionfactorNrf2inastrocytesconfersprotectiontoneighboringneurons.Itiscurrently unknown whetherNO2-FAcanexertprotectiveactivityinALSthroughNrf2activation.Hereinwede- monstratethatnitro-arachidonicacid(NO2-AA) ornitro-oleicacid(NO2-OA)administratedtoastrocytes expressingtheALS-linkedhSOD1G93A induce antioxidantphaseIIenzymeexpressionthroughNrf2ac- tivationconcomitantwithincreasingintracellularglutathionelevels.Furthermore,treatmentof hSOD1G93A-expressingastrocyteswithNO2-FApreventedtheirtoxicitytomotorneurons.Transfectionof siRNAtargetedtoNrf2mRNAsupportedtheinvolvementofNrf2activationinNO2-FA-mediatedpro- tectiveeffects.Ourresultsshowforthe first timethatNO2-FAinduceapotentNrf2-dependentanti- oxidantresponseinastrocytescapableofpreventingmotorneuronsdeathinaculturemodelofALS
