The present work shows the results obtained during the first six months of the Pharmacokinetic Monitoring Section in our University Hospital in Uruguay. Plasma concentrations of Carbamazepine |b Phenytoin and Phenobarbital were monitored for the study. Analysis requests forms |b Sampling Instructions and Pharmacokinetics Algorithms are discussed. Results obtained only from plasma levels in ambulatory and hospitalised patients undergoing chronic therapy are analyzed. Michaelis-Menten elimination parameters of Phenytoin could be estimated for our patients (Km=13 mg/l and Vmax=0.4 mg/kg/h). Sex and comedication with Carbamazepine had no effect on the Phenytoin concentrations. On the other hand |b concomitant treatment with Phenytoin and/or Phenobarbital reduced the elimination half-life of Carbamazepine by 50. Phenobarbital has not been the drug of choice in patients undergoing monotherapy treatment. Collection of trough samples (prior to the administration of a dose) was appropriate for therapeutic drug monitoring of these anticonvulsants.
