Abstract

Glyphosate is the most-used herbicide worldwide, but few studies address ways to mitigate its environmental impact. Biobeds proved to be an effective tool for reducing point-source pesticide contamination because they can degrade many classes of pesticides. However, as the dissipation of glyphosate and its metabolite AMPA in biobeds has rarely been addressed, a degradation study of glyphosate and its metabolites was conducted. For sample preparation, the biomixture used to construct the biobeds was lyophilised, and various methods were evaluated. Finally, the samples treated with 0.6 M potassium hydroxide analysed using IC-MS/MS enabled the successful determination of glyphosate, AMPA, and N-acetyl glyphosate, with recoveries ranging from 74% to 112% (RSDs <15%) and a Lowest Calibration Level of 1 mg/kg using matrix-matched calibration curves due to significant signal suppression. The methodology was applied to study the degradation of glyphosate and its metabolites in biobeds installed at an aerial application company in Uruguay. The highest glyphosate concentrations detected ranged from 354 to 425 mg kg− 1. After one year 98% degradation was confirmed. AMPA reached peak concentrations of 281 to 335 mg kg−1 and degraded from 80% to 67% by the end of the experiment. N-acetyl glyphosate was not detected. Overall, the methodology proved to be simple, efficient, and robust. IC-MS/MS is straightforward and can be extended to other challenging, complex matrices for glyphosate determination, as no derivatisation is required.
